Two proteases, designated I and II, have been isolated from sporulating cells of Bacillus subtilis. They were partially purified by ammonium sulfate fractionation, Sephadex chromatography and affinity columns. Protease I was found to be similar to an already characterized B. subtilis protease. Protease II is trypsin-like in its substrate specificity and is distinct from protease I in its pH optimum, pH stability, molecular weight, substrate specificity, heat stability and sensitivity to various inhibitors. While both enzymes were produced primarily during sporulation, they attained maximum levels of activity at different times. Distinct functions for these proteases in post exponential B. subtilis are likely. 相似文献

BmK M4 is a neutral neurotoxin in the BmK toxin series.It is medially toxic and belongs to group III α-toxins.The purified sample was crystallized in rhombic space group P61.Using an X-ray diffraction technique,the crystal structure of BmK M4 was revealed by molecular replacement at 0.20 nm resolution.The model was refined.The final crystallographic R factor was 0.142 and the free R factor was 0.173.The root mean square deviation is 0.001 5 nm for the bond length and 1.753°for the bond angles.64 water molecules were added to the asymmetric unit.The refined structure showed an unusual non-prolyl cis peptide bond at residue 10.The structure was compared with group II α-toxin BmK M8 (an acidic,weak toxin).The potential structural implications of the cis peptide bond were discussed. 相似文献

The SER Primer on Ecological Restoration provides a succinct introduction to, and overview of, the rapidly growing field of ecological restoration. The Primer was issued initially in 2002 by the Society for Ecological Restoration (SER) and reissued verbatim 2 years later in a more attractive format ( http://www.ser.org/resources;resources-detail-view/ser-international-primer-on-ecological-restoration ). A SER committee recently began deliberations to update the Primer, and much discussion is underway. As two of the Primer's principal authors, we were invited to share our views on how the Primer can be advantageously revised in the light of any changes or new insights since 2002. In particular, we were asked how the Primer might be modified to reflect the ways that ecological restoration address conservation issues raised by climate change and other rapid environmental shifts and global changes. We also touch on questions relating to the benefits of ecological restoration to human society, as this is an area where the Primer needs sharper focus. We have structured the following in a ‘Frequently Asked Questions’ format to highlight issues raised in the recent literature and to focus attention on other issues that merit consideration in the Primer revision process. 相似文献

研究人参茎叶皂甙(GSL)对高胆固醇饮食大鼠心肌再灌注性心律失常(RPA_r)和脂质过氧化的影响。方法:将胆固醇乳剂用灌胃法饲养大鼠14d,建立高脂血症模型,各组大鼠进行心肌缺血再灌注实验,观察高脂血症和GSL对大鼠心肌缺血再灌注2h后血丙二醇(MDA),超氧化物歧化酶(SOD)和一氧化氮(NO)水平的影响和对再灌注性心律失常发生率的影响。结果显示:(1)用胆固醇乳剂饲养大鼠14d,成功建立高脂血症模型。同时给予GSL14d有明显降脂作用。(2)高脂血症状态下,心肌缺血再灌注2h后,血MDA升高(p<0.01),SOD降低(p<0.01)和NO(p<0.05)降低,再灌注10min内RPAr的发生率增高。(3)GSL组再灌注后2h的血MDA降低,而SOD和NO水平显著升高;使RPAr发生率大为降低,无VF发生。实验显示高脂血症加重心肌缺血再灌注损伤和提高RPAr发生率及动物死亡率,GSL可减少高脂饮食大鼠脂质过氧化和诱导体内NO生成而减轻缺血再灌注心肌损伤,降低缺血再灌注性心律失常发生率。 相似文献

Marine benthic communities living in shallow-water habitats(<100 m depth) in Antarctica possess characteristics reminiscentof Paleozoic marine communities and modern deep-sea communities.The absence of crabs and sharks, the limited diversity of teleostsand skates, the dominance of slow-moving invertebrates at highertrophic levels, and the occurrence of dense ophiuroid and crinoidpopulations indicate that skeleton-breaking predation is limitedin Antarctica today, as it was worldwide during the Paleozoicand as it is in the deep sea today. The community structureof the antarctic benthos has its evolutionary roots in the Eocene.Data from fossil assemblages at Seymour Island, Antarctic Peninsulasuggest that shallow-water communities were similar to communitiesat lower latitudes until they were affected by global cooling,which accelerated in the late Eocene to early Oligocene. Thatlong-term cooling trend ultimately resulted in the polar climateand peculiar community structure found in Antarctica today.Declining temperatures beginning late in the Eocene are associatedwith the disappearance of crabs, sharks, and most teleosts.The sudden drop in predation pressure allowed dense ophiuroidand crinoid populations to appear and flourish. These late Eoceneechinoderm populations exhibit low frequencies of sublethaldamage (regenerating arms), demonstrating that there was littleor no predation from skeleton-breaking fish and decapods. Currentscenarios of global climate change include predictions of increasedupwelling and consequent cooling in temperate and subtropicalupwelling zones.
