The structures and conformational peculiarities of five members of the callatostatin family of neuropeptides, i.e. Leu- and Met-callatostatins, ranging in size from 8 to 16 amino acid residues have been investigated by a theoretical conformational analysis method. A comparative analysis of the conformational flexibilities of Met-callatostatin with those of the hydroxylated analogues, [Hyp2]- and [Hyp3]-Met-callatostatin has been carried out. Helically packed C-terminal pentapeptide in the structure of all investigated Leu-callatostatins are shown to be possible. The reason for the great number low-energy conformers for the callatostatin N-terminus is discussed.  相似文献

The organization of the centrosome in PK cells has been analysed according to several parameters: the presence of primary cilium, the number of pericentriolar satellites, the presence of striated rootlets, the distance between two centrioles and their orientation as regards the substrate plane. 2,4-Dinitrophenol (DNP), DNP with deoxyglucose (DOG), sodium azide cause the increase of frequency of occurrence of primary cilia and the growth of mean number of satellites per active centriole. The distribution of active and inactive centrioles in control cells is described by a histogram corresponding to a histogram of accidental distribution. Under the action of DNP or DNP with DOG, but not of sodium azide a part of active centrioles settled down perpendicularly to the substrate plane increases. The orientation of inactive centrioles under all the treatments used doesn't practically change.  相似文献

The disruption of microtubules with nocodazole or microfilaments with cytochalasin B did not prevent mother centrioles from nonrandom, preferentially perpendicular orientation with respect to the substrate plane after FCCP treatment. The microtubules affect negatively the reorientation of centrioles, because after their disruption by nocodazole the percentage of centrioles with the perpendicular orientation (the angle is tipped to the substrate plane by over 74 degrees) is seen to increase. The microfilaments have the positive effect, because after their disruption by cytochalasin B the share of centrioles with the perpendicular orientation decreases. Thus, our observations do not support the hypothesis that the long microtubules can provide the perpendicular orientation of centrioles anchoring them in the cytoplasm.  相似文献

In order to define more accurately normal dimensions and form of the lacrimal point as well as peculiarities in the structure of the vertical part of the lacrimal canaliculus, 315 practically healthy persons (630 eyes) at the age of 18-60 years have been examined. The procedure has been performed by means of the slit lamp (shchl-1) . The lacrimal point has been found to have a round (36.1%) and an oval (63.9%) form. Transition of the lacrimal point into the vertical part of the lacrimal canaliculus in 83.5% of the observations is "fannel"-shaped with a maximal narrowing at the depth of 0.4-0.5 mm, and in 16.5%-- well"-shaped (without any narrowing). The normal diameter of the lacrimal point is 0.2-0.6 mm (the arithmetic mean-- 0.35 mm). When the diameter of the lacrimal point is less than 0.2 mm (at least in one of its sections), the lacrimation function correspondingly deteriorates, indices of the coloured nasolacrimal test become decreased or negative.  相似文献

During recovery after a long (up to 12 h) treatment of pig embryo culture cells (PK) with nocodazole at concentrations of 0.02 g/ml and 0.2 g/ml all c-metaphase cells divide normally into two daughter cells. During recovery after a short (1–4 h) treatment with 0.6 g/ml nocodazole only multipolar mitoses (as a rule tripolar) arise. At the ultrastructural level, the increasing nocodazole concentration leads to progressive disruption of the mitotic spindle. At a nocodazole concentration of 0.2 g/ml kinetochores are not associated with microtubules. At a nocodazole concentration of 0.6 g/ml there are no microtubules around the centrosomes, and in every cell one of the two diplosomes disintegrates. In tripolar telophase centrioles are distributed among the spindle poles generally in a 2:2:0 pattern. Mother and daughter centrioles are always disoriented but not separated. The centriole-free pole contains a cloud of electron-dense material. During tripolar division two of the three daughter cells mainly fuse shortly after telophase forming one binucleate cell. Thus a multipolar mitosis arises as a result of the uncoupling of mother centrioles and spindle microtubules, but not of the duration of the c-mitotic arrest. Centriole-free poles account for the divergence of chromosomes, but mainly they are unable to ensure the normal cytokinesis of daughter cells.by M. Trendelenburg  相似文献

In this work, we report the use of refractive index (RI) tomography for quantitative analysis of unstained DH82 cell line infected with Leishmania infantum. The cell RI is reconstructed by using a modality of optical diffraction tomography technique that employs partially coherent illumination, thus enabling inherent compatibility with conventional wide‐field microscopes. The experimental results demonstrate that the cell dry mass concentration (DMC) obtained from the RI allows for reliable detection and quantitative characterization of the infection and its temporal evolution. The RI provides important insight for studying morphological changes, particularly membrane blebbing linked to an apoptosis (cell death) process induced by the disease. Moreover, the results evidence that infected DH82 cells exhibit a higher DMC than healthy samples. These findings open up promising perspectives for clinical diagnosis of Leishmania.  相似文献

We studied the influence of alien cytoplasm of spring goatgrass Aegilops ovata L. on some physiological parameters in winter wheat (Triticum aestivum L.), Mironovskaya 808, under normal conditions and in the case of modified source-sink relations. Measurements of relative rates of plant dry matter growth and its distribution among organs, CO2 exchange (photosynthesis upon light saturation and dark respiration), content of sugars (sucrose + glucose + fructose) and their ratio in leaves, frost hardiness, and indices of membrane stability and damage of leaves by frost have shown that, on average, alloplasmic hybrid differed from the initial cultivar by almost all parameters. Reduced frost hardiness, increased index of leaf damage by frost, lowered leaf content of sugars, and reduced sucrose/(glucose + fructose) ratio in the alloplasmic hybrid were combined with higher roots/leaves ratio, relative rate of dry matter growth, and photosynthesis and respiration rates. The alloplasmic hybrid was more tolerant to decreased source strength in source-sink relations as compared to the initial cultivar.  相似文献

Spatial structure and conformational properties of the Dippu-AST 8 (allatostatin III) neuropeptide have been investigated by the molecular mechanics method. The conformational energy and geometrical parameters corresponding to the low-energy states of the molecule are obtained. A single backbone conformation with a very restricted set of Ser 3, Phe 4 and Leu 9 amino acids positions is observed for Dippu-AST 8 neuropeptide.  相似文献

Individual ionic channels were shown to be formed in the brain cholesterol containing phospholipid membranes by two-sided addition of the amphotericin B alkyl derivatives. At concentrations between 10(-8) and 10(-7) M, the resulting conductance appeared to be transient. Existence of different antibiotic assemblies was justified by the kinetic analysis of the membrane conductance decline following the antibiotic washing out. In order to account for the transient characteristics of the induced conductance, it was proposed that the antibiotic oligomers incorporate into the membrane from the aqueous phase, form channels aggregating with cholesterol, and then dissociate in the bilayer into non-active degraded oligomeric or monomeric forms.  相似文献
