Heterogeneity of human C4 gene size 总被引:7,自引:0,他引:7
In this article we present a study showing that the human C4 genes differ in length because of the presence or absence of a 6.5 kb intron near the 5 end of the gene. DNA from individuals of known HLA, factor B, and C4 haplotypes was analyzed for restriction fragment length polymorphism (RFLP) by Southern blot analysis with C4-specific cDNA probes. The RFLP patterns obtained showed that the C4 genes are either 22.5 kb or 16 kb in length. They are referred to as long and short C4 genes, respectively. A population study was carried out to examine the distribution of the gene size according to C4 allotypes and haplotypes. Long C4 genes included all C4A genes studied and also some C4B allotypes, e. g., B1 on most C4 A3B1 haplotypes. Similarly, C4B null genes were found to be of the long form. Other C4B allotypes tested were found to be coded for by short C4 genes, including B2, B1 in C4 A6B1 and C4 AQOB1 (with a single C4B gene haplotype).Abbreviations used in this paper C4

The immune response that expels the tapeworm Hymenolepis citelli from the small intestine of its host the white-footed deer mouse is genetically controlled. Patent infections with this tapeworm occur only in individuals that are homozygous for a recessive allele expressed at a single gene locus. By studying this natural host-parasite system in the laboratory it was shown that host genetics contributes to parasite overdispersion in a host population in the absence of all other ecological variables. Thus, the substantive influence of the proportions of resistant and susceptible genotypes in the host population must be considered when developing parasite population models of transmission or control measures. 相似文献

The osmolality and concentrations of Na, K, Cl and the hormones arginine vasotocin (AVT), prolactin, aldosterone and corticosterone were measured in plasma as functions of time in relation to oviposition, changing NaCl content of the diet, and feeding-inanition. AVT was significantly increased immediately after oviposition (but not during the hour before) with a calculated average value of 38.0 +/- 4.1 pg/ml at oviposition. A moderate increase in concentrations of prolactin and corticosterone were observed immediately after oviposition. Oviposition was not associated with detectable changes in plasma osmolality (and electrolyte concentrations) nor with the concentration of aldosterone. After a sudden change from a high NaCl diet to a low NaCl diet the plasma osmolality and concentrations of NaCl, AVT and prolactin reached new stable levels in 24 hr, whereas the plasma aldosterone concentration required more than 4 days to reach a steady level. After resalination plasma aldosterone was suppressed in less than 8 hr. Both osmolality and concentrations of AVT and prolactin showed transient overshoots during the first 24 hr. Maintenance of the cellobiose utilization genes of Escherichia coli in a cryptic state 总被引:6,自引:1,他引:5
The genes for cellobiose utilization are normally cryptic in Escherichia
coli. The cellobiose system was used as a model to understand the process
by which silent genes are maintained in microbial populations. Previously
reported was (1) the isolation of a mutant strain that expresses the
cellobiose-utilization (Cel) genes and (2) that expression of those genes
allows utilization of three beta- glucoside sugars: cellobiose, arbutin,
and salicin. The Cel gene cluster has now been cloned from that mutant
strain. In the course of locating the Cel genes within the cloned DNA
segment, it was discovered that inactivation of the Cel-encoded hydrolase
rendered the host strain sensitive to all three beta-glucosides as potent
inhibitors. This sensitivity arises from the accumulation of the
phosphorylated beta- glucosides. Because even the fully active genes
conferred some degree of beta-glucoside sensitivity, the effects of
cellobiose on a series of five Cel+ mutants of independent origin were
investigated. Although each of those strains utilizes cellobiose as a sole
carbon and energy source, cellobiose also acts as a potent inhibitor that
reduces the growth rate on glycerol 2.5-16.5-fold. On the other hand,
wild-type strains that cannot utilize cellobiose are not inhibited. The
observation that the same compound can serve either as a nutrient or as an
inhibitor suggests that, under most conditions in which cellobiose will be
present together with other resources, there is a strong selective
advantage to having the cryptic (Cel0) allele. In those environments in
which cellobiose is the sole, or the best, resource, mutants that express
the genes (Cel+) will have a strong selective advantage. It is suggested
that temporal alternation between these two conditions is a major factor in
the maintenance of these genes in E. coli populations. This alternation of
environments and fitnesses was predicted by the model for cryptic-gene
maintenance that was previously published.

The complete mitochondrial DNA (mtDNA) of the donkey and mtDNA comparisons among four closely related mammalian species-pairs 总被引:7,自引:0,他引:7
The nucleotide sequence of the complete mitochondrial genome of the donkey, Equus asinus, was determined. The length of the molecule is 16,670 bp. The length, however, is not absolute due to pronounced heteroplasmy
caused by variable numbers of two types of repetitive motifs in the control region. The sequence of the repeats is (a) 5′-CACACCCA
and (b) 5′-TGCGCGCA, respectively. The order of (a) and (b) can be expressed as {n[2(a)+(b)]+m(a)}. In 32 different clones analyzed the number of n and m ranged from 0 to 9 and 1 to 7. The two rRNA genes, the 13 peptide-coding genes, and the 22 tRNA genes of the donkey and the
horse, Equus caballus, were compared in detail. Total nucleotide difference outside the control region was 6.9%. Nucleotide difference between peptide-coding
genes ranged from 6.4% to 9.4% with a mean of 8.0%. In the inferred protein sequences of the 13 peptide-coding genes the amino
acid difference was 0.2–8.8%, and the mean for the 13 concatenated amino acid sequences was 1.9%. In the 22 tRNA genes, the
mean difference was 3.5%, and that in the two rRNA genes was 4.1%. The mtDNA differences between the donkey and the horse
suggest that the evolutionary separation of the two species occurred ≈9 million years ago. Analyses of differences among the
mtDNAs of three other species-pairs, harbor seal/grey seal, fin whale/blue whale, and Homo/common chimpanzee, showed that the relative evolutionary rate of individual peptide-coding genes varies among different species-pairs
and modes of comparison. The findings show that the superimposition of sequence data of one lineage for resolving and dating
evolutionary divergences of other lineages should be performed with caution unless based on comprehensive data.

The complete mitochondrial DNA (mtDNA) molecules ofHomo and of the common chimpanzee were sequenced. Each sequence was established from tissue of one individual and thus nonchimeric. Both sequences were assembled in their entirety from natural (not PCR amplified) clones. Comparison with sequences in the literature identified the chimpanzee specimen asPan troglodytes verus, the West African variety of the species. The nucleotide difference between the complete human and chimpanzee sequences is 8.9%. The difference between the control regions of the two sequences is 13.9% and that between the remaining portions of the sequences 8.5%. The mean amino acid difference between the inferred products of the 13 peptide-coding genes is 4.4%. Sequences of the complete control regions, the complete 12S rRNA genes, the complete cytochromeb genes, and portions of the NADH4 and NADH5 genes of two other chimpanzee specimens showed that they were similar but strikingly different from the same regions of the completely sequenced molecule fromPan troglodytes verus. Preliminary investigations with ethanolic (EtOH) extracts from five Nigerian plants show that extracts of Piper guineense Schum and Thonn (Piperaceae), Cedrela odorata L. (Meliaceae), Dennettia tripetala G. Baker (Annonaceae) and Aframomum melegueta (Rosch) K. Schum (Zingiberaceae) in artificial diets significantly reduced larval growth of European corn borer (ECB), Ostrinia nubilalis Hubner, at a concentration of 1000 ppm (0.1%). An extract of Xylopia aethiopica (Dunal) A. Rich (Annonaceae) was ineffective. When the extracts were subsequently incorporated into artificial diets at 300 ppm and offered to neaonates, larval mortality increased in the order A. melegueta (13%), D. tripetala (13%), P. guineense (27%), and C. odorata (48%). Larval and adult emergence periods increased with increasing concentration of P. guineense, C. odorata and D. tripetala indicating a toxic response. Nutritional indices for habituated third instar larvae with the two most promising plant extracts, P. guineense and C. odorata, showed that the efficiencies of conversion of digested food (ECD) was significantly reduced at 300 ppm suggesting a postdigestive toxicity of the extracts. P. guineense and C. odorata extracts show the best potential for development as botanical insecticides. 相似文献

The complete mitochondrial DNA (mtDNA) molecule of the gorilla was
sequenced. The entire sequence, 16,412 nucleotides, was determined by
analysis of natural (not polymerase chain reaction) restriction fragments
covering the whole molecule. The sequence was established from one
individual and thus nonchimeric. After comparison with the COII gene of
gorilla specimens with known geographical origin, the sequence was
identified as characteristic of the Western lowland gorilla, Gorilla
gorilla gorilla. With the exception of the NADH2 gene, all genes have a
methionine start codon. The inferred start codon of NADH2 is ATT
(isoleucine). The COIII, NASDH4, and cytochrome b genes are not terminated
by a stop codon triplet, and the COI gene is probably terminated by an AAA
triplet rather than by a regular stop codon. The great majority of genic
sequences (rRNAs, peptide-coding genes, tRNAs) of the complete mtDNAs of
Gorilla, Pan, and Homo show a greater similarity between Pan and Homo than
between either of these genera to Gorilla. The analysis of the
peptide-coding genes suggest that relative to comparison between Homo and
Pan a certain degree of transition saturation has taken place in codon
position 3 in comparisons between Gorilla to either Homo or Pan.

The covalent attachment of ubiquitin (Ub) to short-lived or damaged proteins is believed to be the signal that initiates their selective degradation. In several cases, it has been shown that the proteolytic signal takes the form of a multi-Ub chain in which successive Ub molecules are linked tandemly at lysine 48 (K-48). Here we show that Ub molecules can be linked together in vivo at two other lysine positions, lysine 29 (K-29) and lysine 63 (K-63). The formation of these alternative linkages is strongly dependent on the presence of the stress-related Ub conjugating enzymes UBC4 and UBC5. Furthermore, expression of Ub carrying a K-63 to arginine 63 substitution in a strain of Saccharomyces cerevisiae that is missing the poly-Ub gene, UBI4, fails to compensate for the stress defects associated with these cells. Taken together, these results suggest that the formation of multi-Ub chains involving K-63 linkages plays an important role in the yeast stress response. In broader terms, these results also suggest that Ub is a versatile signal in which different Ub chain configurations are used for different functions. 相似文献

Selection-induced mutations are nonrandom mutations that occur as specific
and direct responses to environmental challenge. Examples of
selection-induced mutations have been reported both in bacteria and in
yeast. I previously showed (Hall 1988) that excisions of the mobile genetic
element IS150 from within bglF are selection induced and argued that they
occurred because they were potentially advantageous under the selective
conditions employed. Mittler and Lenski (Mittler and Lenski 1992) have
argued that such excisions are not selection induced but that they occur
randomly in nondividing cells. Here I provide further evidence that IS150
excisions are induced by selection and that the excisions are immediately,
rather than only potentially, advantageous to the cell. I also provide
evidence that excisions, which Mittler and Lenski claim occur randomly in
saturated broth cultures, actually occur after samples from those cultures
are plated onto selective medium.
