The ARF GTPase Activating Protein 1 (ARFGAP1) associates mainly with the cytosolic side of Golgi cisternal membranes where it participates in the formation of both COPI and clathrin-coated vesicles. In this study, we show that ARFGAP1 associates transiently with lipid droplets upon addition of oleate in cultured cells. Also, that addition of cyclic AMP shifts ARFGAP1 from lipid droplets to the Golgi apparatus and that overexpression and knockdown of ARFGAP1 affect lipid droplet formation. Examination of human liver tissue reveals that ARFGAP1 is found associated with lipid droplets at steady state in some but not all hepatocytes.  相似文献

In this paper, we demonstrate a novel salinity sensor based on Tamm-plasmon-polariton (TPP), comprising different shapes of Bragg reflector (ordinary, texturing, and sawtooth) and metallic layer. The finite element method is used to study the considered structure and sensing performance by using the COMSOL multiphysics simulation procedure. Here, we study the effect of surface morphology on the sensitivity; firstly, in the case of one-dimensional photonic crystal-centered defect, it harms the sensitivity; secondly, texturing and sawtooth in the case of Tamm resonance increases the sensitivity, as for texturing the surface, the sensitivity quality factor (Q)?=?236 and figure of merit (FOM)?=?170. For sawtooth surfaces, Q?=?272.4, and FOM?=?199. The consequences of structural parameters on the efficiency of sensing are studied, and new procedures are proposed to enhance TPP-based sensors. A simple and functional alternative to conventional salinity sensors may be the proposed solution.

A bacterium capable of degrading microcystin-RR (MC-RR) was isolated from a Saudi eutrophic lake which was previously reported to have microcystin-producing cyanobacteria. Based on the analysis of the 16S rRNA gene sequence, the isolated strain SSZ01, most likely belong to the genus Bacillus with a highest sequence similarity (99%) with Bacillus flexus strain EMGA5. It was found that B. flexus strain SSZ01, possesses an mlrA gene encoding the most important enzyme for MC degradation. This strain was capable of degrading MC-RR, at a concentration of 10 mg l−1, in batch experiments under environmentally relevant conditions. The degradation of MC-RR was completely removed within 4 d. The degradation of MC-RR by this strain occurred in a rich medium nutrient broth (NB), indicating that this could likely occur along with other organic compounds found in the environment. Therefore, the coexistence of such bacteria with MCs in the same environment can contribute to the self-purification of the ecosystem from such potent toxins. This is the first study to report that B. flexus can degrade MCs.  相似文献

Background and objectiveCoronavirus 2019 (COVID-19) is caused by ‘severe acute respiratory syndrome coronavirus 2′ (SARS-CoV-2), first reported in Wuhan, China in December 2019, which eventually became a global disaster. Various key mediators have been reported in the pathogenesis of COVID-19. However, no effective pharmacological intervention has been available to combat COVID-19 complications. The present study screens nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) as potential inhibitors of this present generation coronavirus infection using an in-silico approach.Materials and methodsThe SARS-CoV-2 proteins (nucleocapsid, proteases, post-fusion core, phosphatase, endoriboruclease) and ACE-2 protein were selected. The 2D structure of nicotinamide ribonucleoside and nicotinamide ribonucleotide was drawn using ChemDraw 14.0 and saved in .cdx format. The results were analyzed using two parameters: full fitness energy and binding free energy (ΔG).ResultsThe full fitness energy and estimated ΔG values from docking of NM, and NMN with selected SARS-CoV-2 target proteins, ADMET prediction and Target prediction indicate the interaction of NR and NMN in the treatment of COVID-19.ConclusionsBased on full fitness energy and estimated ΔG values from docking studies of NM and NAM with selected SARS-CoV-2 target proteins, ADME prediction, target prediction and toxicity prediction, we expect a possible therapeutic efficacy of NR in the treatment of COVID-19.Keyword: COVID-19, SARS-CoV-2, Molecular docking, Poly (ADP-ribose) polymerase enzyme, Nicotinamide  相似文献

BackgroundFoot complications are considered to be a serious consequence of diabetes mellitus, posing a major medical and economical threat. Identifying the extent of this problem and its risk factors will enable health providers to set up better prevention programs. Saudi National Diabetes Registry (SNDR), being a large database source, would be the best tool to evaluate this problem.MethodsThis is a cross-sectional study of a cohort of 62,681 patients aged ≥25 years from SNDR database, selected for studying foot complications associated with diabetes and related risk factors.ResultsThe overall prevalence of diabetic foot complications was 3.3% with 95% confidence interval (95% CI) of (3.16%–3.44%), whilst the prevalences of foot ulcer, gangrene, and amputations were 2.05% (1.94%–2.16%), 0.19% (0.16%–0.22%), and 1.06% (0.98%–1.14%), respectively. The prevalence of foot complications increased with age and diabetes duration predominantly amongst the male patients. Diabetic foot is more commonly seen among type 2 patients, although it is more prevalent among type 1 diabetic patients. The Univariate analysis showed Charcot joints, peripheral vascular disease (PVD), neuropathy, diabetes duration ≥10 years, insulin use, retinopathy, nephropathy, age ≥45 years, cerebral vascular disease (CVD), poor glycemic control, coronary artery disease (CAD), male gender, smoking, and hypertension to be significant risk factors with odds ratio and 95% CI at 42.53 (18.16–99.62), 14.47 (8.99–23.31), 12.06 (10.54–13.80), 7.22 (6.10–8.55), 4.69 (4.28–5.14), 4.45 (4.05–4.89), 2.88 (2.43–3.40), 2.81 (2.31–3.43), 2.24 (1.98–2.45), 2.02 (1.84–2.22), 1.54 (1.29–1.83), and 1.51 (1.38–1.65), respectively.ConclusionsRisk factors for diabetic foot complications are highly prevalent; they have put these complications at a higher rate and warrant primary and secondary prevention programs to minimize morbidity and mortality in addition to economic impact of the complications. Other measurements, such as decompression of lower extremity nerves, should be considered among diabetic patients.  相似文献

At present plants continuously exposed to salinity stress due to the challenging environment that has reduced the crop growth and productivity worldwide. Application of phytohormones by using seed priming method emerges as one of the most reliable and cost effective to alleviate the toxic effect of salinity stress. In this study, we evaluate the effect of seed-primed salicylic acid (SA) to reduce the adverse effect of different salt concentrations (0, 100, 200, and 300 mM NaCl) in pea (Pisum sativum L.) seedlings. After seedling emergence, percent seed germination was calculated; however, after 60 days; plants were sampled for studying the growth and photosynthetic traits, lipid peroxidation level, antioxidant activities, ions accumulation, and its sequestration. The results depicted that salinity treatments hampered overall growth performance and induced oxidative stress in a dose-dependent manner. Salinity also has negatively influence on ion accumulation as Na+ ion increased while K+ ion decreased. On the other hand, seed priming with SA significantly reduced the salinity-induced effects on the overall performance of plants, including growth and photosynthetic attributes. SA alleviated the adverse effect of salinity even at higher salinity level by inducing enzymatic and non-enzymatic antioxidant systems, soluble sugars, and proline accumulation, and regulating ion homeostasis along with up-regulation of Na+/H+ antiporters (SOS1 and NHX1). Thus, seed priming with SA shows a comprehensive role in mitigation of salinity stress and can be used as a model for promising salinity tolerant cultivation.
