Embryonic germ (EG) cells of line EG-1 derived from mouse primordial germ cells were investigated for theirin vitrodifferentiation capacity. By cultivation as embryo-like aggregates EG-1 cells differentiated into cardiac, skeletal muscle and neuronal cells accompanied by the expression of tissue-specific genes and proteins as shown by RT-PCR analysis and indirect immunofluorescence. In comparison to embryonic stem (ES) cells of line D3 the efficiency of differentiation into cardiac and muscle cells was comparatively low, whereas spontaneous neuronal differentiation was more efficient than in D3 cells. Furthermore, the distribution of cell cycle phases as a parameter for the differentiation state was analysed in undifferentiated EG cells and ES cells and compared to data obtained for embryonic carcinoma (EC) cells of line P19 and differentiated, epithelioid EPI-7 cells. Flow cytometric analysis revealed similar cell cycle phase distributions in EG, EC and ES cells. In contrast, the somatic differentiated EPI-7 cells showed a longer G1-phase and shorter S- and G2/M-phases. Together, our results demonstrate that the differentiation state and capacity of EG cellsin vitroresemble that of totipotent ES cells.  相似文献

Elaborate horns or horn‐like structures in male scarab beetles commonly scale with body size either (a) in a linear fashion with horn size increasing relatively faster than body size or (b) in a threshold‐dependent, sigmoid fashion; that is, males smaller than a certain critical body size develop no or only rudimentary horns, whereas males larger than the threshold size express fully developed horns. The development of linear vs. sigmoid scaling relationships is thought to require fundamentally different regulatory mechanisms. Here we show that such disparate regulatory mechanisms may co‐occur in the same individual. Large males of the south‐east Asian Onthophagus (Proagoderus) watanabei (Ochi & Kon) (Scarabaeidae, Onthophagini) develop a pair of long, curved head horns as well as a single thoracic horn. We show that unlike paired head horns in a large number of Onthophagus species, in O. watanabei the relationship between head horns and body size is best explained by a linear model. Large males develop disproportionately longer horns than small males, but the difference in relative horn sizes across the range of body sizes is small compared to other Onthophagus species. However, the scaling relationship between the thoracic horn and body size is best explained by a strongly sigmoid model. Only males above a certain body size threshold express a thoracic horn and males smaller than this threshold express no horn at all. We found a significant positive correlation between head and thoracic horn length residuals, contrary to what would be expected if a resource allocation tradeoff during larval development would influence the length of both horn types. Our results suggest that the scaling relationship between body size and horn length, and the developmental regulation underlying these scaling relationships, may be quite different for different horns, even though these horns may develop in the same individual. ABSTRACT

The mechanism of sound production in tettigoniids is examined by applying the method of ‘cepstrum’ analysis to insect calls. The power cepstrum is defined as the inverse Fourier transform of the logarithmic power spectrum. This analysis shows that the tettigoniid sound signal is a convolution in time of probably two components. The first is caused by the initial impact of teeth of the stridulatory file on the left wing against the plectrum on the right wing (termed the input pulse); the second is caused by the oscillating properties of the tegmina (these being a function of the intrinsic frequencies of dorsal fields and mirror and their damping properties). In the cepstrum each component appears as a varying number of peaks. The tooth impacts cause a very low quefrency peak probably representing the time in which the two tegmina are in contact during each impact and high quefrency peaks representing the impulse repetition rate. The oscillating properties of the tegmina cause two major quefrency peaks which can be clearly related to the size of the dorsal fields and of the mirror respectively, and therefore to their intrinsic frequencies. The high damping factor of the tegmina together with the transient shape of the tegminal input pulse causes a strong time limitation of the impulses and is therefore responsible for the broad frequency bands occurring in the power spectra of the tettigoniid songs. The impulse generation of a synthetic tettigoniid song is discussed.  相似文献

The most conspicuous and species-rich ant-plant mutualism in the Malesian region is found in the important pioneer tree genus Macaranga , yet little is known about the identities or community ecology of the species involved. Our studies have revealed a far more complex system than previously thought. This paper presents the first extensive investigation in the whole distribution area of myrmecophytic Macaranga. All ant-inhabited species were restricted to the moister parts of SE Asia: Peninsular Malaysia, South and East Thailand, Sumatra and Borneo. We found a rather strict and similar altitudinal zonation of myrmecophytic Macaranga species in all regions. Here we focus on the majority of the 19 Macaranga species obligatorily associated with ants of the genus Crematogaster. We identified a total of 2163 ant queens which belonged to at least eight (morpho)species of the small subgenus Decacrema as well as to one non-Decacrema (probably from Atopogyne ). The ant species were not randomly distributed among the Macaranga species but distinct patterns of associations emerged. Despite common sympatric distribution of Macaranga species, in most cases a surprisingly high specificity of ant colonization was maintained which was, however, often not species-specific but groups of certain plant species with identical ant partners could be found. These colonization patterns usually but not always mirror existing taxonomic sections within the genus Macaranga. Possible mechanisms of specificity are discussed. The results are compared with other ant-plant mutualisms.  相似文献

A new group of darkly pigmented root-infecting fungi was isolated from cereal roots obtained from six different locations in northeastern Germany. Similar random amplified polymorphic DNA(RAPD) patterns and restriction profiles of amplified rDNA were used as a basis for classifying the isolates in a separate group. The isolates demonstrating mycelial and infection characteristics typical of Gaeumannomyces graminis could be differentiated from the varieties of G. graminis as well as from Gaeumannomyces cylindrosporus / Phialophora graminicola using RAPD Polymerase chain reaction (PCR) and rDNA Restriction-fragment length polymorphism (RFLP) analysis. Phylogenetic analysis of the Internal transcribed spacer (ITS) regions suggests that the isolates form a distinct group (named group 'E') situated within the Gaeumannomyces – Phialophora complex between the branch of the G. graminis varieties and Gaeumannomyces incrustans / Magnaporthe poae . Isolates of group E produced lobed hyphopodia and were shown in biotests to be non-pathogenic to wheat, oats, Italian Ryegrass and Chewings Fescue, suggesting it is a benign parasite which colonizes cereals or grasses without destroying vascular tissue. Furthermore, curved phialospores could be found. Summarizing the results presented, this new group could be classified as a new species of Phialophora . Although isolates of group E were found at only six of the 32 investigated locations, they composed up to 50% of total isolates of the Gaeumannomyces – Phialophora complex at these sites. Because of the non-pathogenic behaviour, the new group may be of value as biological control agents for pathogenic fungi.  相似文献

Abstract

Comparative studies of sound production and sound emission in seven species of European tettigoniids have been carried out. The species chosen were two Tettigoniines (Tettigonia cantans, Tettigonia viridissima), two Ephippigerines (Ephippiger discoidalis, Ephippiger ephippiger), and three Decticines (Decticus albifrons, Decticus verrucivorus, Psorodonotus illyricus). The factors which determined the choice of species were the different morphology (for example body shape and weight, and wing size) of the three subfamilies. The parameters of the different songs (e.g. dominant frequency, intensity) are normally not correlated to any of the investigated morphological characteristics of the animals. In the brachypterous species intraspecific correlations exist between wing size and the dominant low frequency band of the call. This frequency band is also observable at related higher frequencies in the ultrasonic range (20–60 kHz), the observed band width increasing with frequency. Sound emission in all species is to some extent directional. This directionality is related to body size and wing structure. The song structure of the different species does not appear to be related to any observable characteristic of the habitat of die animals. A possible exception may be the song of Psorodonotus illyricus with a particularly low dominant frequency band. The phylogenetic development of the songs seems to be determined by relationships between the different species rather than to any factors contributed by the habitat.  相似文献

Aromatic amines are important intermediates in industrial manufacturing. They are used in a large number of products, such as pesticides, dyes, plastics and pharmaceuticals. The parent arylamines can be metabolically released from these arylamine-based compounds and form DNA and protein adducts after N-oxidation to N-hydroxy arylamines. Aromatic amine derivatives, including the industrial intermediates acetoacetanilide, acetoacet-m-xylidide and N-ethylaniline, were examined for their ability to form Hb adducts in rats as potential biomarkers of exposure. The haemoglobin binding indices (HBI=binding [mmol mol-1 Hb]/dose [mmol kg-1 body weight]) of the arylamines were determined 24 h after oral administration to female Wistar rats. The precipitated haemoglobin was dissolved in 0.1 M sodium hydroxide in the presence of internal standards. After hexane extraction the released arylamines were analysed by gas chromatography-mass spectrometry (GC-MS). For aniline released from acetoacetanilide an HBI of 15 and for 2,4-dimethylaniline released from acetoacet-m-xylidide an HBI of 0.129 were determined. 1. Using an extensive data set from 18 river restoration projects in the lower mountain ranges of Germany and 5462 river reaches in their surroundings, we estimated the spatial extent of the regional fish species pool from which restored river reaches are colonised. 2. Restoration resulted in a marginally significant increase in fish species richness; however, restored reaches still deviated markedly from natural reference conditions. Nearly all (96.6%) species occurring in restored reaches were present in reaches within a distance of 5 km up‐ or downstream of the restored reach. 3. Species richness in restored reaches was correlated with species richness within a 5‐km species pool. This relationship was more pronounced for common than for rare fishes and applied to both the total number of fish species at the restored reach and the number of additional fish species that were not present at unrestored conditions. 4. The richness of the regional species pools was greatly impoverished. On average, only 50% of all species considered to represent natural reference assemblages were present. The limited success in establishing natural fish assemblages in restored reaches was attributed to spatial limitation (e.g. due to fragmentation) and an impoverished regional species pools from which restored reaches recruit. 5. We recommend that integrated river restoration management should consider not only the abiotic prerequisites of successful restorations, but also the structure and quality of the regional species pool.  相似文献
