In order to know the morphostructural changes that occurs in the enamel during the mineralization and to study the rod tridimensional orientation, the author has examined the literature concerning these subjects comparing it to the results of his researches realized by S.E.M. and Rx-microprobe-analysis. It has been demonstrated that three phases can be distinguished during the mineralization: "fibrillation phase", "rod density phase", "interrod density phase". The fibrillation phase begins in the middle of the ameloblastic secretion phase, ends 8 mm from the cervical ansa and is characterized by the unmasking of the matrix subunits and chemically by the disappearance of the sulphur. The rod density phase occurs in the area included between 8 and 14 mm from the cervical ansa, that is in the ameloblastic modulation phase, and is characterized by the aggregation of the intraprismatic subunits and by the calcium increase. The interrod density phase that occurs during the ameloblastic pigmentation phase, is characterized by the aggregation of the interprismatic subunits and by the presence of the iron in the most superficial enamel layers. The rod orientation in the space has been studied during the second phase of the mineralization suice the intraprismatic subunits join while the interprismatic ones remain in the fibrillation phase; besides the author has compared the results of these studies to the morphology of the external opening of the honeycomb pits. In such a way it has been demonstrated that the rod originates from the dentine-enamel junction forming a 60 degree angle that opens in the cuspidalmesial or cuspidal-lateral direction. In the outer enamel layer the rod loses its mesial or lateral inclination and bends further towards the cusp delimiting a 30 degree angle; finally it bends outward and as a sharpend that is ortogonal to the enamel external surface. It is eventually discussed how this model of rod reconstruction is fit to convert the tangential forces that develop during the mastication into compression forces.  相似文献

Aim of our present work was to investigate a new method to study the three-dimensional arrangement, the length and the diameter of the different parts of the renal tubules. The ureter was cannulated after blocking the urinary flow with a binding of the ureter itself at its intermediate third, and injected in it against flow a synthetic resin (Mercox) normally used for vascular corrosion casts. It was demonstrated that the binding maintained only for 24 hours is adequate for morphological studies of the urinary tracts from papillar ducts until the Henle's loop. On the contrary the binding maintained for 7 days induced marked changes in the tubular architecture similar to the first anatomo-pathological changes of the nephrosclerosis following a chronic obstructive nephropathy.  相似文献

Sarcoglycans are transmembrane proteins that seem to be functionally and pathologically as important as dystrophin. Sarcoglycans cluster together to form a complex, which is localized in the cell membrane of skeletal, cardiac, and smooth muscle. It has been proposed that the dystrophin-glycoprotein complex (DGC) links the actin cytoskeleton with the extracellular matrix and the proper maintenance of this connection is thought to be crucial to the mechanical stability of the sarcolemma. The integrins are a family of heterodimeric cell surface receptors which play a crucial role in cell adhesion including cell-matrix and intracellular interactions and therefore are involved in various biological phenomena, including cell migration, and differentiation tissue repair. Sarcoglycans and integrins play a mechanical and signaling role stabilizing the systems during cycles of contraction and relaxation. Several studies suggested the possibility that integrins might play a role in muscle agrin signalling. On these basis, we performed an immunohistochemical analyzing sarcoglycans, integrins and agrin, on human skeletal muscle affected by sensitive-motor polyneuropathy, in order to better define the correlation between these proteins and neurogenic atrophy due to peripheral neuropathy. Our results showed the existence of a cascade mechanism which provoke a loss of regulatory effects of muscle activity on costameres, due to loss of muscle and neural agrin. This cascade mechanism could determine a quantitative modification of transmembrane receptors and loss of alpha7B could be replaced and reinforced by enhanced expression of the alpha7A integrin to restore muscle fiber viability. Second, it is possible that the reduced cycles of contraction and relaxation of muscle fibers, during muscular atrophy, provoke a loss of mechanical stresses transmitted over cell surface receptors that physically couple the cytoskeleton to extracellular matrix. Consequently, these mechanical changes could determine modifications of chemical signals through variations of pathway structural integrins, and alpha7A could replace alpha7B.  相似文献

Several studies demonstrated that transglutaminases play a key role in extracellular matrix stabilization needed for cell differentiation. We evaluated transglutaminase expression and activity in the pre-secretory stage of differentiation of the continuously erupting rat incisor. We observed that transglutaminase-mediated incorporation of monodansylcadaverine into protein substrates was specifically located in the apical loop, and along the basement membrane joining mesenchyme and inner dental epithelium in the odontogenic organ. Enzyme activity was associated with mRNAs for transglutaminase 1 and 2. Notably, labelling cells for these isoenzymes were observed in both mesenchymal and epithelial compartments, but not in the basement membrane, in the ameloblast facing pulp anterior region, where ameloblast and odontoblast differentiation begins. These findings demonstrate that transglutaminase 1 and transglutaminase 2 are expressed at a major extent in the pre-secretory stage of regenerating rat incisor, where they probably play complementary roles in cell signalling between mesenchyme and epithelium and extracellular matrix.  相似文献

Aims:  The aim of the work was to evaluate the circulation of the viruses and to determine a correlation between faecal indicators and viruses.
Methods and Results:  Raw wastewater and effluent samples were collected from three wastewater treatment plants, during three sampling periods, and analysed, using cultural and molecular methods, to determine bacteria and virus presence. The results show a removal of bacterial indicators, but a limited reduction of the phages. The viral analysis displays the circulation of cultivable enteroviruses and differences in the seasonal-geographical distribution. Hepatitis A virus was found with only two genotypes: IA-IB. Rotavirus was present in 11·11%, 24·14%, 2·78% of the samples in the 1st, 2nd and 3rd sampling periods; Astrovirus in 33·33%, 6·9%, 25%; Adenovirus in 7·41%, 3·45%, 2·78%; Norovirus in 7·41%, 10·34%, 5·56% respectively. Adenovirus was never identified in plants B and C as Rotavirus in plant C.
Conclusions:  The presence of faecal indicators was not predictive of the enteric virus presence, whereas a different circulation of Enteroviruses was found in the wastewater treatment plants.
Significance and Impact of the Study:  The study shows the importance and the usefulness of molecular methods to evaluate the virus circulation and the genetic variability of Enteroviruses.  相似文献
