The distribution and stability of the cellular tumor antigen p53 were studied in baby rat kidney cells transformed by region E1 sequences of nononcogenic adenovirus (Ad) type 5 (Ad5) or oncogenic type 12 (Ad12). In transformed cells expressing the large E1B T antigen of Ad5, p53 was associated with this T antigen. The complexed proteins were concentrated in a cytoplasmic body, which has been shown to consist of a cluster of 8-nm filaments (A. Zantema et al., Virology 142:44-58, 1985). In transformed cells expressing the E1B region of Ad12, however, no association between the viral large T antigen and p53 was detectable. In the latter case, both proteins were found almost exclusively in the nucleus. The stability of p53 in both Ad5- and Ad12-transformed cells was increased relative to that in primary cells or cells immortalized by the E1A region only. Thus, the increased stability of p53 in Ad-transformed cells is not caused by association with a viral T antigen, but it correlates with expression of E1B and with morphological transformation.  相似文献

The consumption of phenanthrene in soil by model plant–microbial associations including natural and transconjugant plasmid-bearing rhizospheric strains of Pseudomonas fluorescens and P. aureofaciens degrading polycyclic aromatic hydrocarbons was studied. It was shown that phytoremediation of soil polluted with phenanthrene in the rhizosphere of barley (Hordeum sativum L.) was inefficient in the absence of the degrading strains. Inoculation of barley seeds with both natural and transconjugant plasmid-bearing Pseudomonas strains able to degrade polycyclic aromatic hydrocarbons (PAH) protected plants from the phytotoxic action of phenanthrene and favored its degradation in soil. Rape (Brassica napus L.) was shown to be an appropriate sentinel plant, sensitive to phenanthrene, which can be used for testing the efficiency of phenanthrene degradation in soil. Biological testing with the use of sensitive rape plants can be applied for estimation of the efficiency of phyto/bioremediation of PAH-polluted soils.  相似文献

Stereospecific binding of apomorphine to rat brain opiate receptors was shown by assaying the competition of 7,8(n)--3H--naloxone and D-ala2-tyrosyl-3,5-3H--enkephalin (5-D-leucine) for opiate receptor binding. EC-NaCl50, the concentration of apomorphine which inhibited 50% binding of the radioactive naloxone and D-ala2, D-leu5-enkephalin in the absence of NaCl were 20 and 42 microM, respectively. EC+NaCl 50, the concentration of apomorphine which inhibited 50% binding of the radioactive naloxone in the presence of 100 mM NaCl was 17 microM. From the ratio of EC+NaCl 50 to EC-NaCl the value of "sodium shift" of effective concentration can be calculated as 0.85. From the data obtained it is concluded that apomorphine, like naloxone, is a "pure" antagonist but it has much less affinity for enkephalin and opiate binding sites. The probable mechanisms of the pharmacological action of apomorphine are discussed.  相似文献

The level of catecholamines was measured in various brain parts in rats of August and Wistar lines, characterized by different stability of cardio-vascular functions in emotional stress. In norm, the August line has a higher content of dopamine (DA), in the mid-brain in particular; the Wistar line has a higher content of noradrenaline (NA) in the hypothalamus. In susceptible animals, which showed different cardio-vascular disorders during 30 hours of immobilization (48% of the Wistar line and 90% of the August), and in animals resistent against emotional stress, the changes in the initial level of catecholamines were observed, both common and specific for two lines. An assumption is made that the stability of cardio-vascular functions in the emotional stress is provided for by an intensive metabolism of the NA-synthesizing neurones in the brain isthmus, and by a moderate activity of the DA-synthesizing neurones of the mid-brain; the susceptibility to cardio-vascular disorders in the emotional stress is due to increase in the activity of the DA-synthesizing mid-brain neurones, the decrease in the activity of the DA-hydroxilase and the exhaustion of NA in the hypothalamus.  相似文献

Ribosomal frameshifting, a process whereby a translating ribosome is diverted from one reading frame to another on a contiguous mRNA, is an important regulatory mechanism in biology and an opportunity for therapeutic intervention in several human diseases. In HIV, ribosomal frameshifting controls the ratio of Gag and Gag-Pol, two polyproteins critical to the HIV life cycle. We have previously reported compounds able to selectively bind an RNA stemloop within the Gag-Pol mRNA; these compounds alter the production of Gag-Pol in a manner consistent with increased frameshifting. Importantly, they also display antiretroviral activity in human T-cells. Here, we describe new compounds with significantly reduced molecular weight, but with substantially maintained affinity and anti-HIV activity. These results suggest that development of more “ligand efficient” enhancers of ribosomal frameshifting is an achievable goal.  相似文献

Bone marrow stroma contains mesenchymal stem cells (MSC) which are progenitor cells, at least for tissues arising from mesechyma. The study of MSC biology yields controversial data. Therefore further experiments are needed to characterize these cells. The aim of our research was to compare primary cultures and subcultures of stromal precursor cells isolated from rat bone marrow. Long-term cultures of these cells isolated from 5 animals have been obtained. Morphological, immunophenotypic, and functional (capacity to osteogenic differentiation) characteristics of the cells have been investigated. We show that the cell morphology in the cultures is highly heterogenic. Morphological cell types are described. Heterogeneity of stromal cells declines on late passages. Cell cultures isolated from different animals have the same immunophenotypic markers (CD90, CD44, CD54, CD106, CD45, CD11b) but different morphological characteristics and a different capacity to osteogenic differentiation during long-term cultivation. The data show that more specific markers and functional tests should be applied to identify MSC.  相似文献

Background  

The integration of many aspects of protein/DNA structure analysis is an important requirement for software products in general area of structural bioinformatics. In fact, there are too few software packages on the internet which can be described as successful in this respect. We might say that what is still missing is publicly available, web based software for interactive analysis of the sequence/structure/function of proteins and their complexes with DNA and ligands. Some of existing software packages do have certain level of integration and do offer analysis of several structure related parameters, however not to the extent generally demanded by a user.  相似文献

H2S+ bacteria responsible for the degradation of sulfur-containing amino acids of fish muscle are currently little used to evaluate the microbiological pal quality of fish. Shewanella putrefaciens greatly predominates in this flora, and was therefore used to define a suitable culture method and medium. Inoculations by the Spiral surface method at 25C, with an incubation of 72h, gave the best counts on a medium containing two sources of sulfur (organic and inorganic) for H2S+ bacteria. The culture medium and the NaCl concentration were determinant in the evaluation of this flora. At present there is no standard medium which meets these requirements.  相似文献

Combination of genetic systems of degradation of polyaromatic hydrocarbons, resistance to heavy metals, and promotion of plant growth/protection is one of the approaches to the creation of polyfunctional strains for phytoremediation of soils after co-contamination with organic pollutants and heavy metals. A plant-growth-promoting rhizosphere strain Pseudomonas chlororaphis PCL1391 (pBS216*, pBS501) has been obtained, in which the nah operon of plasmid pBS216 provides naphthalene biodegradation and the cnr-like operon of plasmid pBS501 provides resistance to cobalt and nickel due to the extrusion of heavy metal cations from the cells. In the presence of 100 μM of nickel, the viability, growth rate, and naphthalene biodegradation efficiency of the resistant strain PCL1391 (pBS216*, pBS501) were much higher as compared with the sensitive PCL1391 (pBS216). During the growth of the resistant strain, in contrast to the sensitive strain, nickel (100 μM) had no inhibiting effect on the activity of the key enzymes of naphthalene biodegradation. Heterogeneity of stromal precursor cells isolated from rat bone marrow   总被引:5,自引:0,他引:5  
Bone marrow stroma contains mesenchymal stem cells (MSC) which are precursor for at least mesenchyma-derived cells. Recent investigations revealed a lot of questions concerning MSC biology that should be further refined. The aim of this study was the comparative analysis of rat bone marrow stroma cells cultures. Mesenchymal precursor cells isolated from rat bone marrow were passed up to 50 times. Comparative morphological and immunophenotypical analysis of these cultures was carried out as well as their ability to osteogenic differentiation was studied. The isolated cultures contained morphologically different types of cells and thus showed a high heterogenity level. Morphology of these cell types was described. The heterogeneity level was reported to decrease over time. It was found out that subcultures isolated from different rats shared the same immunophenotype characteristics (CD90+, CD44+, CD54+, CD 106+, CD45-, CD11b-), but differed in their morphology as well as in ability to osteogenic differentiation. Thus MSC identification requires more specific marker and functional tests to be used.  相似文献
