Summary A series of phosphopeptides Tyr(PO3H2)-Val-Pro-Xxx-Leu (Xxx=Met, Met(O), Nle, Dab or Cys), derived from the native platelet-derived growth factor- receptor (PDGF-) sequence, has been prepared to study their interaction with the src-homology 2 (SH2) domains of the p85 subunit of PI3 kinase. The phosphopeptides were synthesized using Fmoc methodology incorporating N-Boc dibenzyl-protected phosphotyrosine (Boc-Tyr[PO3(Bzl)2]) as the N-terminal amino acid, since the benzyl groups can be removed during resin cleavage with TFA. Only peptides containing methionine were found to exist partially as S-benzyl sulfonium salts after TFA cleavage from the resin. The desired peptide could be obtained from the S-benzyl sulfonium salt by hydrogenolysis. 相似文献

Nocturnal lorises and pottos (Lorisinae) are among the least gregarious of primates. Mothers start to leave their infants alone during the night as early as the day of birth. However, captive studies also indicate that weaning young lorisines closely follow their mothers nearly all the time and obtain their first solid food via scrounging. Accordingly, it has been suggested that young lorisines depend on their mothers to obtain dietary information and to achieve dietary independence by watching their mothers feeding or interacting directly with their mothers over food. We tested for a social dependence on dietary learning by infants in a social network of wild slow lorises (Nycticebus coucang). The social network included one male infant, his mother, and two subadult females. The infant only took to mouth food items that were also part of the females' diet and showed concordance in the frequency of use of food patches with the females. These results contradict dietary learning by trial and error. They indicate that dietary learning by infants depends on information obtained from older conspecifics. However, the infant was never involved in direct interactions with conspecifics over food and fed mostly alone. He was not within a distance where he could see the females feeding more often than expected from the configuration and utilization of home ranges. The infant never looked at conspecifics feeding in his vicinity, which suggests that visual observation or direct interaction over food may not be the mechanisms by which information about food resources is passed from older individuals to young, but that other ways of obtaining such information are used. 相似文献

ERK-2 MAP kinase activation induces inhibitory effects on nuclear protein import in vascular smooth muscle cells. The mechanism and characteristics of this effect of ERK-2 were investigated. An unusual dose-dependent effect of ERK-2 on nuclear protein import was identified. At higher concentrations (1 microg/mL) of ERK-2, nuclear protein import was stimulated, whereas lower concentrations (0.04 microg/mL) inhibited import. Intermediate concentrations exerted intermediate effects. The stimulatory and inhibitory effects at the 2 different ERK-2 concentrations were observed in both conventional, permeabilized cell assays of nuclear protein import and with in situ microinjection of smooth muscle cells. The biphasic effects of ERK-2 on import were also found for the other 2 members of the MAPK family, p38 and JNK. RanGAP was identified by structural analysis as a candidate target protein responsible for mediating the effects of ERK-2. After pretreatment with high concentrations of ERK-2, RanGAP activity was significantly increased by approximately 50%. In contrast, low concentrations of ERK-2 significantly attenuated RanGAP activity. These results demonstrate that all 3 members of the MAPK family can alter nuclear protein import in opposite directions depending upon the concentration of ERK-2 used. RanGAP represents the MAP kinase target whereby nuclear transport can be stimulated or inhibited. 相似文献

Abstract Prays oleae Bern, (OM) and Palpita unionalis Hüb., (JM) are two of the most important pests in olive groves in Egypt. A 3‐year monitoring study using sex pheromone traps in semi‐arid and arid olive groves was performed. In the semi‐arid grove, flight pattern of the OM was the same as in other Mediterranean countries, but in dates concordating plant phenology. The moth completes three generations annually: the first flight is in March to April, the second is in May to June and the third occurs in August to October. In the arid olive grove, an interesting flight pattern was observed. First flight was always very close or overlapped with the second one with no male catches during August to October. However, eggs were present most of the season, indicating unusually high female presence and oviposition activity of the OM during the absence of males in the traps. Generally, moth densities were significantly lower in low fruiting years than in higher ones and were also lower in the arid olive grove than in the semi‐arid one. In contrast, JM males were present all season, exhibiting six to seven and three to four overlapping flight peaks in arid and semi‐arid olive groves, respectively. Moth densities were significantly higher in the arid olive grove than those in the semi‐arid one. This study shows that trapping location and fruit bearing year are characteristics that strongly affect the grove‐specific information needed to estimate correctly adult emergence and thus the timing of control measures. 相似文献

Residual microorganisms and/or re-infections are a major cause for root canal therapy failure. Understanding of the bacterial content could improve treatment protocols. Fifty samples from 25 symptomatic and 25 asymptomatic previously root-filled teeth were collected from Sudanese patients with periradicular lesions. Amplified 16S rRNA gene (V1-V2) variable regions were subjected to pyrosequencing (FLX 454) to determine the bacterial profile. Obtained quality-controlled sequences from forty samples were classified into 741 operational taxonomic units (OTUs) at 3% dissimilarity, 525 at 5% dissimilarity and 297 at 10% dissimilarity, approximately corresponding to species-, genus- and class levels. The most abundant phyla were: Firmicutes (29.9%), Proteobacteria (26.1%), Actinobacteria (22.72%), Bacteroidetes (13.31%) and Fusobacteria (4.55%). Symptomatic patients had more Firmicutes and Fusobacteria than asymptomatic patients, while asymptomatic patients showed more Proteobacteria and Actinobacteria. Interaction of disease status and age was observed by two-way ANOSIM. Canonical correspondence analysis for age, tooth restoration and disease status showed a correlation of disease status with the composition and prevalence of different members of the microbial community. The pyrosequencing analysis revealed a distinctly higher diversity of the microbiota compared to earlier reports. The comparison of symptomatic and asymptomatic patients showed a clear association of the composition of the bacterial community with the presence and absence of symptoms in conjunction with the patients’ age. 相似文献

Cel7A (previously known as cellobiohydrolase I) from Hypocrea jecorina was crystallized in two crystalline forms, neither of which have been previously reported. Both forms co‐crystallize under the same crystallization conditions. The first crystal form belonged to space group C2, with unit‐cell parameters a = 152.5, b = 44.9, c = 57.6 Å, β = 101.2°, and diffracted X‐rays to 1.5 Å resolution. The second crystal form belonged to space group P6322, with unit‐cell parameters a = b≃ 155, c≃ 138 Å, and diffracted X‐rays to 2.5 Å resolution. The crystals were obtained using full‐length Cel7A, which consists of a large 434‐residue N‐terminal catalytic domain capable of cleaving cellulose, a 27‐residue flexible linker and a small 36‐residue C‐terminal carbohydrate‐binding module (CBM). However, a preliminary analysis of the electron‐density maps suggests that the linker and CBM are disordered in both crystal forms. Although numerous epidemiologic studies have examined the long-term safety of silicone breast implants during the past decade, there is a relative lack of surveillance data on short-term health effects and complications following cosmetic surgery of the breast. The Danish Registry for Plastic Surgery of the Breast, established in May of 1999, provides plastic surgeons with a nationwide system for the collection of preoperative, perioperative, and postoperative data on women undergoing breast implantation, breast reduction, or mastopexy. The purpose of the Registry is to examine short-term and, eventually, long-term local complications and possible health effects, and to contribute to an ongoing evaluation of surgical results and surveillance of the products. Furthermore, the Registry will allow the identification of new areas for research into cosmetic and reconstructive breast surgery. Women accepting registration in the Danish Registry for Plastic Surgery of the Breast complete a self-administered questionnaire focusing on medical history and demographic and behavioral factors. Preoperative blood samples are drawn for storage. Surgical data, postoperative results, and complications are registered following surgery and at postoperative visits. Currently, registration has been initiated at 24 private and public clinics, representing more than 80 percent of the plastic surgery clinics in Denmark. As of November of 2001, a total of 1472 women with breast implants and 560 women with breast reduction were included in the Registry. These figures are expected to increase annually by 1000 women undergoing breast implantation and 500 women undergoing breast reduction or mastopexy. The authors present their experience of establishing the first nationwide comprehensive clinical-epidemiologic database and biological bank for cosmetic and reconstructive surgery procedures. 相似文献

The calC2 mutation in Aspergillus nidulans causes hypersensitivity to Calcofluor White, along with other drug sensitivities that indicate a defect in cell wall integrity. We have cloned CalC by complementation, isolating the A. nidulans orthologue of protein kinase C (PkcA). The pkcA allele of the calC2 strain contains a mutation predicted to introduce a charged arginine residue in place of neutral glycine at a conserved site located immediately beside the C1B regulatory domain. Both PkcA and calC2 map to the same region of chromosome VIII. A PkcA::GFP chimera localizes to hyphal apices and growing septa, as well as to the conidiogenous apices of phialides, indicating a role for PkcA in polarized cell wall growth. These observations support the hypothesis that the role of PkcA in A. nidulans, is comparable to that played by Pkc1p in the Saccharomyces cerevisiae cell wall integrity pathway. 相似文献

Aim For a large set of forest herbs we tested: (1) whether there is a positive relationship between local abundance and geographical range size; (2) whether abundance or range size are affected by the niche breadths of species or niche availability; and (3) whether these are affected by the species life‐history traits. Location Northwestern Germany. Methods We measured abundance as mean density in 22 base‐rich deciduous forests and recorded range size as area of occupancy on four different spatial scales (local to national). Niche breadth was expressed in terms of habitat specificity (specialists, generalists) and of the ability to grow across a broad range of soil pH. The species’ pH niche position was used as a measure of the importance of habitat availability. As life‐history traits we used diaspore mass and number, plant height, seed longevity, lifespan/clonality, pollination mode, dispersal capability and flowering time. Results There were mainly no positive relationships between the abundance of species and their range size, as tested across species and across phylogenetically independent contrasts. Forest specialists were generally distributed less widely than generalists, but habitat specificity was not related to local abundance. Species with a broader pH niche breadth were more common, but the positive relationships between niche breadth and abundance or range size disappeared when accounting for sample size effects. Clonal species with few and heavy diaspores were most abundant, as well as early‐flowering species and those lacking dispersal structures. Local and regional range size were determined largely by habitat availability, while national range was positively affected by plant height and diaspore mass. Main conclusions Different processes determine the local density of species and their range size. Abundance within habitat patches appears to be related mainly to the species life histories, especially to their capacity for extensive clonal reproduction, whereas range size appears to be determined strongly by the availability of suitable habitat. 相似文献
