Bent DNA sites promote the curvature of DNA in both eukaryotic and prokaryotic chromosomes. Here, we investigate the localization
and structure of intrinsically bent DNA sites in the extensively characterized Drosophila melanogaster third chromosome DAFC-66D segment (Drosophila amplicon
in the follicle cells). This region contains the amplification control element ACE3, which is a replication enhancer that acts in cis to activate the major replication origin ori-β. Through both electrophoretic and in silico analysis, we have identified three major bent DNA sites in DAFC-66D. The bent DNA site (b1) is localized in the ACE3 element, whereas the other two bent DNA sites (b2 and b3) are localized in the ori-β region. Four additional bent DNA sites were identified in the intron of the S18 gene and near the TATA box of the S15, S19, and S16 genes. The identification of DNA bent sites in genomic regions previously characterized as functionally relevant for DNA
amplification further supports a function for DNA bent sites in DNA replication in eukaryotes.

The conditions of storage, cultivation and maintenance of microbial cultures should preserve the microbiological homogeneity, phenotypic and genotypic characteristics to ensure better reproducibility of metabolic production. To evaluate the influence of the storage condition on the composition of cell fatty acids, genetic profile and biochemical characteristics of Xanthomonas campestris pv. mangiferaeindicae IBSBF 2103, as well as, to identify its relationship with the yielding and viscosity of the xanthan gum produced, this study monitored the strain preserved in two simple and widely used conditions, ultra-freezer (?80 °C) and refrigeration (3–8 °C) during 5 months. Were identified and quantified 13 fatty acids. The cells preserved at ?80 °C showed more stable concentration of all fatty acids, producing more xanthan gum and with higher viscosity. The chromosomal analysis obtained with the enzyme XbaI revealed 17 distinct fragments with maximum size of 485 kilobases, without variations among the subcultures maintained in both storage conditions. The X. campestris pv. mangiferaeindicae subcultures preserved at ?80 °C showed less pronounced phenotypic variations, which had positive influence in the qualitative and quantitative characteristics of the xanthan gum produced. 相似文献

Leaf-litter decomposition is a major component of carbon and nutrient dynamics in tropical forest ecosystems, and moisture availability is widely considered to be a major influence on decomposition rates. Here, we report the results of a study of leaf-litter decomposition of five tree species in response to dry-season irrigation in a tropical forest regrowth stand in the Brazilian Amazon; three experiments differing in the timing of installation and duration allowed for an improved resolution of irrigation effects on decomposition. We hypothesized that decomposition rates would be faster under higher moisture availability in the wet season and during dry-season irrigation periods in the treatment plots, and that decomposition rates would be faster for species with higher quality leaves, independent of treatment. The rates of decomposition ( k ) were up to 2.4 times higher in irrigated plots than in control plots. The highest k values were shown by Annona paludosa (0.97 to 1.26/yr) while Ocotea guianensis (0.73 to 0.85/yr) had the lowest values; intermediate rates were found for Lacistema pubescens (0.91 to 1.02/yr) and Vismia guianensis (0.91 to 1.08/yr). These four tree species differed significantly in leaf-litter quality parameters (nitrogen, phosphorus, lignin, and cellulose concentrations, as well as lignin:nitrogen and carbon:nitrogen ratios), but differences in decomposition rates among tree species were not strictly correlated with leaf-litter quality. Overall, our results show that dry-season moisture deficits limit decomposition in Amazonian forest regrowth. 相似文献

BackgroundWhole breast irradiation reduces loco-regional recurrence and risk of death in patients submitted to breast-conserving treatment. Data show that radiation to the index quadrant alone may be enough in selected patients.AimTo report the experience with intra-operative radiotherapy (IORT) with Electron-beam Cone in Linear Accelerator (ELIOT) and the results in overall survival, local control and late toxicity of patients submitted to this treatment.Materials and Methods147 patients treated with a median follow up of 6.9 years (0.1?11.5 years). The actuarial local control and overall survival probabilities were estimated using the Kaplan Meier method. All tests were two-sided and p ? 0.05 was considered statistically significant.ResultsOverall survival of the cohort in 5 years, in the median follow up and in 10 years was of 98.3%, 95.1% and 95.1%, respectively, whereas local control in 5 years, in the median follow up and in 10 years was of 96%, 94.9% and 89.5%, respectively. Two risk groups were identified for local recurrence depending on the estrogen or progesterone receptors, axillary or margin status and lymphovascular invasion (LVI) (p = 0.016).ConclusionsIORT is a safe and effective treatment. Olfactomedin is a secreted polymeric glycoprotein of unknown function,originally discovered at the mucociliary surface of the amphibian olfactoryneuroepithelium and subsequently found throughout the mammalian brain. As afirst step toward elucidating the function of olfactomedin, itsphylogenetic history was examined to identify conserved structural motifs.Such conserved motifs may have functional significance and provide targetsfor future mutagenesis studies aimed at establishing the function of thisprotein. Previous studies revealed 33% amino acid sequence identity betweenrat and frog olfactomedins in their carboxyl terminal segments. Furtheranalysis, however, reveals more extensive homologies throughout themolecule. Despite significant sequence divergence, cysteines essential forhomopolymer formation such as the CXC motif near the amino terminus areconserved, as is the characteristic glycosylation pattern, suggesting thatthese posttranslational modifications are essential for function.Furthermore, evolutionary analysis of a region of 53 amino acids of fish,frog, rat, mouse, and human olfactomedins indicates that an ancestralolfactomedin gene arose before the evolution of terrestrial vertebrates andevolved independently in teleost, amphibian, and mammalian lineages.Indeed, a distant olfactomedin homolog was identified in Caenorhabditiselegans. Although the amino acid sequence of this invertebrate protein islonger and highly divergent compared with its vertebrate homologs, theprotein from C. elegans shows remarkable similarities in terms of conservedmotifs and posttranslational modification sites. Six universally conservedmotifs were identified, and five of these are clustered in the carboxylterminal half of the protein. Sequence comparisons indicate that evolutionof the N-terminal half of the molecule involved extensive insertions anddeletions; the C-terminal segment evolved mostly through point mutations,at least during vertebrate evolution. The widespread occurrence ofolfactomedin among vertebrates and invertebrates underscores the notionthat this protein has a function of universal importance. Furthermore,extensive modification of its N-terminal half and the acquisition of aC-terminal SDEL endoplasmic-reticulum- targeting sequence may have enabledolfactomedin to adopt new functions in the mammalian central nervoussystem. 相似文献

Sex steroid hormones are critical in gonadal differentiation in turtles. The gonads are not the only organs responsible for producing these hormones during this phase. Mesonephros play an important role in steroidogenesis. The present study aimed to investigate the presence of steroidogenic cells in mesonephros of Podocnemis expansa during gonadal differentiation and to evaluate their morphology and ultrastructure. Ten embryos of P. expansa were collected from 5 nests on day 36 of incubation, during spawning period on an artificial beach. Embryos were extracted from eggs by slicing the shell and euthanized. They were dissected under a stereoscopic microscope to collect the gonad-mesonephro complex, in which were fixed and subsequently processed for light microscopy, immunohistochemistry and transmission electron microscopy analysis. During histological analysis was observed mesonephros has typical morphological structure. Immunohistochemistry showed immunoreaction to aromatase in cells of intertubular space. Confirming these findings, it was possible to observe a type of intertubular cell in several regions of mesonephro, being more predominant in region close to blood vessels, distal and proximal tubules. In ultrastructural analysis these cells were characterized by having a clear, large, and rounded nucleus with evident nucleolus and cytoplasm rich in electron-dense droplets. This study demonstrated for the first time the presence of cells with morphological, immunohistochemical and ultrastructural characteristics similar to steroid-producing cells in P. expansa mesonephrons, suggesting that this organ may contribute to gonadal differentiation in this species. 相似文献

Orthodontic force compresses the periodontal ligament promoting the expression of pro-inflammatory mediators and matrix metalloproteinases
responsible for tooth movement. The extent in time while periodontal cells are being treated and the increment in the amount
of mechanical stress caused by the orthodontic force is thought to regulate the levels of metalloproteinases in the periodontal
tissue. To study the possible regulation in the activity of metalloproteinases 2, 3, 7, 9, and 10 by simulated orthodontic
force, human periodontal ligament fibroblast cultures were centrifuged (141×g) for 30, 60, 90, and 120 min, simulating the orthodontic force. Cell viability, protein quantification, and activity of metalloproteinases
by zymography were evaluated at 24, 48, and 72 h after centrifugation in both cell lysates and growth medium. The activity
of the 72-kDa matrix metalloproteinase 2 was decreased at 24 h regardless of the duration of centrifugation and at 48 h in
cells centrifuged for 30 min only. Decrease in the amount of total protein in lysates was seen at 48 and 72 h with no change
in cell viability. The data seem to indicate that the amount of mechanical stress regulates the levels of secreted matrix
metalloproteinase 2. In addition, the centrifugation as a model for simulated orthodontic force may be used as a simple and
reliable method to study the role played by matrix metalloproteinases in periodontal ligament when submitted to mechanical

Sex differences in animal coloration often result from sex‐dependent regulatory mechanisms. Still, some species exhibit incomplete sexual dimorphism as females carry a rudimentary version of a costly male trait, leading to intralocus sexual conflict. The underlying physiology and condition dependence of these traits can inform why such conflicts remain unresolved. In eastern fence lizards (Sceloporus undulatus), blue iridophore badges are found in males and females, but melanin pigmentation underneath and surrounding badges is male‐exclusive. We track color saturation and area of badges across sexual maturity, and their relationship to individual quality (body condition and immunocompetence) and relevant hormones (testosterone and corticosterone). Saturation and testosterone were positively correlated in both sexes, but hormone and trait had little overlap between males and females. Saturation was correlated with body condition and immunocompetence in males but not in females. Co‐regulation by androgens may have released females from resource allocation costs of color saturation, even when in high condition. Badge area was independent of testosterone, but associated with low corticosterone in females, indicating that a nonsex hormone underlies incomplete sexual dimorphism. Given the evidence in this species for female reproductive costs associated with ornamentation, this sex‐nonspecific regulation of an honest signal may underlie intralocus sexual conflict. 相似文献

ABSTRACT The reasons for the decline of bee diversity and abundance include the destruction and loss of natural habitats. Protected areas are created for biodiversity conservation, but these areas vary strongly in their level of vegetation disturbance. Using trap-nests, we assessed changes in solitary bee abundance, richness, and composition in areas ranging from naturally conserved to degraded. Solitary bees were sampled during an 18-month period in three areas of southeastern Brazil: a preserved area in Rio Preto State Park – PERP; a restored/altered area with exotic plants at the Federal University of the Jequitinhonha and Mucuri Valleys – JK Campus; and a degraded area in Biribiri State Park – PEBi. A total of seven species of bees built 115 nests. In the degraded area, only two nests were built. Abundance of built nests was higher in the preserved area (PERP), but diversity was higher in the restored area (JK Campus). Our results show that the solitary bee population responds positively to habitat complexity (local scale). The presence of a diverse solitary bee fauna in the restored area indicates that altered areas should also be protected as suitable areas for re-colonization of cavity-nesting bees. 相似文献
