In the present study we describe a novel functional cell surface molecule, designated as Kp43, which is expressed among leukocytes by NK cells, TCR-gamma/delta + T lymphocytes, and some CD8+ CD56+TCR-alpha/beta + T cell clones. The Kp43 Ag is a 70-kDa disulfide-linked dimer, which migrates in SDS-PAGE under reducing conditions as a single 43-kDa band. Two-color immunofluorescence staining of fresh PBL revealed that only a fraction of CD16+, and of TCR-gamma/delta + T lymphocytes expressed the Ag. The analysis of TCR-alpha/beta + T cell clones showed that a small proportion (2 out of 20) weakly expressed Kp43 together with the CD8 and CD56 molecules. By immunoperoxidase staining of different tissues the anti-Kp43, reactivity was detected exclusively in lymphoid organs, where a minority of scattered cells was stained, and in some liver sinusoidal cells. Essentially all NK cells acquired Kp43 when stimulated with a B lymphoblastoid cell line. By contrast, the pattern of distribution of Kp43 remained stable upon in vitro culture of T-gamma/delta lymphocytes, thus delineating two subsets according to its expression. In lymphokine-activated killer populations, obtained by culturing either PBL or NK cells with high concentration of IL-2, most CD16+ and CD56+ cells became Kp43+. The Kp43-specific mAb inhibited the IL-2-dependent proliferative response of cultured NK and TCR-gamma/delta + T cells without affecting their non-MHC-restricted cytotoxicity. The partial inhibitory effect, which was mediated as well by pepsin digested F(ab')2 fragments, was lost upon reduction to Fab. The anti-Kp43 mAb did not interfere with the specific binding of IL-2 to its surface receptors. Altogether the data point out that the Kp43 dimer is involved in the regulation of the IL-2-dependent proliferative response of NK cells and a subset of TCR-gamma/delta + T lymphocytes. 相似文献

Balloon-occluded transarterial chemoembolisation (B-TACE) is an intraarterial transcatheter treatment for liver cancer. In B-TACE, an artery-occluding microballoon catheter occludes an artery and promotes collateral circulation for drug delivery to tumours. This paper presents a methodology for analysing the haemodynamics during B-TACE, by combining zero-dimensional and three-dimensional modelling tools. As a proof of concept, we apply the methodology to a patient-specific hepatic artery geometry and analyse two catheter locations. Results show that the blood flow redistribution can be predicted in this proof-of-concept study, suggesting that this approach could potentially be used to optimise catheter location. 相似文献

Tomato spotted wilt virus (TSWV) causes serious diseases of many economically important crops. Disease control has been achieved by breeding tomato and pepper cultivars with the resistance genes Sw‐5 and Tsw, respectively. However, TSWV isolates overcoming these genetic resistances have appeared in several countries. To evaluate the risk of spread of these resistance‐breaking isolates, we tested their ability of transmission by the main vector of TSWV, the thrips Frankliniella occidentalis. We compared the transmission rate by thrips of six TSWV isolates of different biotype (able or unable to overcome this resistance in pepper and tomato), and with divergent genotype (A and B). Our results indicate that the transmission rate was related to the amount of virus accumulated in thrips but not to virus accumulation in the source plants on which thrips acquired the virus. No correlation was found between transmission efficiency by thrips and the genotype or between transmission efficiency and the ability of overcoming both resistances. This result suggests that resistance‐breaking isolates have the same potential to be transmitted as the isolates unable to infect resistant tomato and pepper cultivars. 相似文献

A disease characterized by symptomless leaves and fruit decolouration, loss of consistency and mild deformation on ripening was detected in tomato fields in north‐eastern Spain during 2003 and 2004. DAS‐ELISA analysis revealed the presence of the Cucumber mosaic virus (CMV) in all diseased plants. CMV isolates were characterized by polyacrylamide gel electrophoresis (PAGE) analysis of double‐stranded RNAs (dsRNAs) and nucleotide sequence analysis, and compared with some CMV isolates belonging to different subgroups used as controls. A total of 12 isolates obtained from the infected tomato plants selected for this study gave the same electrophoresis pattern for the three genomic dsRNAs, which was different to the patterns showed by the CMV isolates collected in the same region a few years ago. The identity of the complete nucleotide sequence of one of these CMV isolates and the partial sequence of other five isolates compared to the Tfn strain from Italy and the BAR92/1 isolate from tomato collected in Barcelona in 1992 was higher than 99%, both belonging to subgroup IB of CMV. The CMV isolates of this subgroup found in eastern Spain in previous studies were not detected after 1996. The nucleotide sequences of two isolates that were chosen as representatives of the CMV isolates more frequently detected in previous years revealed that they belonged to the CMV subgroups IA. The origin and the possible causes of reappearance of CMV IB isolates in north‐eastern Spain are discussed. 相似文献

The avirulence determinant triggering the resistance conferred by the tomato gene Sw‐5 against Tomato spotted wilt virus (TSWV) is still unresolved. Sequence comparison showed two substitutions (C118Y and T120N) in the movement protein NSm present only in TSWV resistance‐breaking (RB) isolates. In this work, transient expression of NSm of three TSWV isolates [RB1 (T120N), RB2 (C118Y) and non‐resistance‐breaking (NRB)] in Nicotiana benthamiana expressing Sw‐5 showed a hypersensitive response (HR) only with NRB. Exchange of the movement protein of Alfalfa mosaic virus (AMV) with NSm supported cell‐to‐cell and systemic transport of the chimeric AMV RNAs into N. tabacum with or without Sw‐5, except for the constructs with NBR when Sw‐5 was expressed, although RB2 showed reduced cell‐to‐cell transport. Mutational analysis revealed that N120 was sufficient to avoid the HR, but the substitution V130I was required for systemic transport. Finally, co‐inoculation of RB and NRB AMV chimeric constructs showed different prevalence of RB or NBR depending on the presence or absence of Sw‐5. These results indicate that NSm is the avirulence determinant for Sw‐5 resistance, and mutations C118Y and T120N are responsible for resistance breakdown and have a fitness penalty in the context of the heterologous AMV system. 相似文献
