SYNOPSIS. Out of 64 nutrients tested, none replaced glycerol for growth in the dark of Prymnesium parvum. Some metabolites enahanced growth in the light, and in the presence of glycerol also in the clark. Good growth with glycerol could be obtained in the absence of CO2. Survival of cultures in the dark in media without glycerol was prolonged by various nutrients. Thioglycerol in glycerol-containing media inhibited growth in light and darkness. Apparently Prymnesium parvum has a specific glycerol requirement for dark growth.  相似文献

Xylem resistance to water stress‐induced cavitation is an important trait that is associated with drought tolerance of plants. The level of xylem cavitation experienced by a plant is often assessed as the percentage loss in conductivity (PLC) at different water potentials. Such measurements are constructed with samples that are excised underwater at native tensions. However, a recent study concluded that cutting conduits under significant tension induced cavitation, even when samples were held underwater during cutting. This resulted in artificially increased PLC because of what we have termed a ‘tension‐cutting artefact’. We tested the hypothesized tension‐cutting artefact on five species by measuring PLC at native tension compared with after xylem tensions had been relaxed. Our results did not support the tension‐cutting artefact hypothesis, as no differences were observed between native and relaxed samples in four of five species. In a fifth species (Laurus nobilis), differences between native and relaxed samples appear to be due to vessel refilling rather than a tension‐cutting effect. We avoided the tension‐cutting artefact by cutting samples to slightly longer than their measurement length and subsequent trimming of at least 0.5 cm of sample ends prior to measurement.  相似文献

We examined whether extensive dry season dieback and mortality in a South African fynbos community were due to drought or pathogen attack. Plant dieback and mortality have been reported elsewhere in similar plant communities suggesting potential for a widespread climatic or biotic threat to this community. We collected tissue samples from Brunia noduliflora, the dominant plant in the community, and cultured them for potential plant pathogens. We also measured dry season predawn and midday water potentials of healthy and stressed plants and constructed pressure‐volume curves to assess turgor loss point. Plant stress and mortality were monitored over a 2‐year study period. Both healthy plants and plants that displayed moderate signs of stress had dry season predawn water potentials well above their turgor loss point suggesting plants were not water stressed. However, plants displaying >60% crown dieback had much lower water potentials (as low as ?12 MPa). A previously undescribed fungus (Pythium sp.) was isolated from the root vascular tissue of all stressed plants and was not present in healthy plants. The proximate cause of plant stress was likely pathogen‐induced, while the ultimate cause of plant death appears to be extreme water stress. The present study suggests that Brunia (Bruniceae), Leucadendron (Proteaceae) and Erica (Ericaceae), all emblematic and dominant genera within the diverse fynbos community, may be susceptible to Pythium infection. This may pose a serious threat to communities already threatened by climate change.  相似文献

The rainbow darter Etheostoma caeruleum is a small fish in the perch family (Percidae) that is adapted to fast‐flowing streams in eastern North America. It is relatively sensitive to habitat degradation and is widely used as a sentinel of stream condition. To provide a complementary tool for assessing the integrity of stream ecosystems, 16 highly polymorphic tetranucleotide microsatellite markers were identified for these darters. Between four and 16 loci were found to be useful in five congeneric species. These markers will be useful for characterizing population genetic structure and diversity of rainbow darters and related fishes.  相似文献

Aquaporins (AQPs) are water channels that allow cells to rapidly alter their membrane water permeability. A convenient model for studying AQP expression and activity regulation is Black Mexican Sweet (BMS) maize cultured cells. In an attempt to correlate membrane osmotic water permeability coefficient (Pf) with AQP gene expression, we first examined the expression pattern of 33 AQP genes using macro-array hybridization. We detected the expression of 18 different isoforms representing the four AQP subfamilies, i.e. eight plasma membrane (PIP), five tonoplast (TIP), three small basic (SIP) and two NOD26-like (NIP) AQPs. While the expression of most of these genes was constant throughout all growth phases, mRNA levels of ZmPIP1;3 , ZmPIP2;1 , ZmPIP2;2, ZmPIP2;4 and ZmPIP2;6 increased significantly during the logarithmic growth phase and the beginning of the stationary phase. The use of specific anti-ZmPIP antisera showed that the protein expression pattern correlated well with mRNA levels. Cell pressure probe and protoplast swelling measurements were then performed to determine the Pf. Interestingly, we found that the Pf were significantly increased at the end of the logarithmic growth phase and during the steady-state phase compared to the lag phase, demonstrating a positive correlation between AQP abundance in the plasma membrane and the cell Pf.  相似文献

The compound 5-chloro-3-methyl-4-nitro-1H-pyrazole (CMNP) is a pyrazole-derivative that induces abscission selectively in mature citrus (Citrus sinensis) fruit when applied to the canopy and has herbicidal activity on plants when applied to roots. Despite the favourable efficacy of this compound, the mode of action remains unknown. To gain information about the mode of action of CMNP, the effect of application to mature citrus fruit and Arabidopsis thaliana roots was explored. Peel contact was essential for mature fruit abscission in citrus, whereas root drenching was essential for symptom development and plant decline in Arabidopsis. CMNP was identified as an uncoupler in isolated soybean (Glycine max) mitochondria and pea (Pisum sativum) chloroplasts and an inhibitor of alcohol dehydrogenase in citrus peel, but not an inhibitor of protoporphyrinogen IX oxidase. CMNP treatment reduced ATP content in citrus peel and Arabidopsis leaves. Phospholipase A2 (PLA2) and lipoxygenase (LOX) activities, and lipid hydroperoxide (LPO) levels increased in flavedo of citrus fruit peel and leaves of Arabidopsis plants treated with CMNP. An inhibitor of PLA2 activity, aristolochic acid (AT), reduced CMNP-induced increases in PLA2 and LOX activities and LPO levels in citrus flavedo and Arabidopsis leaves and greatly reduced abscission in citrus and delayed symptoms of plant decline in Arabidopsis. However, AT treatment failed to halt the reduction in ATP content. Reduction in ATP content preceded the increase in PLA2 and LOX activities, LPO content and the biological response. The results indicate a link between lipid signalling, abscission in citrus and herbicidal damage in Arabidopsis.  相似文献

THE unusual modified base1 of the nucleic acid of the large, generalized transducing phage of Bacillus subtilis SP-15 was predicted from the low denaturation temperature of the native nucleic acid. The base was detected as a fifth ultraviolet-absorbing component once the nucleic acid had been hydrolysed with formic acid; it partially replaces the thymine and represents 12 mol per cent of all the bases1. It is responsible for the low Tm of the nucleic acid, since once all other unusual components have been removed, the remaining material melts at the same temperature as the native nucleic acid. We report here the isolation and characterization of this new pyrimidine.  相似文献
