Low-density quantitative real-time PCR (qPCR) arrays are often used to profile expression patterns of microRNAs in various biological milieus. To achieve accurate analysis of expression of miRNAs, non-biological sources of variation in data should be removed through precise normalization of data. We have systematically compared the performance of 19 normalization methods on different subsets of a real miRNA qPCR dataset that covers 40 human tissues. After robustly modeling the mean squared error (MSE) in normalized data, we demonstrate lower variability between replicates is achieved using various methods not applied to high-throughput miRNA qPCR data yet. Normalization methods that use splines or wavelets smoothing to estimate and remove Cq dependent non-linearity between pairs of samples best reduced the MSE of differences in Cq values of replicate samples. The cancer stem cell (CSC) model encompasses an advantageous paradigm that in recent decades provides a better elucidation for many important biological aspects of cancer initiation, progression, metastasis, and, more important, development of multidrug resistance (MDR). Such several other hematological malignancies and solid tumors and the identification and isolation of ovarian cancer stem cells (OV-CSCs) show that ovarian cancer also follows this hierarchical model. Gaining a better insight into CSC-mediated resistance holds promise for improving current ovarian cancer therapies and prolonging the survival of recurrent ovarian cancer patients in the future. Therefore, in this review, we will discuss some important mechanisms by which CSCs can escape chemotherapy, and then review the recent and growing body of evidence that supports the contribution of CSCs to MDR in ovarian cancer. 相似文献

The analysis of myenteric neurons is becoming increasingly important for the assessment of enteric nervous system injury and
degeneration occurring in motor disorders of the gut. Limited information is presently available on the quantitative estimation
of myenteric neurons and glial cells in paraffin-embedded colonic sections; additional data would be useful for diagnostic
purposes. In this morphometric study, we performed immunohistochemistry to count myenteric neurons and glial cells in paraffin
sections of human colon. Serial cross sections of formalin-fixed paraffin-embedded full-thickness normal human left colon
(n = 10, age-range: 50–72 years) were examined. HuC/D and S100β antigens were found to be the best markers for the detection
of neurons and glial cells, respectively. Significant correlations were noted between the numbers of neurons/glial cells and
the respective myenteric ganglion areas. These findings suggest that HuC/D-S100β-immunostained paraffin cross sections of
human colon can be regarded as valuable tools for the quantitative estimation of myenteric neurons and glial cells. Based
on the present method, only a limited number of paraffin sections are needed for reliable quantitative assessments of myenteric
ganglion cells, thus allowing fast and simple approaches in the settings of the histopathological diagnosis of colonic motility
disorders and retrospective evaluations of pathological archival tissue specimens.
