The electrophysiological properties of cultured human melanocytes were investigated using the whole-cell configuration of the patch-clamp technique. Depolarizations to membrane potentials more positive than -30 mV resulted in the rapid development (<1 ms to peak) of an inward current. The maximum peak current was observed at +10 mV and reached an average amplitude of about 270 pA. During the depolarizations, the current inactivated with a time constant of about 2 ms. The current was abolished by the addition of 0.3 μM tetrodotoxin, a blocker of voltage-gated Na+-channels, and disappeared when Na+ was omitted from the extracellular medium. In addition, the melanocytes contain at least two types of outward K+-current. The first type, observed in every cell, was highly sensitive (Ki 1 mM) to the K+-channel blocker TEA, required depolarizations beyond zero to be activated and did not inactivate. The second type was less regularly observed (10% of the cells). This current activated at more negative voltages (–20 mV), was resistant to TEA (20 mM) but was blocked by 2 mM 4-aminopyridine and inactivated rapidly during depolarizations. We conclude that human melanocytes are equipped with voltage-dependent Na+-channels, a delayed rectifying K+-current and a K+-current similar to the A-current in neurones.  相似文献

A morphological and anatomical investigation is presented of the two species of Heberdenia, H. bahamensis (Macaronesia) and H. penduliflora (Mexico). A cladistic analysis including 27 taxa of the Myrsinaceae and using Jacquinia (Theophrastaceae) and Manilkara (Sapotaceae) as outgroup was performed to provide a hypothesis of the relationships of the two species of Heberdenia. It was concluded that H. bahamensis and H. penduliflora are not more closely related to each other than either is to many other species of the Myrsinaceae, and that they should not be referred to the same genus. Heberdenia bahamensis appears to be most closely related to the Old World genera Pleiomeris, Embelia , and Grenacheria , whereas H. penduliflora is nested within Ardisia s. L , possibly being most closely related to the segregate genus Gentlea. It is suggested that H. penduliflora is probably best referred to a genus of its own.  相似文献

An instrument is described which functions as a spectrophotometer, In addition it measures action spectra of photobiological processes, excitation and emission spectra of fluorescence, and gives data for calculation of action spectra of partial reactions of photosynthesis and photocontrolled enzymatic reactions. The instrument has a Bausch and Lomb 250 mm grating monochromator. Equal numbers of quanta at different wavelengths are adjusted electronically with control of the slit through a quantacorrected photocell and a servo motor which is programmed by a cam connected to the wavelength drive.  相似文献

Two new species, Dorvillea bermudensis sp.n. and D. albomaculata sp.n., with obligate asexual reproduction are described. Both are closely related to sexually reproducing members of the same genus. The new species undergo repeated fission of the architomy type, with the rate of regeneration and subsequent production of segments being positively correlated with the energy content of the fragments. Regeneration and reproductive potentials vary between head and tail fragments with tail lineages producing higher rates than head lineages. Both species are very tolerant of starvation which may cause considerable degrowth and initiation of dispersal behavior.  相似文献
