Aspergillus fumigatus is responsible for many allergic respiratory diseases, the most notable of which - due to its severity - is allergic bronchopulmonary aspergillosis. Aspf1 is a major allergen of this fungus: this 149-amino acid protein belongs to the ribotoxin family, whose best characterized member is alpha-sarcin (EC 3.1.27.10). The proteins of this group are cytotoxic ribonucleases that degrade a unique bond in ribosomal RNA impairing protein biosynthesis. Aspf1 and its deletion mutant Aspf1Delta(7-22) have been produced as recombinant proteins; the deleted region corresponds to an exposed beta-hairpin. The conformation of these two proteins has been studied by CD and fluorescence spectroscopy. Their enzymatic activity and cytotoxicity against human rhabdomyosarcoma cells was also measured and their allergenic properties have been studied by using 58 individual sera of patients sensitized to Aspergillus. Aspf1Delta(7-22) lacks cytotoxicity and shows a remarkably reduced IgE reactivity. From these studies it can be concluded that the deleted beta-hairpin is involved in ribosome recognition and is a significant allergenic region. 相似文献

Conversion of local structural state of a protein from an α-helix to a β-strand is usually associated with a major change in the
tertiary structure. Similar changes were observed during the self assembly of amyloidogenic proteins to form fibrils, which
are implicated in severe diseases conditions, e.g., Alzheimer disease. Studies have emphasized that certain protein sequence
fragments known as chameleon sequences do not have a strong preference for either helical or the extended conformations.
Surprisingly, the information on the local sequence neighborhood can be used to predict their secondary at a high accuracy
level. Here we report a large scale-analysis of chameleon sequences to estimate their propensities to be associated with
different local structural states such as α -helices, β-strands and coils. With the help of the propensity information derived
from the amino acid composition, we underline their complexity, as more than one quarter of them prefers coil state over to
the regular secondary structures. About half of them show preference for both α-helix and β-sheet conformations and either

The renin–angiotensin system (RAS), critically involved in the control of blood pressure and volume homeostasis, is a dual system comprising a circulating component and a local tissue component. The rate limiting enzyme is renin, which in the circulating RAS derives from the kidney to generate Ang II, which in turn regulates cardiovascular function by binding to AT1 and AT2 receptors on cardiac, renal and vascular cells. The tissue RAS can operate independently of the circulating RAS and may be activated even when the circulating RAS is suppressed or normal. A functional tissue RAS has been identified in brain, kidney, heart, adipose tissue, hematopoietic tissue, gastrointestinal tract, liver, endocrine system and blood vessels. Whereas angiotensinsinogen, angiotensin converting enzyme (ACE), Ang I and Ang II are synthesized within these tissues, there is still controversy as to whether renin is produced locally or whether it is taken up from the circulation, possibly by the (pro)renin receptor. This is particularly true in the vascular wall, where expression of renin is very low. The exact function of the vascular RAS remains elusive, but may contribute to fine-tuning of vascular tone and arterial structure and may amplify vascular effects of the circulating RAS, particularly in pathological conditions, such as in hypertension, atherosclerosis and diabetes. New concepts relating to the vascular RAS have recently been elucidated including: (1) the presence of functionally active Ang-(1-7)-Mas axis in the vascular system, (2) the importance of the RAS in perivascular adipose tissue and cross talk with vessels, and (3) the contribution to vascular RAS of Ang II derived from immune and inflammatory cells within the vascular wall. To better understand the roles of gammadelta T cells in mucosal infection, we utilized Salmonella enterica serovar Typhimurium (Salmonella serovar Typhimurium) infection in cattle as it closely approximates Salmonella serovar Typhimurium-induced enterocolitis in humans. Protein and gene expression in alphabeta and gammadelta T cells derived from lymphatic ducts draining the gut mucosa in Salmonella serovar Typhimurium-infected calves were analyzed. In calves with enterocolitis, general gene expression trends in gammadelta T cells suggested subtle activation and innate response, whereas alphabeta T cells were relatively quiescent following Salmonella serovar Typhimurium infection. An increase in IL-2R alpha expression on gammadelta T cells from infected calves and results from in vitro assays suggested that gammadelta T cells were primed by Salmonella serovar Typhimurium LPS to better respond to IL-2 and IL-15. Together with gene expression trends in vivo, these data support early priming activation of target tissue gammadelta T cells during Salmonella serovar Typhimurium infection. 相似文献

Two Lower Liassic sections of the Banos de Alhama de Granada outcrop are studied. The Hettangian and Sinemurian ammonite assemblages are detailed and a reinterpretation of the stratigraphy of the outcrop made.The ammonites collected show a clear mediterranean character. A biostratigraphy for the Hettangian and Sinemurian can be made but the correlation with the northeuropean zonal divisions and chronostratigraphy is rather difficult. These are the only west mediterranean sections where a succession of Hettangian ammonite assemblages have been made. 相似文献

Mesenchymal stem cells (MSCs) have been isolated from a variety of tissues, such as bone marrow, skeletal muscle, dental pulp, bone, umbilical cord and adipose tissue. MSCs are used in regenerative medicine mainly based on their capacity to differentiate into specific cell types and also as bioreactors of soluble factors that will promote tissue regeneration from the damaged tissue cellular progenitors. In addition to these regenerative properties, MSCs hold an immunoregulatory capacity, and elicit immunosuppressive effects in a number of situations. Not only are they immunoprivileged cells, due to the low expression of class II Major Histocompatibilty Complex (MHC-II) and costimulatory molecules in their cell surface, but they also interfere with different pathways of the immune response by means of direct cell-to-cell interactions and soluble factor secretion. In vitro, MSCs inhibit cell proliferation of T cells, B-cells, natural killer cells (NK) and dendritic cells (DC), producing what is known as division arrest anergy. Moreover, MSCs can stop a variety of immune cell functions: cytokine secretion and cytotoxicity of T and NK cells; B cell maturation and antibody secretion; DC maturation and activation; as well as antigen presentation. It is thought that MSCs need to be activated to exert their immunomodulation skills. In this scenario, an inflammatory environment seems to be necessary to promote their effect and some inflammation-related molecules such as tumor necrosis factor-α and interferon-γ might be implicated. It has been observed that MSCs recruit T-regulatory lymphocytes (Tregs) to both lymphoid organs and graft. There is great controversy concerning the mechanisms and molecules involved in the immunosuppressive effect of MSCs. Prostaglandin E2, transforming growth factor-β, interleukins- 6 and 10, human leukocyte antigen-G5, matrix metalloproteinases, indoleamine-2,3-dioxygenase and nitric oxide are all candidates under investigation. In vivo studies have shown many discrepancies regarding the immunomodulatory properties of MSCs. These studies have been designed to test the efficacy of MSC therapy in two different immune settings: the prevention or treatment of allograft rejection episodes, and the ability to suppress abnormal immune response in autoimmune and inflammatory diseases. Preclinical studies have been conducted in rodents, rabbits and baboon monkeys among others for bone marrow, skin, heart, and corneal transplantation, graft versus host disease, hepatic and renal failure, lung injury, multiple sclerosis, rheumatoid arthritis, diabetes and lupus diseases. Preliminary results from some of these studies have led to human clinical trials that are currently being carried out. These include treatment of autoimmune diseases such as Crohn's disease, ulcerative colitis, multiple sclerosis and type 1 diabetes mellitus; prevention of allograft rejection and enhancement of the survival of bone marrow and kidney grafts; and treatment of resistant graft versus host disease. Determining the potential range of invasive alien species under current conditions is important. However, we also need to consider future distributions under scenarios of climate change and different management interventions when formulating effective long‐term intervention strategies. This paper combines niche modelling and fine‐scale process‐based modelling to define regions at high risk of invasion and simulate likely dynamics at the landscape scale. Our study species is Schinus molle (Peruvian pepper tree; Anacardiaceae), a native of central South America, introduced to South Africa in about 1850 where it was widely planted along roads. Localities of planted and naturalized trees were mapped along 5380 km of roads – a transect that effectively samples a large part of western South Africa. Correlative modelling was used to produce profiles of present and future environmental conditions characterizing its planted and naturalized ranges. A cellular‐automata simulation model was used to estimate the dynamics of S. molle under future climates and different management scenarios. The overall potential range of S. molle in the region is predicted to shrink progressively with predicted climate change. Some of the potential range of S. molle defined based on current conditions (including areas where it is currently highly invasive) is likely to become less favourable. The species could persist where it is well established long after conditions for recruitment have deteriorated. Some areas where the species is not widely naturalized now (notably the fynbos biome) are likely to become more favourable. Our modelling approach allows for the delineation of areas likely to be invaded in future by considering a range of factors at different scales that mediate the interplay of climatic variables and other drivers that define the dimensions of human intervention such as distance from planted trees and the density of planted plants, both of which affect propagule pressure. 相似文献
