Three protease-containing fractions were recovered by gel filtration from concentrated crude extracellular products produced by Aeromonas hydrophila grown in a defined medium. The recovery of a heat-stable protease was differentially prevented when the crude preparation was stored for 48 h at -20 degrees C but was unaffected by storage of the crude preparation at either 4 or -70 degrees C. Once fractionated, the heat-stable protease appeared to be unaffected by subsequent storage at 4, -20, or -70 degrees C.  相似文献

The ability of two strains of Lactobacillus acidophilus, CRL 640 and CRL 800, to survive and retain their biological activities under frozen storage was determined. Freezing and thawing, as well as frozen storage, damaged the cell membrane, rendering the microorganisms sensitive to sodium chloride and bile salts. Both lactic acid production and proteolytic activity were depressed after 21 days at -20 degreesC, whereas beta-galactosidase activity per cell unit was increased. Incorporation of the amino acid selenocysteine into a growing protein chain involves the interaction between a hairpin in the mRNA termed the selenocysteine insertion sequence (SECIS) and the special elongation factor SelB. Here we present the structure of the SECIS from the thermophilic organism Moorella thermoacetica (SECIS-MT) determined using nuclear magnetic resonance (NMR) spectroscopy. The SECIS-MT hairpin structure contains a pentaloop with the first and fourth nucleotides of the loop forming a noncanonical GC base pair; the fifth loop nucleotide is bulged out and unstructured. The G and U in positions two and three are on opposite sides of the loop and solvent exposed. The backbone resonances of the SECIS-binding domain from the M. thermoacetica SelB protein were assigned, and the degree of chemical shift perturbations that occur upon SECIS binding were mapped onto the structure of the complex. We demonstrate that a region in the third winged-helix domain of SelB, not previously implicated in binding, is affected by SECIS binding.  相似文献

Summary Organ cultures containing the sex pheromone gland ofDiatraea saccharalis (F.) (sugarcane borer) were maintained for over 2 months. Juvenile hormone but not ecdysterone appeared to be essential for the maintenance of integrity of the sex pheromone gland in the organ culture system. Muscles associated with these cultures pulsated during hte entire culture period with or without juvenile hormone, suggesting that the hormone requirement was specific for the sex pheromone gland and not a general requirement of the cells of the organ-cultured segment.  相似文献

An ultrastructural study of the sex pheromone gland of female adult sugarcane borers suggests that pheromone is not produced during the first 2 hr after emergence but reaches a peak 48 hr after emergence. The sex pheromone gland is composed of two cell types and a number of structural changes observed to occur in the cells of the sex pheromone gland prior to pheromone production are described.  相似文献

One hundred and twenty-six beef bulls on southwestern Louisiana coastal range were evaluated for breeding soundness. Samples were taken to determine the incidence of bovine leukemia virus (BLV) infection, and the prepuce was cultured for potential pathogens. A high incidence (47.6%) of questionable and unsatisfactory potential breeders resulted mainly from 37.0% of the bulls exhibiting high numbers of abnormal sperm cells in the semen. Only bulls in the 4-to 5-yr age group exhibited the expected incidence of normal spermiograms. Genital campylobacteriosis was not diagnosed but there was genital trichomoniasis in three of the seven herds. Hemophilus somnus , mycoplasma and ureaplasma were isolated from the prepuce of 13.3, 48.8 and 36.7% of the bulls, respectively. Isolation of these organisms from the prepuce did not appear to be associated with abnormal spermiograms. Of the bulls studied, 34.4% had positive AGID reactions for BLV. Bulls seropositive to BLV had an increased incidence of leukocyte counts that were above the normal range. There was no apparent relationship between BLV infection and abnormal spermiograms.  相似文献
