A distribution survey of the endangered Sclater's monkey ( Cercopithecus sclateri ) was conducted over a wide area in southern Nigeria using forest surveys and hunter interviews. Sclater's monkey, Nigeria's only endemic primate species, is restricted to a land area of about 28,500 km2 in the densely human-populated, oil-producing region of southern Nigeria. Results indicate that this species is not as rare as previously thought; we confirmed its presence in 27 formerly unknown sites. Based on encounter-rate and distribution data, Sclater's monkey is one of the two most abundant diurnal primate taxa across its range. However, the species primarily occupies isolated and degraded forest fragments. Although hunting is widespread, selective hunting of larger-bodied primate taxa offers some respite for the smaller Sclater's monkey. We encountered this species more frequently in forests with relatively high hunting pressure, possibly indicating competitive release in the heavily hunted forests of southern Nigeria. Long-term persistence of Sclater's monkey, which has no official protection throughout its range, depends on the willingness of hunters to target smaller-bodied wildlife (effort-profit trade-off), local bushmeat demand and protection of key forest fragments and the few larger forests in the region.  相似文献

Although primates are hunted on a global scale, some species are protected against harassment and killing by taboos or religious doctrines. Sites where the killing of sacred monkeys or the destruction of sacred groves is forbidden may be integral to the conservation of certain species. In 2004, as part of a distribution survey of Sclater's guenon (Cercopithecus sclateri) in southern Nigeria, we investigated reports of sacred monkeys in the Igbo‐speaking region of Nigeria. We confirmed nine new sites where primates are protected as sacred: four with tantalus monkeys (Chlorocebus tantalus) and five with mona monkeys (Cercopithecus mona). During 2004–2006, we visited two communities (Akpugoeze and Lagwa) previously known to harbor sacred populations of Ce. sclateri to estimate population abundance and trends. We directly counted all groups and compared our estimates with previous counts when available. We also estimated the size of sacred groves and compared these with grove sizes reported in the literature. The mean size of the sacred groves in Akpugoeze (2.06 ha, n=10) was similar to others in Africa south of the Sahel, but larger than the average grove in Lagwa (0.49 ha, n=15). We estimated a total population of 124 Sclater's monkeys in 15 groups in Lagwa and 193 monkeys in 20 groups in Akpugoeze. The Akpugoeze population was relatively stable over two decades, although the proportion of infants declined, and the number of groups increased. As Sclater's monkey does not occur in any official protected areas, sacred populations are important to the species' long‐term conservation. Despite the monkeys' destruction of human crops, most local people still adhere to the custom of not killing monkeys. BACKGROUND: Poor graft function without rejection may occur after stem cell transplantation (SCT). CD34(+) stem cell boost (SCB) can restore marrow function but may induce or exacerbate GvHD. We therefore investigated the feasibility and efficacy of CD34(+)-selected SCB in some patients with poor graft function. We present the results for eight patients (median age 46 years) transplanted initially for myelofibrosis, acute leukemia, myeloma and NHL. Six patients had received HLA-matched and two mismatched grafts (PB, BM; n=5, 3). After a median of 128 days post-transplant, the median leukocyte and platelet counts were, respectively, 2.05/nL and 18/nL. None had achieved platelet counts >50/nL even though donor chimerism was >95% in seven recipients. METHODS: Positive selection of CD34(+) stem cells was performed on a CliniMACS device, observing GMP and achieving a median of 98.5% purity. The patients received a median of 1.7 x 10(6)/kg CD34(+) cells and 2.5 x 10(3)/kg CD3(+) T lymphocytes. RESULTS: Hemograms at days +30, +60 and +90, respectively, showed steadily increasing median leukocyte (2.55, 3.15 and 4.20/nL) and platelet (29, 39 and 95/nL) counts. After a median follow-up of 144 days, five patients remained alive. No patient had developed acute or chronic GvHD. One patient died of leukemic relapse and two others of systemic mycosis. DISCUSSION: These preliminary results point to the possibility of safely improving graft function using CD34(+) positively selected stem cells without necessarily increasing the incidence of GvHD in patients with poor graft function post-SCT. Experience with more patients and longer follow-up should clarify the optimal role for this procedure.  相似文献

Count-based indices and distance sampling are widely used to monitor primate populations. Indices are often confounded by variation in detectability, whereas distance sampling is generally ineffective with species that flee or hide from observers and where it is difficult to accurately measure detection distances. We tested occupancy modeling as a means to monitor Sclater’s monkey (Cercopithecus sclateri), an endemic of Nigeria. We evaluated effects of survey methodology, habitat, and human disturbance on detection probability and site occupancy. Average detectability was high (p = 0.81), but varied substantially between two observers. Occupancy was highest in areas with intermediate levels (20–40%) of farmland and secondary forest, and was unaffected by human disturbance. Sampling plots (4 and 6.25 ha) did not concurrently contain >1 monkey group, were likely closed to monkey movements during the replicate surveys of each plot, and were spatially separated so that it was unlikely the same group was observed in >1 plot. These conditions enabled the conversion of occupancy to group density. Scaled to 6.25 ha, model-weighted occupancy averaged 0.230 (SE 0.103), yielding an estimate of 3.7 groups/km2 (95% CI 1.4–7.7 groups/km2). Because some groups straddled plot boundaries, we assumed that half of these groups were inside the plots, resulting in an adjusted estimate of 3.1 groups/km2. Our results illustrate that occupancy can be suitable for monitoring vigilant forest primates where detection distances are difficult to measure. However, special attention is required to choose spatial and temporal scales that accommodate the method’s closure and independent-detection assumptions.  相似文献

The International Stem Cell Initiative characterized 59 human embryonic stem cell lines from 17 laboratories worldwide. Despite diverse genotypes and different techniques used for derivation and maintenance, all lines exhibited similar expression patterns for several markers of human embryonic stem cells. They expressed the glycolipid antigens SSEA3 and SSEA4, the keratan sulfate antigens TRA-1-60, TRA-1-81, GCTM2 and GCT343, and the protein antigens CD9, Thy1 (also known as CD90), tissue-nonspecific alkaline phosphatase and class 1 HLA, as well as the strongly developmentally regulated genes NANOG, POU5F1 (formerly known as OCT4), TDGF1, DNMT3B, GABRB3 and GDF3. Nevertheless, the lines were not identical: differences in expression of several lineage markers were evident, and several imprinted genes showed generally similar allele-specific expression patterns, but some gene-dependent variation was observed. Also, some female lines expressed readily detectable levels of XIST whereas others did not. No significant contamination of the lines with mycoplasma, bacteria or cytopathic viruses was detected.  相似文献

The emergence of medicinal indications for stem cell therapies has seen a need to develop the manufacturing capacity for adherent cells such as mesenchymal stem cells (MSCs). One such development is in the use of microcarriers, which facilitate enhanced cell densities for adherent stem cell cultures when compared with 2D culture platforms. Given the variety of stem cell expansion systems commercially available, novel methods of non‐invasive and automated monitoring of cell number, confluence, and aggregation, within disparate environments, will become imperative to process control, ensuring reliable and consistent performance. The in situ epi‐illumination of mouse embryonic fibroblasts and human mesenchymal stem cells attached to Cytodex 1 and 3 microcarriers was achieved using a bespoke microscope. Robust image processing techniques were developed to provide quantitative measurements of confluence, aggregate recognition, and cell number, without the need for fluorescent labeling or cell detachment. Large datasets of cells counted on individual microcarriers were statistically analyzed and compared with NucleoCounter measurements, with an average difference of less than 7% observed from days 0 to 6 of a 12‐day culture noted, prior to the onset of aggregation. The developed image acquisition system and post‐processing methodologies were successfully applied to dynamically moving colonized microcarriers. The proposed system offers a novel method of cell identification at the individual level, to consistently and accurately assess viable cell number, confluence, and cell distribution, while also minimizing the variability inherent in the current invasive means by which cells adhered to microcarriers are analyzed. Four cycles of S(1) family recurrent selection to improve grain yield and resistance to Striga hermonthica have been completed in TZE-Y Pop STR C(0.) In order to determine whether or not to continue with the recurrent scheme, it was desirable to evaluate the amount of residual genetic variance and associated parameters in the population. The objective of this study was to characterize the relative changes in the levels of the genetic variances, heritability estimates and genetic correlation coefficients, and to predict future gains from selection for grain yield, Striga resistance and other agronomic traits. Fifty S(1) families, derived from each cycle, were evaluated under Striga-infested and Striga-free conditions at Mokwa, Ikenne and Abuja, Nigeria, in 2005 and 2007. Under Striga infestation, genetic variances for grain yield, days to anthesis, plant height and Striga damage generally increased in the advanced cycles of selection. In contrast, the genetic variances for days to silk, anthesis-silking interval, ears per plant, ear aspect and number of emerged Striga plants decreased with selection. The advanced cycles of selection significantly out-yielded the original cycle in both research environments. Heritabilities for grain yield, Striga damage and number of emerged Striga plants were significantly greater than zero. The realized gains from selection for grain yield under Striga infestation (52?kg?ha(-1)?cycle(-1)) and Striga-free conditions (130?kg?ha(-1)?cycle(-1)) were remarkably lower than the predicted gains (350 and 250?kg?ha(-1?)cycle(-1), respectively). Adequate genetic variability exists in cycle 4 of the scheme to ensure future gains from selection.  相似文献

Modifications of neurobehavioral activities related to single episodes of consumption of different doses of bee honey were examined in rats under conditions of the hole-board (HB) test (to evaluate the level of anxiety) and open-field (OF) test (where the intensities of locomotion, rearing, and grooming were measured). Animals of all subgroups had free access to normal saline, while rats of the three experimental subgroups consumed bee honey in the doses of 0.5, 1.0, and 2.0 g per 1 kg body mass (in the form of 10, 20, and 40% solutions, respectively). Among the doses tested, only higher ones induced considerable changes in the behavioral indices. The highest dose (2.0 g/kg) provided a more than twofold increase in the number of examined holes in the HB test; in the OF test, it also increased the numbers of crossed squares, rearings, and grooming episodes by 30, 37, and 164%, respectively. Thus, our experiments demonstrated a rather significant ability of the natural product tested to relieve anxiety and intensify motor, research/orientational, and grooming aspects of behavior even upon single acts of consumption. Possible neurophysiological mechanisms underlying the behavioral modifications observed are discussed.  相似文献

Salmonellae are widely distributed but nomenclaturally controversial pathogens of both humans and animals. Despite elaborate studies, much still remain to be discovered about these organisms. Although Salmonella nomenclature has proved to be rather complex, in 2005, Salmonella enterica finally gained official approval as the type species of the genus Salmonella. In addition, one other species has been approved and recognised in the genus Salmonella, namely, Salmonella bongori. New serovars (serotypes) are continually being discovered each year and reported in the journal Research in Microbiology. Salmonella serovars and their antigenic formulae are listed in the White–Kauffmann–Le Minor scheme and updated by the World Health Organization’s Collaborating Centre for Reference and Research on Salmonella at the Pasteur Institute, Paris, France.  相似文献
