Photoinduced changes of structural and functional properties of lactatedehydrogenase isoenzymes from human erythrocytes in free state and in the presence of serotonin have been studied by means of gel chromatography, electrophoresis, IR-spectrophotometry and by the method of definition of catalytic activity. UV-light influence induces photoinactivation of erythrocyte's LDH, while its inhibitory effect intensifies with the increase of irradiation dose. The complicated character of changes in electrophoretic mobility and percentage content of isoenzymes LDH-1, LDH-2, LDH-3 under the influence of UV-rays testifies that the decrease of total enzyme activity of these isoforms in connected with their different photosensitiveness and represents the result of many-staged process which is characterized both by the consistent and parallel proceeding of its individual photochemical reactions. A pronounced photoprotective effect of serotonin towards the molecules of erythrocytic LDH isoenzymes has been discovered. It seems to be caused by formation of enzyme--biogenous amine complex affecting the secondary protein structure.  相似文献

The changes of photo- and peroxide resistance of erythrocytic and lymphocytic cells of human under influence of UV and some active oxygen forms in presence of biogenic amines such as serotonin, dopamine, adrenalin, histamine had been investigated. While investigating the degree of photohemolysis of erythrocytes it was discovered that biogenic compounds raise UV stability of erythrocytic membranes. By using the method of chemiluminescence it was established that biogenic amines increased the degree of peroxide resistance of human erythrocytic and lymphocytic cells. The decrease of the level of erythrocytic diene conjugates under influence of UV by serotonin and histamine was also discovered.  相似文献

During oxydase reaction spectral characteristics of ceruloplasmin at absorption of copper ions and protein part of the molecule are shown to change. It has been ascertained, that when irradiating ceruloplasmin by UV-light the functioning of intramolecular electron transport chain is broken, the degree of positive cooperativity (a Hill's constant) on substrate decreases. It is supposed, that these changes are caused by disturbance of interdomain interactions in a protein molecule.  相似文献

The effect of staining with commonly used nuclear dyes (orsein, light green, methylene blue, azure-eosin by Romanovsky-Giemsa) on the frequency of occurrence of nuclear disorders (micronuclei, perinuclear vacuoles, notches, protrusions, such as "tongue" and "broken egg") in human buccal mucosa was determined. It is shown that Romanovsky-Giemsa dye is the best type of dye both in the quality of the micropreparations and the spectrum of detected abnormalities.  相似文献

It has been shown by classical biophysical and biochemical methods in combination with atomic microscopy that lipase from Rhizopus niveus exists in a water solution as a dimer with a molecular weight of 96 kDa. The rate of splitting of triglycerides by a dimeric molecules is twice that of monomers. The heat stability of the monomeric form of lipase at temperatures of 20-60 degrees C is significantly higher than that of the native molecule.  相似文献

Cytogenetic characteristics of the seed progeny of birch (Betula pendula Roth), growing in colony Urazovo Belgorod region exposed by the impact of Chernobyl precipitation in 1986, were determinated. The changing of cytogenetic characteristics in comparison with the control (mitotic index and level of mitosis pathologies grown, their spectrum widens part of persistent nucleolies at the stages of metaphase, anaphase, telophase of mitosis enlarges, square of surface of single nucleolies decreases, part of moderate-active nucleolies "bark-core vacuolisated" type increase) on the experimental squares is revealed. The most considerable effects were observed in 2000, which connected with the increasing of the contaminations of mentioned territory as a result of brick factory work. By means of cluster analysis methods it was established that the cleanest in northwestern part of colony Urazovo, the most contaminated is central part. It was purposed, that chemical compounds, are main agents caused the changing of cytogenetic properties of test-object after the normalization of the radiation level.  相似文献

The effect of UV radiation on the spectral characteristics of human nitrosohemoglobin in a wide dose range (151-4530 J/m2) was studied. It was shown that the irradiation of hemoprotein solutions with low doses of UV light (151-453 J/m2) led to local conformational rearrangements of the iron porphyrin moiety of the molecule. The apoprotein exhibited a high photostability under the conditions of the experiment. High doses of UV radiation (1359-4530 J/m2) induced the accumulation of methemoglobin in the protein sample. A scheme of the photoinduced formation of methemoglobin was elaborated.  相似文献

The contribution of hem and globin components of electrophoretic fractions of UV-irradiated human carboxyhemoglobin to photodestruction of the protein was studied. The changes observed are the result of summation of some processes unequal in intensity and direction that take place in microheterogeneous media of photomodified protein. Photosensitivity of hemoproteid in electrophoretic fraction depends on apoprotein condition, whereas the hem photoresistance cannot be the evidence of the photostability of the whole molecule.  相似文献
