Impact of land‐use change to Jatropha bioenergy plantations on biomass and soil carbon stocks: a field study in Mali 下载免费PDF全文

Small‐scale Jatropha cultivation and biodiesel production have the potential of contributing to local development, energy security, and greenhouse gas (GHG) mitigation. In recent years however, the GHG mitigation potential of biofuel crops is heavily disputed due to the occurrence of a carbon debt, caused by CO2 emissions from biomass and soil after land‐use change (LUC). Most published carbon footprint studies of Jatropha report modeled results based on a very limited database. In particular, little empirical data exist on the effects of Jatropha on biomass and soil C stocks. In this study, we used field data to quantify these C pools in three land uses in Mali, that is, Jatropha plantations, annual cropland, and fallow land, to estimate both the Jatropha C debt and its C sequestration potential. Four‐year‐old Jatropha plantations hold on average 2.3 Mg C ha?1 in their above‐ and belowground woody biomass, which is considerably lower compared to results from other regions. This can be explained by the adverse growing conditions and poor local management. No significant soil organic carbon (SOC) sequestration could be demonstrated after 4 years of cultivation. While the conversion of cropland to Jatropha does not entail significant C losses, the replacement of fallow land results in an average C debt of 34.7 Mg C ha?1, mainly caused by biomass removal (73%). Retaining native savannah woodland trees on the field during LUC and improved crop management focusing on SOC conservation can play an important role in reducing Jatropha's C debt. Although planting Jatropha on degraded, carbon‐poor cropland results in a limited C debt, the low biomass production, and seed yield attained on these lands reduce Jatropha's potential to sequester C and replace fossil fuels. Therefore, future research should mainly focus on increasing Jatropha's crop productivity in these degraded lands. 相似文献

muscle carnosine content measured within a population can vary by a factor 4. The aim of this study was to further characterize
suggested determinants of the muscle carnosine content (diet, gender and age) and to identify new determinants (plasma carnosinase
activity and testosterone). We investigated a group of 149 healthy subjects, which consisted of 94 men (12 vegetarians) and
55 women. Muscle carnosine was quantified in M. soleus, gastrocnemius and tibialis anterior using magnetic resonance proton
spectroscopy and blood samples were collected to determine CNDP1 genotype, plasma carnosinase activity and testosterone concentrations. Compared to women, men have 36, 28 and 82% higher
carnosine concentrations in M. soleus, gastrocnemius and tibialis anterior muscle, respectively, whereas circulating testosterone
concentrations were unrelated to muscle carnosine levels in healthy men. The carnosine content of the M. soleus is negatively
related to the subjects’ age. Vegetarians have a lower carnosine content of 26% in gastrocnemius compared to omnivores. In
contrast, there is no difference in muscle carnosine content between omnivores with a high or low ingestion of β-alanine.
Muscle carnosine levels are not related to the polymorphism of the CNDP1 gene or to the enzymatic activity of the plasma carnosinase. In conclusion, neither CNDP1 genotype nor the normal variation in circulating testosterone levels affects the muscular carnosine content, whereas vegetarianism,

Carnosine is present in high concentrations in skeletal muscle where it contributes to acid buffering and functions also as a natural protector against oxidative and carbonyl stress. Animal studies have shown an anti-diabetic effect of carnosine supplementation. High carnosinase activity, the carnosine degrading enzyme in serum, is a risk factor for diabetic complications in humans. The aim of the present study was to compare the muscle carnosine concentration in diabetic subjects to the level in non-diabetics. Type 1 and 2 diabetic patients and matched healthy controls (total n=58) were included in the study. Muscle carnosine content was evaluated by proton magnetic resonance spectroscopy (3 Tesla) in soleus and gastrocnemius. Significantly lower carnosine content (-45%) in gastrocnemius muscle, but not in soleus, was shown in type 2 diabetic patients compared with controls. No differences were observed in type 1 diabetic patients. Type II diabetic patients display a reduced muscular carnosine content. A reduction in muscle carnosine concentration may be partially associated with defective mechanisms against oxidative, glycative and carbonyl stress in muscle. 相似文献

The inevitable switch from standard molecular methods to next-generation sequencing for the molecular profiling of tumors is challenging for most diagnostic laboratories. However, fixed validation criteria for diagnostic accreditation are not in place because of the great variability in methods and aims. Here, we describe the validation of a custom panel of hotspots in 24 genes for the detection of somatic mutations in non-small cell lung carcinoma, colorectal carcinoma and malignant melanoma starting from FFPE sections, using 14, 36 and 5 cases, respectively. The targeted hotspots were selected for their present or future clinical relevance in solid tumor types. The target regions were enriched with the TruSeq approach starting from limited amounts of DNA. Cost effective sequencing of 12 pooled libraries was done using a micro flow cell on the MiSeq and subsequent data analysis with MiSeqReporter and VariantStudio. The entire workflow was diagnostically validated showing a robust performance with maximal sensitivity and specificity using as thresholds a variant allele frequency >5% and a minimal amplicon coverage of 300. We implemented this method through the analysis of 150 routine diagnostic samples and identified clinically relevant mutations in 16 genes including KRAS (32%), TP53 (32%), BRAF (12%), APC (11%), EGFR (8%) and NRAS (5%). Importantly, the highest success rate was obtained when using also the low quality DNA samples. In conclusion, we provide a workflow for the validation of targeted NGS by a custom-designed pan-solid tumor panel in a molecular diagnostic lab and demonstrate its robustness in a clinical setting. 相似文献

Purpose
The main aim of the study is to assess the environmental and economic impacts of the lodging sector located in the Himalayan region of Nepal, from a life cycle perspective. The assessment should support decision making in technology and material selection for minimal environmental and economic burden in future construction projects.Methods
The study consists of the life cycle assessment and life cycle costing of lodging in three building types: traditional, semi-modern and modern. The life cycle stages under analysis include raw material acquisition, manufacturing, construction, use, maintenance and material replacement. The study includes a sensitivity analysis focusing on the lifespan of buildings, occupancy rate and discount and inflation rates. The functional unit was formulated as the ‘Lodging of one additional guest per night’, and the time horizon is 50 years of building lifespan. Both primary and secondary data were used in the life cycle inventory.Results and discussion
The modern building has the highest global warming potential (kg CO2-eq) as well as higher costs over 50 years of building lifespan. The results show that the use stage is responsible for the largest share of environmental impacts and costs, which are related to energy use for different household activities. The use of commercial materials in the modern building, which have to be transported mostly from the capital in the buildings, makes the higher GWP in the construction and replacement stages. Furthermore, a breakdown of the building components shows that the roof and wall of the building are the largest contributors to the production-related environmental impact.Conclusions
The findings suggest that the main improvement opportunities in the lodging sector lie in the reduction of impacts on the use stage and in the choice of materials for wall and roof.6.

This study reports on the carbon, water, and energy footprints of tomatoes grown in a greenhouse in Northern Italy and two possible future variations of heating and carbon dioxide (CO2) fertilization on the current setup. The heat supply in place, consisting of natural gas (NG) and canola oil combustion, is compared to cogeneration and incineration of municipal solid waste for heating and CO2 from industrial exhaust for fertilization. As a benchmark, the current system is also compared to a conventional system, in which heat is delivered solely based on NG. Each kilogram (kg) of fresh tomatoes (“Cuore di Bue” variety) produced in the current greenhouse emits 2.28 kg CO2 equivalents (eq) and uses 95.5 megajoules (MJ) eq energy and 122 liters (L) of water. Relative to the system in place, the carbon footprint (CF) is 57.5% and 18% higher with conventional NG heating and cogeneration and is 40% lower with waste valorization. Further, 33%, 55%, and 63% less energy and 9%, 96%, and 14% less water are used in the conventional, cogeneration, and waste valorization scenarios, respectively. This confirms that there are multiple strategies to reduce the impact of the tomato production under consideration. 相似文献

Patterns of DNA methylation in complex genomes like those of mammalian cells have been viewed as indicators of different levels of genetic activities. It is as yet unknown how these complicated patterns are generated and maintained during cell replication. There is evidence from many different biological systems that the sequence-specific methylation of promoters in higher eukaryotes is one of the important factors in controlling gene activity at a long-term level. In general, the fifth nucleotide 5-methyldeoxycytidine can be considered as a modulator of protein-DNA interactions. The degree and direction of this modulation has to be assessed experimentally in each individual instance. The establishment of de novo patterns of DNA methylation is characterized by the gradual non-random spreading of DNA methylation by an essentially unknown mechanism. In this review, some of the general concepts of DNA methylation in mammalian systems are presented, and research currently performed in the authors' laboratory has been summarized. 相似文献

Carnosine (beta-alanyl-l-histidine) is present in high concentrations in human skeletal muscle. The ingestion of beta-alanine, the rate-limiting precursor of carnosine, has been shown to elevate the muscle carnosine content. We aimed to investigate, using proton magnetic resonance spectroscopy (proton MRS), whether oral supplementation with beta-alanine during 4 wk would elevate the calf muscle carnosine content and affect exercise performance in 400-m sprint-trained competitive athletes. Fifteen male athletes participated in a placebo-controlled, double-blind study and were supplemented orally for 4 wk with either 4.8 g/day beta-alanine or placebo. Muscle carnosine concentration was quantified in soleus and gastrocnemius by proton MRS. Performance was evaluated by isokinetic testing during five bouts of 30 maximal voluntary knee extensions, by endurance during isometric contraction at 45% maximal voluntary contraction, and by the indoor 400-m running time. beta-Alanine supplementation significantly increased the carnosine content in both the soleus (+47%) and gastrocnemius (+37%). In placebo, carnosine remained stable in soleus, while a small and significant increase of +16% occurred in gastrocnemius. Dynamic knee extension torque during the fourth and fifth bout was significantly improved with beta-alanine but not with placebo. Isometric endurance and 400-m race time were not affected by treatment. In conclusion, 1) proton MRS can be used to noninvasively quantify human muscle carnosine content; 2) muscle carnosine is increased by oral beta-alanine supplementation in sprint-trained athletes; 3) carnosine loading slightly but significantly attenuated fatigue in repeated bouts of exhaustive dynamic contractions; and 4) the increase in muscle carnosine did not improve isometric endurance or 400-m race time. 相似文献

Due to higher oil prices, abundance of labor and suitable land and its stable political climate, Tanzania attracted many investments in Jatropha. Although several studies on Jatropha's economic potential are available, its true economics are still uncertain. This paper aims to add to the growing body of knowledge on the socio‐economic performance of the Jatropha system by (i) studying the economic potential (net present value – NPV) of the current most prevailing Jatropha system for Tanzanian farmers and its regional differences, by (ii) making a greenhouse gas (GHG) balance and its economic value of the Jatropha activities on regional level, and by (iii) calculating break‐even thresholds for yield and seed price. Therefore, regional yield modeling, regional life‐cycle assessment, and NPV calculations based on Monte Carlo simulations, each with its set of assumptions, are combined. This study shows positive economic potential of Jatropha cultivation in most of the Tanzanian regions. However, the results also show that 13 of 20 Tanzanian regions will not attain a net positive GHG balance within 10 years. This indicates that the environmental impacts might be more restrictive for Jatropha's sustainability potential in Tanzania than the socio‐economic potential. These results are based on the combination of three models, which consists of strong interdisciplinary modeling work. However, this modeling also contains simplifications (e.g., no opportunity cost for ‘marginal’ land) and uncertainties (e.g., using globally modeled potential yield estimations), which have to be considered in the interpretation of the results. 相似文献
