The role of the cysteine-rich region of the β2 integrin subunit in the leukocyte function-associated antigen-1 (LFA-1, αLβ2, CD11a/CD18) heterodimer formation and ligand binding

The cysteine-rich region (CRR) of the β2 integrin subunit was replaced by that of β1 to give the chimera β2NV1. β2NV1 can combine with αL to form a variant leukocyte-function-associated antigen (LFA)-1 on COS cell surface, suggesting that the specificity of the β2 interaction with αL does not lie in the CRR. Unlike those expressing wild-type LFA-1, COS cells expressing αLβ2NV1 are constitutively active in intercellular adhesion molecule (ICAM)-1 adhesion. These results suggest that activation of LFA-1 involves the release of an intramolecular constraint, which is maintained, in part, by the authentic β2 CRR.     

Escherichia coli RecBC pseudorevertants lacking chi recombinational hotspot activity

Pseudorevertants of an Escherichia coli exonuclease V (RecBC enzyme)-negative mutant have been isolated after ethyl methane sulfonate mutagenesis of a recC73 (presumed missense) mutant. The remedial mutations in each of the four pseudorevertants studied in detail map and complement as recC mutations. By several criteria, such as recombination proficiency, support of phage growth, RecBC nuclease activity, and cell viability, the pseudorevertants appear to have regained partially or completely various aspects of RecBC activity. However, chi recombinational hotspots, which stimulate exclusively the RecBC pathway of recombination, have no detectable activity in lambda vegetative crosses in the pseudorevertants. The properties of these mutants, in which the RecBC pathway of recombination is active yet in which chi is not active, are consistent with the hypothesis that wild-type RecBC enzyme directly interacts with chi sites; alternatively, the mutants may block or bypass the productive interaction of another recombinational enzyme with chi.     

Skeletal Ultrastructure in some Tubuliporine Cyclostome Bryozoans

The ultrastructure of the calcareous skeleton is described in twenty–one species of recent tubuliporine cyclostome bryozoans, using field emission SEM. The succession of skeletal fabrics in interior walls may be classified into four different fabric suites. The first–formed part of the calcitic skeleton in all species for which it has been observed is a precursory fabric of tiny, wedge–shaped crystallites. This is succeeded in about half of the species studied by a fabric of transverse fibres, followed by foliated fabric and often semi–nacre (fabric suite 1). Most of the remaining species lack transverse fibres and have interior walls largely comprising semi–nacre (fabric suite 2). A few species have skeletons consisting of predominantly distally–oriented, irregularly or regularly foliated fabric (fabric suite 3). A single species has a skeleton of proximally–oriented foliated fabric (fabric suite 4). Basal exterior walls in all species have a precursory fabric of tiny wedge–shaped crystallites without a strong preferred orientation, deposited directly upon the organic cuticle, followed by a layer of planar spherulitic structure, which in turn is succeeded by a similar fabric to that developed in the interior wall of the species concerned. Outermost layers of frontal exterior walls exhibit one of the following combinations of three fabrics: an outer layer of (1) finely granular or wedge–shaped crystallites; a thin dense granular layer followed by (2) distally accreting planar spherulitic fabric., or (3) obliquely accreting planar spherulitic fabric growing partly towards the midline of the frontal wall. Terminal diaphragms usually have outer layers dominated by planar spherulitic ultrastructure with centripetal growth directions. The fabric suites present in tubuliporines encompass most known fabrics found in the other cyclostome suborders and support the notion that this species–rich suborder occupies a central position in cyclostome evolution.     

Thermoregulation during prolonged exercise in heat: alterations with beta-adrenergic blockade

Thermoregulation and cardiovascular drift were studied under conditions of prolonged exercise in a warm environment (dry bulb temperature 31.7 +/- 0.3 degrees C, rh 44.7 +/- 4.7%) during beta-adrenergic blockade. Fourteen subjects performed 90-min rides on a cycle ergometer at a work rate equivalent to 40% of their control maximal O2 uptake under each of three treatments provided in a randomized double-blind manner: atenolol (100 mg/day), propranolol (160 mg/day), and a placebo. Exercise during the propranolol trial resulted in significantly higher forearm vascular resistance values and significantly lower forearm blood flows (FBF) compared with the placebo trial. However, the significantly lower FBF during propranolol did not significantly alter the rectal temperature (Tre) response to prolonged exercise. In addition, both beta-blockers produced lower FBF for any given Tre, suggesting that beta-adrenergic blockade affects FBF through nonthermal factors. The slight differences in Tre, despite the large differences in FBF between the various treatments, are apparently the result of an enhanced sweat loss and a lower mean skin temperature during exercise with beta-blockade. The uncoupling of FBF and sweat loss provides evidence of independent regulation. The reduction in FBF at any given Tre was concomitant to lower blood pressure values during beta-blockade and suggests that baroreflexes provide significant input to the control of skin blood flow when both pressure and temperature maintenance are simultaneously challenged.
