The uptake of six 9-aminoacridinecarboxamide derivatives by L1210 cells in relation to their lipophilicity and cytotoxic activity was studied. The amount of acridines taken up by cells was estimated by fluorimetric measurements. It was found that the uptake efficiency of this class of compounds by cells depends on the size of carboxamide residue as well as on position of the substituent. The increase of size of carboxamide chain resulted in the loss of capability of acridines to penetrate cell membrane. Cytotoxic effects of acridines were well correlated with the level of drugs accumulated by cells, whereas no clear correlation between uptake and lipophilicity was observed. It is concluded that uptake of 9-aminoacridinecarboxamides is the most important factor determining their antiproliferative activity.  相似文献

Habitat spatial distribution, seasonal variation, and activity patterns influence changes in vertebrate assemblages over time. Terrestrial birds play major roles in the dynamics of tropical forests, but there are few effective methods to study these species due to their cryptic coloration and elusive behavior. We used camera‐trap data collected during 16 mo (February 2017–June 2018) to describe the terrestrial avifauna in southeastern Peru, assess to what extent the composition of terrestrial avifauna changes among seasons and across two major habitats (terra firme and floodplain forests), and determine daily activity patterns of terrestrial birds. We used overlap analyses to examine temporal co‐occurrence between ecologically similar and sympatric species. Camera traps recorded 16 species, including eight species in the family Tinamidae. Capture rates were highest for Pale‐winged Trumpeters (Psophia leucoptera; Psophiidae) and Gray‐fronted Doves (Leptolila rufaxilla; Columbidae). Species composition did not differ between habitats or seasons, and capture rates between habitats only differed for White‐throated Tinamous (Tinamus guttatus). Overlaps of activity patterns were high between ecologically similar species and species found in terra firme habitats (White‐throated Tinamous and Cinereous Tinamous, Crypturellus cinereus) and in both habitat types (Pale‐winged Trumpeters and Gray‐fronted Doves). Low numbers of captures of possibly locally rare or less abundant species hindered a complete analysis of spatial and seasonal patterns of terrestrial bird assemblages. We suggest a greater sampling effort and greater spatial replication to better understand the spatial and seasonal dynamics of the terrestrial avifauna. Further studies that assess the mechanisms that allow the coexistence of sympatric tinamous would be valuable, both in our study area and elsewhere. The use of camera traps in long‐term monitoring projects proved to be an effective tool for monitoring terrestrial birds, identifying cryptic and often rare animals to species level, and providing valuable ecological information at species and community levels.  相似文献

Biodiversity information in Angola is limited or nonexistent, hindering the design and implementation of conservation strategies. The Escarpment forest is one of the most important areas for bird diversity in the country. However, there is almost no information about the territorial needs and habitat preferences of its threatened endemic birds. This study evaluated these needs and preferences in Gabela akalat Sheppardia gabela, a range‐restricted endemic to the Central Escarpment. Eighteen individuals of Gabela akalat were captured and radio‐tracked with the objectives of establishing their territory size (through home‐range size estimates) and habitat preferences using compositional analysis. Home‐range sizes were slightly larger than other Sheppardia species and Gabela akalat evidently avoided clearings and preferred forest habitat, although it was also able to use farmland areas and secondary growth to a lesser extent. Conservation measures should focus on the preservation of remaining old‐growth forest through the establishment of a nature reserve in Kumbira. To assure the success of such an initiative, the local population should participate in planning, administration and enforcement. We outline some measures that could help address the economic needs of the local community while maintaining forest cover.  相似文献

Herbivorous animals face shortages of different minerals in different geographic areas. In the Amazon Basin, sodium is often limiting, driving herbivores to seek supplemental sources. In the lowlands of the western Amazon Basin, parrots commonly consume sodium‐rich soils at clay licks but lick use varies widely among species, and to date, parrots in the region have not been reported consuming other supplemental sodium sources. We document 11 species of psittacines consuming sodium‐rich leaves and trunks of Attalea butyracea palms growing on sodium‐rich soils in lowland Peru. Consumed palms had more sodium and less potassium than uneaten A. butyracea palms and other palm species in the area. Among A. butyracea palm parts, sodium and Na:K ratios were highest in trunks (consumed by parrots in 94% of the 387 foraging bouts recorded) and lowest in leaves (consumed in only 14% of foraging bouts). The low potassium and high Na:K ratio suggest that birds may be seeking not just any sodium sources, but those low in potassium, as potassium is known to exacerbate dietary sodium shortages. Use of the palms and species’ abundance in the study area were not correlated. Instead, parrot species that consumed palms the most were those that use relatively few traditional soil clay licks. This finding suggests that parrot species in the region have fundamental differences in preferred strategies for obtaining supplemental sodium and may help explain documented interspecific differences in geophagy.  相似文献

A fast screening method of whole blood was proposed for enantiorecognition of free L‐T3 , L‐T4, and D‐T4. Stochastic microsensors based on four inulins (IN, IQ, TEX, and HD) immobilized on diamond paste (DP) were used for recognition of free L‐T3 , L‐T4, and D‐T4. For the enantiorecognition of free L‐T4 and D‐T4 in whole blood and pharmaceutical samples, the best microsensor was the one based on TEX/DP (wide linear concentration ranges, and low limits of quantification). The best limit of detection for the assay of free L‐T3 (400 fmol/L) was recorded using the microsensors based on HD/DP, while for the assay of free L‐T4, and D‐T4 the best limit of determination (1 pmol/L) was recorded using the TX/DP‐based microsensor. For the enantiorecognition of free L‐T3 in whole blood and pharmaceutical samples the best microsensor was the one based on HD/DP (the wider linear concentration range, and the lower limit of quantification – of pmol/L magnitude order). In this work, non-invasive high-spatial resolution three-dimensional (3D) X-ray micro-computed tomography (μCT) of healthy mouse lung vasculature is performed. Methodologies are presented for filtering, segmenting, and skeletonizing the collected 3D images. Novel methods for the removal of spurious branch artefacts from the skeletonized 3D image are introduced, and these novel methods involve a combination of distance transform gradients, diameter-length ratios, and the fast marching method (FMM). These new techniques of spurious branch removal result in the consistent removal of spurious branches without compromising the connectivity of the pulmonary circuit. Analysis of the filtered, skeletonized, and segmented 3D images is performed using a newly developed Vessel Network Extraction algorithm to fully characterize the morphology of the mouse pulmonary circuit. The removal of spurious branches from the skeletonized image results in an accurate representation of the pulmonary circuit with significantly less variability in vessel diameter and vessel length in each generation. The branching morphology of a full pulmonary circuit is characterized by the mean diameter per generation and number of vessels per generation. The methods presented in this paper lead to a significant improvement in the characterization of 3D vasculature imaging, allow for automatic separation of arteries and veins, and for the characterization of generations containing capillaries and intrapulmonary arteriovenous anastomoses (IPAVA).  相似文献

WNT signaling is critical in most aspects of skeletal development and homeostasis, and antagonists of WNT signaling are emerging as key regulatory proteins with great promise as therapeutic agents for bone disorders. Here we show that Sost and its paralog Sostdc1 emerged through ancestral genome duplication and their expression patterns have diverged to delineate non-overlapping domains in most organ systems including musculoskeletal, cardiovascular, nervous, digestive, reproductive and respiratory. In the developing limb, Sost and Sostdc1 display dynamic expression patterns with Sost being restricted to the distal ectoderm and Sostdc1 to the proximal ectoderm and the mesenchyme. While Sostdc1−/− mice lack any obvious limb or skeletal defects, Sost−/− mice recapitulate the hand defects described for Sclerosteosis patients. However, elevated WNT signaling in Sost−/−; Sostdc1−/− mice causes misregulation of SHH signaling, ectopic activation of Sox9 in the digit 1 field and preaxial polydactyly in a Gli1- and Gli3-dependent manner. In addition, we show that the syndactyly documented in Sclerosteosis is present in both Sost−/− and Sost−/−; Sostdc1−/− mice, and is driven by misregulation of Fgf8 in the AER, a region lacking Sost and Sostdc1 expression. This study highlights the complexity of WNT signaling in skeletal biology and disease and emphasizes how redundant mechanism and non-cell autonomous effects can synergize to unveil new intricate phenotypes caused by elevated WNT signaling.  相似文献
