Mite populations in a forest reserve, where accidental fire swept through almost half of the total area covered by the forest, were enumerated at one, three, and six months after burning. Mite densities in both burnt and unburnt plots were extremely low immediately after burning. Six months after burning, the density of juvenile cryptostigmatid mites were higher in the burnt area than the unburnt area and several mite genera had recovered from the deleterious effect of burning, but overall mite densities were still lower in the burnt than the unburnt area. Some of the typical dominant mite genera of the tropical rainforest, namely Haplozetes sp., Scheloribates sp., Nothrus sp. and Carabodes sp. did not recover from the effect of burning within six months. The selective influence of burning on mite genera/families, as well as the different rates of recovery observed in this study, were attributed to differences in feeding requirements, phenological patterns and life history tactics of the mites. Juvenile cryptostigmatid mites were more in the burnt than unburnt plots suggesting that reproductive activities of this group increased after burning.  相似文献

The Smirnoff-Wheeler (SW) pathway has been proven to be the only significant source of l-ascorbic acid (AsA; vitamin C) in the seedlings of the model plant Arabidopsis thaliana. It is yet uncertain whether the same pathway holds for all other plants and their various organs as AsA may also be synthesized through alternative pathways. In this study, we have cloned some of the genes involved in the SW-pathway from acerola (Malpighia glabra), a plant containing enormous amount of AsA, and examined the expression patterns of these genes in the plant. The AsA contents of acerola leaves were about 8-fold more than that of Arabidopsis with 5-700-fold higher mRNA abundance in AsA-biosynthesizing genes. The unripe fruits have the highest AsA content but the accumulation was substantially repressed as the fruit transitions to maturation. The mRNAs encoding these genes showed correlation in their expression with the AsA contents of the fruits. Although very little AsA was recorded in the seeds the mRNAs encoding all the genes, with the exception of the mitochondrially located L-galactono-1,4-lactone dehydrogenase, were clearly detected in the seeds of the unripe fruits. In young leaves of acerola, the expression of most genes were repressed by the dark and induced by light. However, the expression of GDP-D-mannose pyrophosphorylase similar to that encoded by A. thaliana VTC1 was induced in the dark. The expressions of all the genes surged after 24h following wounding stress on the young leaves. These findings will advance the investigation into the molecular factors regulating the biosynthesis of abundant AsA in acerola.  相似文献

Unraveling the role of genes annotated as protein of unknown function is of importance in progression of plant science. l-Galactono-1,4-lactone (l-GalL) is the terminal precursor for ascorbic acid (AsA) biosynthesis in Arabidopsis thaliana, and a previous study showed two DUF (domains of unknown function) 642 family genes (At1g80240 and At5g25460, designated as DGR1 and DGR2, respectively) to be sensitive to it. In this work, leaves from wild-type Arabidopsis were fed with d-glucose, l-galactose, l-GalL and AsA, and the expression level of the At1g80240 and At5g25460 genes showed a specific response to l-GalL, but not to the other supplements despite the increases of the tissue AsA contents. Analysis of promoter-β-glucuronidase (GUS) transgenic plants showed the two genes to be complementarily expressed at the root apex and in the rest of the root excluding the apex, respectively, in both young and old seedlings, and to be expressed at the leaf primordia. The GUS activity under the control of the At5g25460 promoter was high in the cotyledon and leaf veins of young seedlings. These findings were consistent with the results of quantitative real-time PCR. Interestingly, the T-DNA insertion mutant of At5g25460 (SALK_125079) displayed shorter roots and smaller rosettes than Col-0; however, no phenotypic difference was observed between the T-DNA insertion mutant of At1g80240 and the wild type. This is the first report on the expression and functional analysis of these two DUF642 family genes, with the results revealing the contribution of DGR genes to the development of Arabidopsis.  相似文献

Constructed wetland is an innovative and emerging ecological technology for wastewater treatment. This study was conducted to investigate the effectiveness of a Vegetated Submerged Bed Constructed Wetland (VSBCW) for removal of heavy metals from industrial wastewater in a steel manufacturing company. A pilot Effluent Treatment Plant (ETP) consisting of equalization basin, two VSBCW basins and a storage tank was constructed. The VSBCW was constructed using 10–30 mm round granite for the different zones. This was overlaid by 200 mm deep granite and 150 mm washed sand with Phragmites karka, Vetiveria nigritana and Cana lilies as macrophytes. Irrigation of macrophytes using effluent from the industry was done after 3 months of planting and ETP monitored. Industrial wastewater samples were collected and analyzed for heavy metals such as zinc (Zn), lead (Pb), iron (Fe), manganese (Mn), magnesium (Mg) and chromium (Cr) to know the treatment efficiency of the ETP. Results indicated that the removal efficiencies of the VSBCW for Pb, Mg and Cr were 15.4%, 79.7% and 97.9% respectively. Fe and Mn were seen to increase by 1.8% and 33% respectively. The ETP using locally available macrophytes is effective in the phytoremediation of heavy metals, particularly Cr from the wastewater.  相似文献

Various hydrocarbons have been released into the environment as a result of industrialization. An effective way of removing these materials without further environmental contamination is microbial bioremediation. Mycobacterium gilvum PYR-GCK, a bacteria isolated from a PAH polluted estuary, was studied using comparative shotgun proteomics to gain insight on its molecular activity while using pyrene and glucose as sole carbon and energy sources. Based on annotated genomic information, a confirmation analysis was first performed to confirm its pyrene degradation activity, using gas chromatography–mass spectrometry technology. One dimensional gel electrophoresis and liquid chromatography–mass spectrometry technologies employed in the proteomics analysis revealed the expression of pyrene degrading gene products along with upregulated expression of proteins functioning in the glyoxylate and shikimate pathways, in the pyrene-induced cells. The study also revealed the pathway of pyrene degraded intermediates, via partial gluconeogenesis, into the pentose phosphate pathway to produce precursors for nucleotides and amino acids biosynthesis.  相似文献
