The in vivo nitrate reductase activity in leaf tissue of cotton (Gossypium hirsutum L.) was characterized. Enzymatic activity was linear with time up to 60 min. The assay for nitrate reductase activity was optimized in leaf slices 400 μm wide incubated in an anaerobic system at 30°C, in a 0.02 M KNO3 medium at pH 7.0 with 1 % propanol. In vivo activity was highest in recently matured leaves at the top of the plant. Both light and nitrate enhanced in vivo enzymatic activity. The activity was highest after 9 hours in the light and then decreased steadily for several more hours even in the presence of light. The nitrate reductase activity was more strongly correlated to the levels of NO3-N in the culture solution than to the NO3-N level in the tissue. The utility of this technique in nitrate reductase assay in a tissue containing large amounts of phenolic compounds is discussed.  相似文献

Abstract A new subgenus and species, Glyptomorpha (Zanporiu) egyptiaca from Egypt, Arabia, Yemen and Pakistan are described and illustrated. G. Zanporia differs from G. Glyptomorpha in the form of the scapus and the lengh of the ovipositor. The status of Teraturus Kokujev is reconsidered and it is proposed that it should be treated as a subgenus of Glyptomorpha Holmgren, The relationship of Glyptomorpha to Rhadinobracon Szépligeti and Victoroviella Tobias is also discussed.  相似文献

Abstract: Two new primitive macroscelideans, Eotmantsoius perseverans gen. et sp. nov. and Nementchatherium rathbuni sp. nov. from the late Middle or Late Eocene locality of Dur At‐Talah, Libya, are described. Eotmantsoius is known from a single tooth and is characterized by low crown height, isolated conules, and lack of a preprotocrista and prehypocrista. Based on these characters, it appears to be more plesiomorphic than Chambius (late Early Eocene, Chambi, Tunisia), which was, to date, universally accepted as the oldest and most basal macroscelidean. Eotmantsoius appears, however, somewhat derived in having an entostyle, a pericone, a shorter mesial cingulum and larger intercusp valleys. The phylogenetic position of Eotmantsoius is unclear. The second new species, N. rathbuni, allows a reassessment of Nementchatherium. We suggest that it is the most derived of the herodotiines in having long preprotocrista and prehypocrista, reduced M2/m2, a robust entostyle twinned with the hypocone on M1 and the posterior aspect of the root of the maxillary jugal process placed along the length of M2. Nementchatherium rathbuni appears more derived than N. senarhense, from the Eocene locality of Bir El Ater, Algeria, in having smaller m2‐3 with no paraconid, a smaller hypoconulid on m2 and a more reduced talonid on m3 (with smaller hypoconid and entoconid). Nementchatherium rathbuni also has, distal to the m3, a coronoid canal. This character has commonly been proposed as a synapomorphy for paenungulates (e.g. proboscideans, sirenians and hyracoids); we investigate the possibility that it unites macroscelideans and paenungulates, as the superordinal clade Afrotheria suggests. Comparisons reveal that a coronoid canal also occurs in several nonafrotherian orders (lagomorphs, xenarthrans, artiodactyls and perissodactyls), suggesting that is not an exclusive synapomorphy for paenungulates plus macroscelideans. Moreover, the homology of the character within paenungulates is doubtful.  相似文献

Using NaCl or polyethylene glycol (PEG) solutions to progressively decrease the external osmotic potential of the peat casing of the growing medium used to culture the mushroom Agaricus bisporus resulted in proportionately decreased yields of sporophores. Over the range of -0.07 to -0.37 MPa, the extent of decrease in yield was similar with both types of osmoticum. However, with further decrease in external osmotic potential (from -0.37 to -0.62 MPa) there was a further proportional decrease in sporophore yield with PEG but a complete suppression of sporophore production with NaCl. Treatments with both NaCl and PEG decreased the concentrations of P, Mg, K, Fe and Mn, but not N and Cu, in sporophore dry matter. Treatment with NaCl solutions increased the concentrations of Na and CI ions in sporophore dry matter and decreased the concentration of Ca; PEG solutions had no effect. Ion toxicity associated with excessive accumulation of Na and C1 ions, or ionic imbalance associated with the concomittant decrease in Ca ions appear to be additional factors to osmotic stress in decreasing yield of sporophores when the growing medium becomes highly saline. The critical concentration of NaCl which caused 10% reduction in sporophore yield was 28 mM; A. bisporus is, therefore, moderately salt-sensitive.  相似文献
