Flagellar-mediated motility is an indispensable function for cell types as evolutionarily distant as mammalian sperm and kinetoplastid parasites, a large group of flagellated protozoa that includes several important human pathogens. Despite the obvious importance of flagellar motility, little is known about the signalling processes that direct the frequency and wave shape of the flagellar beat, or those that provide the motile cell with the necessary environmental cues that enable it to aim its movement. Similarly, the energetics of the flagellar beat and the problem of a sufficient ATP supply along the entire length of the beating flagellum remain to be explored. Recent proteome projects studying the flagella of mammalian sperm and kinetoplastid parasites have provided important information and have indicated a surprising degree of similarities between the flagella of these two cell types.  相似文献

Caulerpenyne, the major secondary metabolite synthesized by the green marine alga Caulerpa taxifolia, is cytotoxic against several cell lines. To identify possible targets of this toxin, we investigated the effect of caulerpenyne on the neuroblastoma SK-N-SH cell line. Caulerpenyne induced an inhibition of SK-N-SH cell proliferation with an IC50 of 10 +/- 2 microM after 2 hr of incubation.We observed no blockage in G2/M phase and an increase in cell death. On immunofluorescence microscopy, caulerpenyne affected the microtubule network in SK-N-SH cell line; we observed a loss of neurites and a compaction of the microtubule network at the cell periphery. In vitro, after 35 min of incubation, caulerpenyne inhibited the polymerization of pig brain purified tubulin or microtubule proteins, with an IC50 of 21 +/- 2 microM and 51 +/- 6 microM respectively. Analysis by electron microscopy indicated that caulerpenyne induced aggregation of tubulin, which may be responsible for inhibition of microtubule polymerization and bundling of residual microtubules.  相似文献

DNA profiling is a well-established technology for use in the criminal justice system, both in courtrooms and elsewhere. The fact that DNA profiles are based on non-coding DNA and do not reveal details about the physical appearance of an individual has contributed to the acceptability of this type of evidence. Its success in criminal investigation, combined with major innovations in the field of genetics, have contributed to a change of role for this type of evidence. Nowadays DNA evidence is not merely about identification, where trace evidence is compared to a sample taken from a suspect. An ever-growing role is anticipated for DNA profiling as an investigative tool, a technique aimed at generating a suspect where there is none. One of these applications is the inference of visible traits. As this article will show, racial classifications are at the heart of this application. The Netherlands and its legal regulation of 'externally visible traits' will serve as an example. It will be shown that, to make this technology work, a large number of actors has to be enrolled and their articulations invited. This indicates that instead of a 'silent witness', a DNA profile should rather be seen as an 'articulate collective'. Based on two cases, I argue that the normativity of visible traits is context-dependent. Taking into account the practices in which technology is put to use alerts us to novel ethical questions raised by their application.  相似文献

Shallow basal root growth angle (BRGA) increases phosphorus acquisition efficiency by enhancing topsoil foraging because in most soils, phosphorus is concentrated in the topsoil. Root hair length and density (RHL/D) increase phosphorus acquisition by expanding the soil volume subject to phosphorus depletion through diffusion. We hypothesized that shallow BRGA and large RHL/D are synergetic for phosphorus acquisition, meaning that their combined effect is greater than the sum of their individual effects. To evaluate this hypothesis, phosphorus acquisition in the field in Mozambique was compared among recombinant inbred lines of common bean (Phaseolus vulgaris) having four distinct root phenotypes: long root hairs and shallow basal roots, long root hairs and deep basal roots, short root hairs and shallow basal roots, and short root hairs and deep basal roots. The results revealed substantial synergism between BRGA and RHL/D. Compared with short-haired, deep-rooted phenotypes, long root hairs increased shoot biomass under phosphorus stress by 89%, while shallow roots increased shoot biomass by 58%. Genotypes with both long root hairs and shallow roots had 298% greater biomass accumulation than short-haired, deep-rooted phenotypes. Therefore, the utility of shallow basal roots and long root hairs for phosphorus acquisition in combination is twice as large as their additive effects. We conclude that the anatomical phene of long, dense root hairs and the architectural phene of shallower basal root growth are synergetic for phosphorus acquisition. Phene synergism may be common in plant biology and can have substantial importance for plant fitness, as shown here.Suboptimal phosphorus availability is a primary limitation to plant growth in terrestrial ecosystems (Vance et al., 2003). Large areas of tropical and subtropical soils in Africa, Latin America, and Asia have phosphorus availability limited by low total phosphorus content as well as high phosphorus fixation (Sanchez and Uehara, 1980). The use of phosphorus fertilizer to correct phosphorus deficiency is only a partial solution, since phosphorus fertilizers are costly, nonrenewable, potentially harmful to the environment, and often marginally effective in tropical soils because of immobilization by the soil (Cathcart, 1980). Therefore, the development of crop cultivars with enhanced ability to acquire phosphorus is an important strategy to increase agricultural productivity in low-input agroecosystems and to reduce input requirements in intensive agriculture (Vance et al., 2003; Gahoonia and Nielsen, 2004; Lambers et al., 2006; Lynch, 2007, 2011).Several root phenes (i.e. basic units of the phenotype; Serebrovsky, 1925; Lynch, 2011; for discussion, see York et al., 2013) enhance phosphorus acquisition, including root architectural phenes for topsoil foraging (Lynch and Brown, 2001), such as shallow root growth angles (Liao et al., 2004; Ho et al., 2005), increased basal root whorl number (Lynch and Brown, 2012; Miguel et al., 2013), and adventitious rooting (Miller et al., 2003); phenes to enhance soil exploitation, including root hair length and density (RHL/D; Bates and Lynch, 2000a, 2000b, 2001; Ma et al., 2001a; Gahoonia and Nielsen, 2004; Yan et al., 2004) and phosphorus-solubilizing root exudates (Ryan et al., 2001); mycorrhizal symbioses (Smith and Read, 2008); and phenes that reduce the metabolic cost of soil exploration (Lynch and Ho, 2005), such as root etiolation and root cortical aerenchyma (Fan et al., 2003; Postma and Lynch, 2010, 2011). It is probable that interactions among these phenes are important in determining the phosphorus acquisition of integrated phenotypes. Results from the structural-functional model SimRoot indicate that RHL/D, the distance from the root tip to the first appearance of root hairs, and the pattern of root hair-bearing epidermal cells (trichoblasts) among non-hair-bearing cells (atrichoblasts) are synergetic for phosphorus acquisition in Arabidopsis (Arabidopsis thaliana; Ma et al., 2001b). Another SimRoot study showed that on low-phosphorus soils, the utility of root cortical aerenchyma in maize (Zea mays) may be 2.9 times greater in plants with increased lateral branching density than in plants with normal branching (Postma and Lynch, 2011). Morphological, anatomical, symbiotic, and biochemical phenes expressed by root axes should have significant synergies with architectural phenes, since architectural phenes determine the position of root axes in time and space and, therefore, the soil domain in which spatially localized phenes are expressed (Lynch, 2011).Phosphorus availability is greater in the topsoil, with a steep decline with depth. Therefore, root architectural phenes that increase topsoil foraging can improve phosphorus acquisition (Lynch and Brown, 2001). Root shallowness regulated by basal root growth angle (BRGA) has been demonstrated to be of particular importance for topsoil foraging (Bonser et al., 1996; Liao et al., 2001; Rubio et al., 2001; Ho et al., 2005). These studies show that common bean (Phaseolus vulgaris) genotypes with smaller BRGA (i.e. shallower roots) have better performance in low-phosphorus soils. Shallow root distribution is also important for phosphorus acquisition in maize (Zhu et al., 2005).RHL/D are also important for phosphorus acquisition (Bates and Lynch, 2000a, 2000b, 2001; Gahoonia and Nielsen, 2004). Since phosphorus mobility in soil is governed by diffusion rather than mass flow, phosphorus uptake by roots is limited by localized phosphorus depletion in the rhizosphere (Barber, 1995). Long root hairs extend the phosphorus depletion zone surrounding the root, thereby increasing the total amount of phosphorus accessible by the roots and phosphorus acquisition. In many plant species, the length and density of root hairs increase in response to low phosphorus availability (Bates and Lynch, 1996; Ma et al., 2001a). Increased RHL/D increases phosphorus accumulation in Arabidopsis growing in low-phosphorus conditions (Bates and Lynch, 2000a, 2000b), and mutants lacking root hairs have reduced phosphorus acquisition (Bates and Lynch, 2000b; Gahoonia et al., 2001). Species that develop more and/or longer root hairs (e.g. Lolium perenne) are more efficient in accessing inorganic phosphorus from soils and thus show greater growth response to phosphorus fertilization than species that lack these traits (e.g. Podocarpus totara). Genotypic variation for root hairs is associated with increased phosphorus acquisition in several species, including barley (Hordeum vulgare; Gahoonia and Nielsen, 2004), common bean (Miguel, 2004; Yan et al., 2004), and maize (Zhu et al., 2010).We hypothesize that the utilities of BRGA and RHL/D for phosphorus acquisition are synergetic. Root hairs will be more valuable for phosphorus acquisition if located in surface soil horizons by arising from roots with a shallow growth angle; shallow roots will have greater benefit for phosphorus acquisition if they have long and dense hairs. Therefore, genotypes possessing long, dense root hairs on shallow roots should have greater phosphorus acquisition than genotypes with either long root hairs on deep roots or short root hairs on shallow roots. We expect the combined benefit of long root hairs and shallow root growth angles to exceed the sum of their individual effects, since they permit greater exploitation of soil strata with the greatest phosphorus availability.In this study, we evaluated the potential synergism between the architectural phene of BRGA and the morphological phene of RHL/D for phosphorus acquisition by comparison of contrasting phenotypes of common bean growing in a weathered tropical soil.  相似文献

Glioblastoma multiforme (GBM) is the most common malignant adult brain tumor. Standard GBM treatment includes maximal safe surgical resection with combination radiotherapy and adjuvant temozolomide (TMZ) chemotherapy. Alarmingly, patient survival at five-years is below 10%. This is in part due to the invasive behavior of the tumor and the resulting inability to resect greater than 98% of some tumors. In fact, recurrence after such treatment may be inevitable, even in cases where gross total resection is achieved. The Cancer Genome Atlas (TCGA) research network performed whole genome sequencing of GBM tumors and found that GBM recurrence is linked to epigenetic mechanisms and pathways. Central to these pathways are epigenetic enzymes, which have recently emerged as possible new drug targets for multiple cancers, including GBM. Here we review GBM treatment, and provide a systems approach to identifying epigenetic drivers of GBM tumor progression based on temporal modeling of putative GBM cells of origin. We also discuss advances in defining epigenetic mechanisms controlling GBM initiation and recurrence and the drug discovery considerations associated with targeting epigenetic enzymes for GBM treatment.  相似文献

Six new steroidal saponins, pandarosides K–M (13) and their methyl esters (46), were isolated as minor components, after a careful chemical reinvestigation of the Caribbean sponge Pandaros acanthifolium. Their structures were established on the basis of spectroscopic analyses and comparison with the data obtained from previous metabolites of this family. All new compounds showed moderate to weak activity against four parasitic protozoa. Additionally, these compounds and previously reported pandarosides and acanthifoliosides were tested on three human tumour cell lines, and their haemolytic and liposome permeabilizing activity were assessed. Two pandarosides exhibited moderate to strong cytotoxic effect, while three acanthifoliosides showed strong haemolytic activity.  相似文献

The chemical composition of the Caribbean sponge Pandaros acanthifolium was investigated and led to the isolation of seven new steroidal glycosides namely pandarosides A-D (1, 3, 4 and 6) along with the three methyl esters of pandarosides A, C, and D (2, 5 and 7). Their structures were characterized as 3β-[β-glucopyranosyl-(1→2)-β-glucopyranosyloxyuronic acid]-16-hydroxy-5α,14β-poriferast-16-ene-15,23-dione (1) and its methyl ester (2), 3β-[β-glucopyranosyloxyuronic acid]-16-hydroxy-5α,14β-poriferast-16-ene-15,23-dione (3), 3β-[β-glucopyranosyl-(1→2)-β-glucopyranosyloxyuronic acid]-16-hydroxy-5α,14β-cholest-16-ene-15,23-dione (4) and its methyl ester (5), 3β-(β-glucopyranosyloxyuronic acid)-16-hydroxy-5α,14β-cholest-16-ene-15,23-dione (6) and its methyl ester (7) on the basis of detailed spectroscopic analyses, including 2D NMR and HRESIMS studies. Pandarosides A-D and their methyl esters (1-7) are all characterized by a rare 2-hydroxycyclopentenone D-ring with a 14β configuration. The absolute configuration of the aglycon part of pandaroside A (1) was assigned by comparison between experimental and TDDFT calculated circular dichroism spectra on the more stable conformer.  相似文献

Background

Identifying and quantifying pathological changes in brain electrical activity is important for investigations of brain injury and neurological disease. An example is the development of epilepsy, a secondary consequence of traumatic brain injury. While certain epileptiform events can be identified visually from electroencephalographic (EEG) or electrocorticographic (ECoG) records, quantification of these pathological events has proved to be more difficult. In this study we developed MATLAB-based software that would assist detection of pathological brain electrical activity following traumatic brain injury (TBI) and present our MATLAB code used for the analysis of the ECoG.

Methods

Software was developed using MATLAB(?) and features of the open access EEGLAB. EEGgui is a graphical user interface in the MATLAB programming platform that allows scientists who are not proficient in computer programming to perform a number of elaborate analyses on ECoG signals. The different analyses include Power Spectral Density (PSD), Short Time Fourier analysis and Spectral Entropy (SE). ECoG records used for demonstration of this software were derived from rats that had undergone traumatic brain injury one year earlier.

Results

The software provided in this report provides a graphical user interface for displaying ECoG activity and calculating normalized power density using fast fourier transform of the major brain wave frequencies (Delta, Theta, Alpha, Beta1, Beta2 and Gamma). The software further detects events in which power density for these frequency bands exceeds normal ECoG by more than 4 standard deviations. We found that epileptic events could be identified and distinguished from a variety of ECoG phenomena associated with normal changes in behavior. We further found that analysis of spectral entropy was less effective in distinguishing epileptic from normal changes in ECoG activity.

Conclusion

The software presented here was a successful modification of EEGLAB in the Matlab environment that allows detection of epileptiform ECoG signals in animals after TBI. The code allows import of large EEG or ECoG data records as standard text files and uses fast fourier transform as a basis for detection of abnormal events. The software can also be used to monitor injury-induced changes in spectral entropy if required. We hope that the software will be useful for other investigators in the field of traumatic brain injury and will stimulate future advances of quantitative analysis of brain electrical activity after neurological injury or disease.

Ethnicity is a frequently used measure in research into youth and sexuality in the Netherlands, a country known and admired for its favourable sexual health outcomes. This paper critically examines the production of knowledge about sexuality and ethnicity in the Netherlands. It traces the concept of ethnicity through four research practices (rationales of taking up ethnicity and compiling research populations; determining ethnicity; statistical calculations and making recommendations). It shows how the notion of ethnicity is flexible, slippery and changeable, yet at the same time becomes solidified and naturalized in relation to sexuality. The paper is based on a literature review of youth and sexuality in the Netherlands.  相似文献
