The cardiac adrenoceptors of lower vertebrates were characterized in atrial preparations. Adrenaline (A) potentiated the force and frequency of contraction in the spontaneously beating atria of the frog, trout and flounder and in electrically paced atrial strips from the shark. The inotropic responses of A were most pronounced at the lower temperatures for the frog and trout, while A enhanced frequency to a greater extent at higher temperatures in the frog atria. Atrial alpha-receptors activated by A at 8 degrees C could not be detected in any of the species under study. The apparent affinities for the inotropic and chronotropic responses of agonist in the frog (15 degrees C) and trout (8 degrees C) atria were: Iso greater than Sal greater than or equal to A greater than NA. A cocaine-sensitive uptake for A and NA was apparent in these atria, consistent with sympathetic innervation. The affinities for the catecholamines in the flounder and shark atria were not increased by cocaine, in accordance with absence of sympathetic innervation of the atria in these species. These atria were also insensitive to corticosterone. The affinities for A and NA were on the other hand higher in the sympathetically non-innervated atria of the flounder than in the innervated atria of the frog and trout. The apparent orders of relative affinities for agonists were Iso greater than A = NA greater than Sal for the flounder, and of the relative potencies Iso = A greater than NA greater than Sal for the shark atrium. The results are consistent with the hypothesis that catecholamines enhance cardiac performance in lower vertebrates chiefly via "adrenaline" receptors which resemble the beta 2-type of mammalian adrenoceptors in many respects. Unlike that in mammals, cardiac adrenaline receptors in the frog and trout are activated by the sympathetic neurotransmitter ("innervated" receptors). On the other hand, the adrenaline receptors of the flounder and shark are responding to the circulating catecholamines ("humoral" receptors). However, the flounder atrium, with equal affinities for A and NA, appears as an exception to the rule by having a mixed population of humoral beta 1- and beta 2-adrenoceptors, indicating a role for circulating NA in cardiac regulation in this species.  相似文献

Summary The atrial adrenoceptors of the rainbow trout heart (Salmo gairdneri) were characterized in spontaneously beating preparations at 8 °C.The order of affinity for the inotropic and chronotropic responses was: Isoprenaline > adrenaline salbutamol > phenylephrine noradrenaline. Selective agonists and antagonists for mammalian - and 1-adrenoceptors were without effect on the trout preparations.A potent neuronal uptake for adrenaline and noradrenaline could be demonstrated by means of cocaine blockade, consistent with considerable sympathetic innervation of the trout atrium.The results show that catecholamines increase the force and frequency of the atrium via a single, 2-type of adrenoceptor. It is suggested that there is no distinction between the innervated and the humoral -adrenoceptors in the myocardium of this species.  相似文献

Abstract

A phosphotriester method for the synthesis of dithymidine phosphoromonothoates and phosphorodithioates with new S-protecting groups has been investigated. Four of the S-protecting groups possesed catalytic activity, however side reactions occurred during deprotection. The best S-protecting group was 4-chloro-2-nitlobenzyl which could be removed with a minimum of side reactions (0.3 %). The coupling reagent PyFNOP (14) gave protected dithymidine phosphoromonothioate in 96 % yield after 15 min coupling. Furthermore PyFNOP chemoselectively activates oxygen in nucleoside phosphorodithioate monomers 9 and can be used for the synthesis of oligodeoxynucleoside phosphorodithioates with mixed base sequences.  相似文献

Information about forest biodiversity has so far been collected mostly by using field inventories, but it is desirable to find other methods that can cover large areas at a lower cost. In a forest landscape covering 2000 ha in southern Sweden we tested if colour-infrared (CIR) aerial photographs on the scale of 1:30000 can be used to interpret forest stand characteristics correlated to the occurrence of epiphytic lichens that are Red-listed or otherwise indicate high nature conservation value (signal species). Using logistic regression we found that the interpreted stand variables tree height and crown structure class were significantly correlated to the occurrence of Red-listed species. For signal species, the variables tree height, percentage of southern deciduous trees and crown structure class were significantly correlated to the occurrence. The logistic regression models successfully predicted a significantly higher probability to find Red-listed species in the stands that actually contained such species compared to stands without Red-listed species. The same was true for stands with signal species. We conclude that interpretation of CIR aerial photographs could be a useful method to find certain groups of epiphytic lichens in surveys covering large areas.  相似文献

TGF-beta and Smad3 signaling link inflammation to chronic fibrogenesis   总被引:6,自引:0,他引:6  
Transient adenovirus-mediated gene transfer of IL-1beta (AdIL-1beta), a proinflammatory cytokine, induces marked inflammation and severe and progressive fibrosis in rat lungs. This is associated with an increase in TGF-beta1 concentration in bronchoalveolar lavage (BAL) fluid. TGF-beta1 is a key cytokine in the process of fibrogenesis, using intracellular signaling pathways involving Smad2 and Smad3. In this study we investigate whether inflammation induced by IL-1beta is able to independently induce lung fibrosis in mice deficient in the Smad3 gene. Seven days after AdIL-1beta administration, similar levels of IL-1beta transgene are seen in BAL in both wild-type (WT) and knockout (KO) mice, and BAL cell profiles demonstrated a similar marked neutrophilic inflammation. Phospho-Smad2 staining was positive in areas of inflammation in both WT and KO mice at day 7. By day 35 after transient IL-1beta expression, WT mice showed marked fibrosis in peribronchial areas, quantified by picrosirius red staining and morphometry. However, there was no evidence of fibrosis or collagen accumulation in IL-1beta-treated KO mice, and peribronchial areas were not different from KO mice treated with the control adenovector. TGF-beta1 and phospho-Smad2 were strongly positive at day 35 in fibrotic areas observed in WT mice, but no such staining was detectable in KO mice. The IL-1beta-induced chronic fibrotic response in mouse lungs is dependent on Smad3. KO and WT animals demonstrated a similar inflammatory response to overexpression of IL-1beta indicating that inflammation must link to the Smad3 pathway, likely through TGF-beta, to induce progressive fibrosis.  相似文献

The perennial herbaceous crop Arundo donax is a potential feedstock for second-generation bioethanol production. In the present work, two different process options were investigated for the conversion of two differently steam-pretreated batches of A. donax. The pretreated raw material was converted to ethanol with a xylose-consuming Saccharomyces cerevisiae strain, VTT C-10880, by applying either separate hydrolysis and fermentation (SHF) or simultaneous saccharification and fermentation (SSF). The highest overall ethanol yield and final ethanol concentration were achieved using SHF (0.27 g g?1 and 20.6 g L?1 compared to 0.24 g g?1 and 19.0 g L?1 when SSF was used). The performance of both SHF and SSF was improved by complementing the cellulolytic enzymes with hemicellulases. The higher amount of acetic acid in one of the batches was shown to strongly affect xylose consumption in the fermentation. Only half of the xylose was consumed when batch 1 (high acetic acid) was fermented, compared to that 94% of the xylose was consumed in fermentation of batch 2 (lower acetic acid). Furthermore, the high amount of xylooligomers present in the pretreated materials considerably inhibited the enzymatic hydrolysis. Both the formation of xylooligomers and acetic acid thus need to be considered in the pretreatment process in order to achieve efficient conversion of A. donax to ethanol.  相似文献

Background

Micro-computed tomography (micro-CT) is a novel tool for monitoring acute and chronic disease states in small laboratory animals. Its value for assessing progressive lung fibrosis in mice has not been reported so far. Here we examined the importance of in vivo micro-CT as non-invasive tool to assess progression of pulmonary fibrosis in mice over time.

Methods

Pulmonary fibrosis was induced in mice by intratracheal delivery of an adenoviral gene vector encoding biologically active TGF-ß1 (AdTGF-ß1). Respiratory gated and ungated micro-CT scans were performed at 1, 2, 3, and 4 weeks post pulmonary adenoviral gene or control vector delivery, and were then correlated with respective histopathology-based Ashcroft scoring of pulmonary fibrosis in mice. Visual assessment of image quality and consolidation was performed by 3 observers and a semi-automated quantification algorithm was applied to quantify aerated pulmonary volume as an inverse surrogate marker for pulmonary fibrosis.

Results

We found a significant correlation between classical Ashcroft scoring and micro-CT assessment using both visual assessment and the semi-automated quantification algorithm. Pulmonary fibrosis could be clearly detected in micro-CT, image quality values were higher for respiratory gated exams, although differences were not significant. For assessment of fibrosis no significant difference between respiratory gated and ungated exams was observed.

Conclusions

Together, we show that micro-CT is a powerful tool to assess pulmonary fibrosis in mice, using both visual assessment and semi-automated quantification algorithms. These data may be important in view of pre-clinical pharmacologic interventions for the treatment of lung fibrosis in small laboratory animals.  相似文献

Northern ecosystems are experiencing some of the most dramatic impacts of global change on Earth. Rising temperatures, hydrological intensification, changes in atmospheric acid deposition and associated acidification recovery, and changes in vegetative cover are resulting in fundamental changes in terrestrial–aquatic biogeochemical linkages. The effects of global change are readily observed in alterations in the supply of dissolved organic matter (DOM)—the messenger between terrestrial and lake ecosystems—with potentially profound effects on the structure and function of lakes. Northern terrestrial ecosystems contain substantial stores of organic matter and filter or funnel DOM, affecting the timing and magnitude of DOM delivery to surface waters. This terrestrial DOM is processed in streams, rivers, and lakes, ultimately shifting its composition, stoichiometry, and bioavailability. Here, we explore the potential consequences of these global change‐driven effects for lake food webs at northern latitudes. Notably, we provide evidence that increased allochthonous DOM supply to lakes is overwhelming increased autochthonous DOM supply that potentially results from earlier ice‐out and a longer growing season. Furthermore, we assess the potential implications of this shift for the nutritional quality of autotrophs in terms of their stoichiometry, fatty acid composition, toxin production, and methylmercury concentration, and therefore, contaminant transfer through the food web. We conclude that global change in northern regions leads not only to reduced primary productivity but also to nutritionally poorer lake food webs, with discernible consequences for the trophic web to fish and humans.  相似文献

This study was conducted to explore the potency of morphine to induce reductions of specific cytochrome P450 isoenzyme functions. Male Sprague-Dawley rats were treated with escalating doses (20-125 mg/kg per day) of morphine for 2 weeks in order to study the effects on the following cytochrome P450 catalyzed reactions: 16 alpha-hydroxylation of dehydroepienderosterone (DHA) and progesterone; 17 alpha- and 21-hydroxylation of progesterone; N-demethylation of ethymorphine, codeine and morphine as well as O-dealkylation of ethylmorphine and codeine. 16 alpha-Hydroxylation of DHA and progesterone and 17 alpha-hydroxylation of progesterone decreased to 18, 12 and 10% of control activities, respectively. The N-demethylation of ethylmorphine and codeine decreased to 34 and 43% of control activities, respectively. Morphine treatment had no effect on the 21-hydroxylation reactions or the O-dealkylation of ethylmorphine or codeine. A monoclonal antibody (Mab) against rat liver cytochrome P450 2 c/RLM 5 exerted a 66-73% inhibition of the N-demethylation of ethylmorphine and codeine, respectively, whereas the O-dealkylation reactions were not affected. This Mab inhibited the 16 alpha- and 17 alpha-hydroxylation of DHA and progesterone, whereas the 21-hydroxylation reactions were unaffected. The steroid hydroxylation reactions in rat adrenals were not altered upon morphine treatment. Our data suggest that a major part of the 16 alpha- and 17 alpha-steroid hydroxylations are catalyzed by the same (or closely related) cytochrome(s) P450 as the opioid N-demethylation reactions.  相似文献
