The thermodynamic and kinetic aspects of molecular recognition for the methotrexate (MTX)-dihydrofolate reductase (DHFR) ligand-protein system are investigated by the binding energy landscape approach. The impact of 'hot' and 'cold' errors in ligand mutations on the thermodynamic stability of the native MTX-DHFR complex is analyzed, and relationships between the molecular recognition mechanism and the degree of ligand optimization are discussed. The nature and relative stability of intermediates and thermodynamic phases on the ligand-protein association pathway are studied, providing new insights into connections between protein folding and molecular recognition mechanisms, and cooperativity of ligand-protein binding. The results of kinetic docking simulations are rationalized based on the thermodynamic properties determined from equilibrium simulations and the shape of the underlying binding energy landscape. We show how evolutionary ligand selection for a receptor active site can produce well-optimized ligand-protein systems such as MTX-DHFR complex with the thermodynamically stable native structure and a direct transition mechanism of binding from unbound conformations to the unique native structure.  相似文献

Elevated levels of serum saturated fatty acid palmitate have been shown to promote insulin resistance, increase cellular ROS production, and trigger cell apoptosis in hepatocytes during the development of obesity. However, it remains unclear whether palmitate directly impacts the circadian clock in hepatocytes, which coordinates nutritional inputs and hormonal signaling with downstream metabolic outputs. Here we presented evidence that the molecular clock is a novel target of palmitate in hepatocytes. Palmitate exposure at low dose inhibits the molecular clock activity and suppresses the cyclic expression of circadian targets including Dbp, Nr1d1 and Per2 in hepatocytes. Palmitate treatment does not seem to alter localization or reduce protein expression of BMAL1 and CLOCK, the two core components of the molecular clock in hepatocytes. Instead, palmitate destabilizes the protein-protein interaction between BMAL1-CLOCK in a dose and time-dependent manner. Furthermore, we showed that SIRT1 activators could reverse the inhibitory action of palmitate on BMAL1-CLOCK interaction and the clock gene expression, whereas inhibitors of NAD synthesis mimic the palmitate effects on the clock function. In summary, our findings demonstrated that palmitate inhibits the clock function by suppressing SIRT1 function in hepatocytes.  相似文献

There are few insecticidal options for potato tuber moth (PTM), Phthorimaea operculella Zeller (Lepidoptera: Gelechiidae), control late in the growing season. We evaluated the PTM granulovirus (PoGV) and Bacillus thuringiensis Berliner ssp. kurstaki (Btk) for season‐long control of PTM on potato foliage in 2006 and 2007. Compared to untreated controls, 10 weekly applications of PoGV (1013 occlusion bodies/ha) reduced PTM populations in replicated 1‐m3 field cages by 86–96% on pre‐harvest foliage and 90–97% on tubers added to cages shortly before harvest. Infection rates of 82–95% of L4 larvae by PoGV were noted within individual larval cohorts. Equivalently timed Btk treatments (1.12 kg product/ha) were significantly less effective at population suppression, with a 36–76% reduction in larvae recovered from tubers added to cages. A PoGV/Btk alternation was significantly more effective than Btk alone and as effective as PoGV in 2007, but not in 2006. There was some evidence that reduced rate PoGV treatments (10% rate or 50% application frequency) were less effective than the standard program. There were no treatment effects on percentage of tubers growing in the ground that were infested at harvest, which remained comparatively low at ≤8.1%. Bioassays were conducted to evaluate the residual activities of foliar deposits. Early‐season applications were highly effective for the first 24 h (≥93% mortality) with a steady decline in activity over 10 days. A second application, applied later in the season, showed similar patterns, although in this case Btk was less persistent than PoGV, whereas both agents provided significant larval mortality compared with controls over 14 days. Both PoGV and Btk provide alternatives to manage field infestations of PTM prior to harvest, thus reducing the risk of tuber infestations in storage.  相似文献

Commercial formulations of the codling moth, Cydia pomonella L., granulovirus (CpGV) are limited by their short residual activity under orchard conditions in the Pacific Northwest. We evaluated spray-dried lignin-encapsulated formulations of CpGV for improved solar stability based on laboratory bioassays with a solar simulator and in field tests in an infested apple orchard. In laboratory tests, aqueous lignin formulations containing a high dosage of 3 x 10(10) occlusion bodies (OB)/L, with and without the additives titanium dioxide (TiO(2)) and sugar, provided significant solar protection of virus, i.e., mortality of codling moth exposed to lignin formulations that had been irradiated with 9.36 x 10(6) joules/m(2) was 92-94%, compared with 66-67% from a glycerin-stabilized product (Cyd-X) or suspension of pure unformulated virus at the same rates. By comparison, a lower dosage of the lignin formulation (3 x 10(8)OB/L) did not provide significant solar protection. Equivalent dosage-dependent patterns in solar protection were observed in further tests with the lignin formulation, when an intermediate (3 x 10(9)OB/L) as well as the low dosage provided no solar protection. Equivalent rates of a blank lignin formulation (containing no virus) did not affect larval mortality, suggesting a protective effect of the lignin on the virus at the high rate. The use of several spray adjuvants, 'NuFilm-17' and 'Organic Biolink' (sticker-spreaders at 0.06% v/v), 'Raynox' (sunburn protectant at 5% v/v), and 'Trilogy'(neem oil at 1% v/v) did not provide solar protection of a commercial CpGV preparation in laboratory tests. In season long orchard tests (Golden Delicious), the lignin formulation of CpGV applied at 6.57 x 10(12)OB/ha did not significantly improve control of codling moth or protection of fruit compared with Cyd-X at equivalent rates. Our studies show that lignin-based CpGV formulations provided solar protection at relatively high virus dosages. The testing of lignin formulations containing reduced virus concentrations may allow virus solar protection to be achieved at more economical rates.  相似文献

The identification of effective solar protectants for field application of the codling moth, Cydia pomonella (L.), granulovirus (CpGV) is of interest to improve its efficacy and commercial viability as a biological pesticide. We evaluated several materials as potential adjuvants to protect CpGV from ultraviolet degradation. In laboratory tests with a solar simulator (9.36×106 J/m2), the addition of kaolin clay (Surround® WP at 3 and 6%, w/v), a paraffin wax-based emulsion (SPLAT? at 5%, v/v), and a bark extract trans-cinnamaldehyde combined with a film agent (both at 1%, v/v) did not significantly reduce larval entries or increase larval mortality in irradiated apples that were treated with a commercial CpGV product (Cyd-X). In semi-field tests in an apple orchard, a spray-dried lignin formulation containing CpGV (6.57×1012 OBs/ha) and a lignin-based adjuvant used with Cyd-X (both applied at 4.7–5.6 kg lignin/ha) significantly improved residual activity of CpGV compared with Cyd-X alone applied at the same rate. However, the benefits were short lived and could not be detected after 7 days. In orchard tests, we evaluated two additional refined lignin-products (Lignosulfonate and Vanisperse? CB at 5.61 kg/ha) and two particle film materials (kaolin clay, ‘Cocoon?’, and calcium with boron ‘Eclipse?’) as adjuvants for UV protection of Cyd-X (6.57×1012 OBs/ha) in tests against a dense codling moth infestation. Although all virus treatments were highly effective (causing ≥90% larval mortality), no significant effects of the adjuvant treatments could be detected. In these latter tests, the use of a silicone based wetting agent at 0.025% (v/v) may have been beneficial at increasing mortality among older larvae inside the fruit.  相似文献

We evaluated biopesticides based on entomopathogenic fungi, azadirachtin and horticultural oils for the management of the chilli thrips, Scirtothrips dorsalis Hood, an economically significant invasive pest in the United States. Insecticides were applied four times at 7‐ to 10‐day intervals against established S. dorsalis infestations on shrub roses KnockOut®, Rosa x ‘Radrazz’ under simulated nursery conditions. When applied as stand‐alone treatments, Beauveria bassiana GHA (BotaniGard® ES), Metarhizium brunneum F52 (Met‐52 EC), a horticultural oil (SuffOil‐X®) and azadirachtin (Molt‐X®) at label rates provided significant control, reducing populations of S. dorsalis by 48–71% compared with control over 4–6 weeks. Similar results were observed when the biopesticides were applied in rotation with each other. A conventional standard, spinosad (Conserve® SC), was consistently the most effective treatment in these studies, reducing thrips populations by >95% overall. In another study, more effective control (87%–92%) was achieved in biopesticide rotation programmes that included spinosad, when compared with those that did not. Results also showed that these biopesticides can be tank‐mixed. However, there was no evidence that B. bassiana or M. brunneum combined with azadirachtin resulted in additive or synergistic control, as neither tank‐mix treatment improved control compared with azadirachtin alone. These findings highlight the potential use of biopesticides in rotation programmes with conventional insecticides to manage S. dorsalis on roses. Biopesticides evaluated in this study can be incorporated into an IPM programme for roses.  相似文献

Large quantities of insecticides are used on warm season turfgrasses to combat pest infestations. To investigate the potential for microbial control, we screened commercially available entomopathogenic nematode products against Herpetogramma phaeopteralis Guenée, an economically injurious pest in the south‐eastern United States and Caribbean islands. All tested products, based on Steinernema carpocapsae, S. feltiae, Heterorhabditis bacteriophora, H. megidis and H. indica, were pathogenic to H. phaeopteralis larvae in the laboratory, but S. carpocapsae caused the highest mortality. Amongst nematode species, median lethal concentration (LC50) was not different for three different larval sizes (based on 95% CL) with the exception of H. indica, which had higher LC50 for small larvae. The number of infective juvenile stages (IJs) produced per White trap was significantly greater from larvae infected by Hbacteriophora and least for those infected by H. indica. A proprietary formulation of S. carpocapsae ‘Millenium®’ was chosen for further greenhouse experiments. Overall, the neonicotinoid insecticide clothianidin provided the best control, but greenhouse experiments also revealed that the label rate of Millenium (106 IJ/l at 2500 l/ha) reduced webworm populations by 83–93% and was as effective as clothianidin against larger‐size larvae. Our data suggest that commercial formulations of S. carpocapsae can be a good option for H. phaeopteralis biocontrol, but further field studies are warranted to confirm effectiveness under different environmental scenarios.  相似文献

Gasoline spills were simulated in the laboratory with three air‐dried soils, using a synthetic gasoline and unsaturated soil to quantify passive volatilization over a period of up to 16 d. The total and individual gasoline components were monitored as a function of time and depth in the soil.

The time required to deplete the overall gasoline concentration in the soil to 40% of the initial concentration ranged from 0.25 to 10 d for the three soils. Sand was the fastest, followed by loamy sand and silt loam. The volatilization rate of gasoline from soil was found to be dependent on soil, chemical type, and depth. Observation of individual components indicated that a wicking mechanism contributed to the gasoline flux toward the surface.  相似文献
