Manggur, which means to plane down or sharpen, is a manual process in producing copper blades of Balinese gamelan orchestra. The craftsmen of Manggur work 6 to 8 hours a day, sitting on the floor with folded legs and hunched back. Because the craftsmen often complain about musculoskeletal problems after having completed a full day work, an ergonomic intervention was made by changing their usual working posture (the first working posture) into working on tables while sitting on chairs for one hour and alternately standing for half an hour (the second working posture). Treatment by subject design was applied to 22 randomly chosen craftsmen. Resting heart rate and working heart rate were measured by using a stopwatch, and the number of musculoskeletal complaints were recorded with Nordic Body Map Questionnaire. As a result, the second working posture caused significant reductions in working heart rate, work pulse (the difference between working heart rate and resting heart rate) and the number of musculoskeletal complaints. These results suggest that the change of working posture in manggur decreases cardiovascular load and musculoskeletal strain among Balinese gamelan craftsmen.  相似文献

Development process brings the positive as well as negative impacts. Among others, the negative impacts include the extinction of some medicinal plants. The research issue is what strategy can be used for preserving the medicinal plants while little attention is given to them. A research action was thus started for preserving and conserving these plants. Methods used were participatory observation, field surveys, and demonstration-plots. The action consisted of: an awareness program, a preparation phase and the establishment of a park of medicinal plants. In doing that, the Systemic, Holistic, Interdisciplinary and Participatory (SHIP) approach was applied. Up to now, the results found are: 1) the awareness program was conducted several times; 2) two pieces of lands were provided from the provincial government of Bali and from the local community; 3) the field surveys identified about 200 kinds of medicinal plants in the house yards, government offices, hotels and median strips of roads; 4) in the traditional medicine texts (lontar), more then 500 kinds of medicinal plants were found to be cited; 5) the medicinal plants were classified into wooden trees, shrubs, grasses, watery plants, epiphytes and parasitic plants. It is concluded that preserving and conserving the medicinal plants in Bali is urgent and that there is a willingness to do that with the active support of the stakeholders. It is encouraging that good examples have already been established in Ubud, Tampaksiring and Karangasem. Therefore, it is recommended to establish a medicinal plants park in Bali since it will have multiplier effects.  相似文献

Species and sex identification are among the most important parameters for conservation management. However, it is extremely difficult to perform such identification in Formosa landlocked salmon (Oncorhynchus masou formosanus). Both sexual dimorphism in landlocked dwarf form Formosa landlocked salmon and morphological difference among cherry salmon complex are minimal. We developed a simple, rapid and noninvasive method for identifying sex and species of this critically endangered species using a loop-mediated isothermal amplification (LAMP) technique. The LAMP assay showed the advantage of simple detection (evaluated by visual inspection), rapid reaction time (< 1 h), isothermal condition (less equipment required) and high efficiency (only 0.5-5 pg of DNA was required in the reaction mixture). Therefore, the method is more economical and practical than PCR. The LAMP assay can be easily performed in the field and is a valuable tool for detecting sex ratios in wild populations and identifying species in commercial imports. This is the first application of LAMP in identifying species and sex of salmonids as far as we know and clearly shows the potential application of LAMP in molecular ecology and conservation efforts.  相似文献

Xenobiotics directed against sterol biosynthesis have proved to be useful tools in the determination of which sterol molecules are necessary for successful plant cell growth. However, the exact mode of action by which sterols are able to trigger cell growth remains to be elucidated. Previous studies using the triazole paclobutrazol, demonstrated that in Apium graveolens (cv. New Dwarf White) suspension cultures, sterol and phosphatidylcholine biosynthesis are co-ordinately regulated (C. E. Rolph and L. J. Goad 1991, Physiol. Plant. 83: 605–610). The studies presented herein, were designed to investigate the possible role of phosphatidylcholine in the stimulation of plant cell growth. Sterol biosynthesis, and hence cell growth, was inhibited by the use of the azole xenobiotic miconazole. Treatment of the cultures with miconazole lead to compositional changes in the free sterol content of the cells. For example, 30 μM miconazole treatment led to a reduction in the stigmasterol/sitosterol ratios from 1.53 to 1.24. In contrast, the phospholipid content of the cells remained relatively unchanged with phosphatidylcholine accounting for approximately 25% of the total phospholipids present in both control and miconazole-treated cells. The cytostatic effect of miconazole could be partially counteracted by supplementation of the growth medium with the phytosterol stigmasterol and/or the unsaturated fatty acids oleate and linoleate. The activity of CTP:cholinephosphate cytidylyltransferase (EC 2.7.7.15), a rate-limiting enzyme in phosphatidylcholine biosynthesis, was significantly reduced in cells whose growth had been arrested by miconazole treatment. In miconazole-treated cultures whose growth had been partially restored by supplementation with either specific sterols or unesterified fatty acids, the activity of this key enzyme was increased. In the case of stigmasterol, oleate and linoleate supplementation, the microsomal activity was found to be similar to that exhibited by control cultures. From these studies, it may be concluded that certain phytosterols and unsaturated fatty acids play key roles with respect to phosphatidylcholine biosynthesis and that phosphatidylcholine biosynthesis via the CDP-base pathway is an important pre- and/or co-requisite for successful culture growth.  相似文献

Background

Epidemiological studies have shown that imposing travel restrictions to prevent or delay an influenza pandemic may not be feasible. To delay an epidemic substantially, an extremely high proportion of trips (~99%) would have to be restricted in a homogeneously mixing population. Influenza is, however, strongly influenced by age-dependent transmission dynamics, and the effectiveness of age-specific travel restrictions, such as the selective restriction of travel by children, has yet to be examined.

Methods

A simple stochastic model was developed to describe the importation of infectious cases into a population and to model local chains of transmission seeded by imported cases. The probability of a local epidemic, and the time period until a major epidemic takes off, were used as outcome measures, and travel restriction policies in which children or adults were preferentially restricted were compared to age-blind restriction policies using an age-dependent next generation matrix parameterized for influenza H1N1-2009.

Results

Restricting children from travelling would yield greater reductions to the short-term risk of the epidemic being established locally than other policy options considered, and potentially could delay an epidemic for a few weeks. However, given a scenario with a total of 500 imported cases over a period of a few months, a substantial reduction in the probability of an epidemic in this time period is possible only if the transmission potential were low and assortativity (i.e. the proportion of contacts within-group) were unrealistically high. In all other scenarios considered, age-structured travel restrictions would not prevent an epidemic and would not delay the epidemic for longer than a few weeks.

Conclusions

Selectively restricting children from traveling overseas during a pandemic may potentially delay its arrival for a few weeks, depending on the characteristics of the pandemic strain, but could have less of an impact on the economy compared to restricting adult travelers. However, as long as adults have at least a moderate potential to trigger an epidemic, selectively restricting the higher risk group (children) may not be a practical option to delay the arrival of an epidemic substantially.  相似文献

Grasslands are one of the most widespread landscapes worldwide, covering approximately one-fifth of the world’s land surface, where grazing is a common practice. How carbon storage responds to grazing in steppes remains poorly understood. We quantified the effects of grazing on community composition and species diversity, and carbon storage in two typical grasslands of northeastern China, one in Horqin and the other one in Hulunbeier. In both grasslands, grazing did not influence plant species diversity. However, it substantially decreased aboveground carbon by 31% and 54% in Horqin and Hulunbeier, respectively. Fenced and grazing treatments showed a similar belowground carbon at both locations. The predominant carbon pool in the study grassland ecosystem was found in the upper 100 cm soil depth, from 98.2 to 99.1% of the total carbon storage. There were no significant effects of grazing on soil carbon neither in the whole profile nor in the uppermost 20 cm soil depth in the two study grasslands. Studies on the effects of varying rangeland management, such as region disparity and grazing systems, may have important consequences on species diversity and carbon partitioning, and thus on rangeland stability and ecosystem functioning.  相似文献

The Notch pathway contributes to self-renewal of tumor-initiating cell and inhibition of normal colonic epithelial cell differentiation. Deregulated expression of Notch1 and Jagged1 is observed in colorectal cancer. Hairy/enhancer of split (HES) family, the most characterized targets of Notch, involved in the development of many cancers. In this study, we explored the role of Hes1 in the tumorigenesis of colorectal cancer. Knocking down Hes1 induced CRC cell senescence and decreased the invasion ability, whereas over-expression of Hes1 increased STAT3 phosphorylation activity and up-regulated MMP14 protein level. We further explored the expression of Hes1 in human colorectal cancer and found high Hes1 mRNA expression is associated with poor prognosis in CRC patients. These findings suggest that Hes1 regulates the invasion ability through the STAT3-MMP14 pathway in CRC cells and high Hes1 expression is a predictor of poor prognosis of CRC.  相似文献

Information on steroid hormones and fatty acids that play roles in lobster reproduction is still very limited although the data are indispensable to seed production in hatchery. The study was designed to identify steroid hormones and fatty acids during gonadal maturation of spiny lobster (Panulirus homarus). Male spiny lobsters were treated with and without thyroxine injection. Female spiny lobsters were treated with and without eyestalk ablations during mature and immature gonad developments. Androst-5-en-17-one,3β (androst) and estran-3-one,17β (estran), two steroid hormones were identified at different levels of gonadal maturity of spiny lobsters. High concentrations of androst and estran were detected in the male spiny lobsters treated with thyroxine injections. Estran showed high concentrations in female brood stock of spiny lobsters during oogenesis stages both without eyestalk ablation and with ablation of one or two eyestalks, except in the immature female gonads. It was found that stearic acid was the highest and dominant fatty acid in mature male spiny lobster. Stearic acid, oleic acid, palmitic acid and caprylic acid were fatty acids with high concentrations in immature and mature female spiny lobsters. After 30 days in captivity, only stearic acid and oleic acid were found dominantly in eyestalk ablated mature female spiny lobsters.  相似文献
