Benzothiadiazole (BTH) induces resistance to the downy mildew pathogen, Peronospora sparsa, in arctic bramble, but the basis for the BTH‐induced resistance is unknown. Arctic bramble cv. Mespi was treated with BTH to study the changes in leaf proteome and to identify proteins with a putative role in disease resistance. First, BTH induced strong expression of one PR‐1 protein isoform, which was also induced by salicylic acid (SA). The PR‐1 was responsive to BTH and exogenous SA despite a high endogenous SA content (20–25 µg/g fresh weight), which increased to an even higher level after treatment with BTH. Secondly, a total of 792 protein spots were detected in two‐dimensional gel electrophoresis, eight proteins being detected solely in the BTH‐treated plants. BTH caused up‐ or down‐regulation of 72 and 31 proteins, respectively, of which 18 were tentatively identified by mass spectrometry. The up‐regulation of flavanone‐3‐hydroxylase, alanine aminotransferase, 1‐aminocyclopropane‐1‐carboxylate oxidase, PR‐1 and PR‐10 proteins may partly explain the BTH‐induced resistance against P. sparsa. Other proteins with changes in intensity appear to be involved in, for example, energy metabolism and protein processing. The decline in ATP synthase, triosephosphate isomerase, fructose bisphosphate aldolase and glutamine synthetase suggests that BTH causes significant changes in primary metabolism, which provides one possible explanation for the decreased vegetative growth of foliage and rhizome observed in BTH‐treated plants.  相似文献

Between 1988 and 2007, set‐aside, a European Commission production control measure, took an average of 10% of arable farmland in the EU out of production each year. In 2007, the set‐aside rate was set to 0% and the scheme was later abandoned altogether. By assessing associations of farmland birds with set‐aside and quantifying the extent of set‐aside loss, this study aims to assess the implications of set‐aside loss for farmland bird conservation. During the lifespan of set‐aside, a large number of studies assessed the biodiversity value of set‐aside and other agricultural crops and habitats. Where possible we considered measurable benefits of set‐aside. However, some studies did not specify the type of set‐aside and in some cases set‐aside fields were grouped with cereal stubble fields. In these cases, we took the pragmatic approach of assessing the value of generic stubble fields as a conservative minimum estimate of the value of set‐aside fields. A re‐analysis of data from 30 intensive studies demonstrates that farmland bird densities tended to be higher on set‐aside than on either cereal or oilseed rape crops. Without mitigation, these are the two crops likely to replace most set‐aside fields. We estimate that 26–52% of the farmland populations of key granivorous passerines were present on stubble fields, giving an indication of the proportion of birds likely to be present on set‐aside fields within this broader category. An extensive survey of lowland farmland during winters 1999/2000, 2000/2001 and 2002/2003, repeated in February 2008, showed a doubling of the number of 1‐km squares with no stubble and a halving of the number of squares with more than 10 ha of stubble. After set‐aside abandonment, 72% of squares had no stubble in the important late winter period, confirming that many of the former stubble fields were retained as set‐aside. A simple correlative model suggests that this could cause a small increase in the rate of decline of Skylark Alauda arvensis and Yellowhammer Emberiza citrinella populations, assuming causal links between stubble area and demography. However, even if this assumption cannot be supported, these results clearly indicate that a significant proportion of some farmland bird populations will need to find alternative breeding and foraging habitats.  相似文献

Evidence for the indirect effects of pesticides on farmland birds   总被引:4,自引:0,他引:4  
Indirect effects of pesticides, operating through the food chain, have been proposed as a possible causal factor in the decline of farmland bird species. To demonstrate such a link, evidence is needed of (1) an effect of food abundance on breeding performance or survival; (2) an effect of breeding performance or survival on population change; and (3) pesticide effects on food resources, sufficient to reduce breeding performance or survival, and hence to affect the rate of population change. Evidence under all three categories is only available for one species, the Grey Partridge Perdix perdix , although data showing effects of pesticides on food resources and relationships between food resources and breeding performance are also available for some other species. This paper reports on recent work investigating the effects of pesticides on Yellowhammer Emberiza citrinella and Skylark Alauda arvensis during the breeding season. The probability of brood reduction in Yellowhammer was affected by the proportion of the foraging area around the nest which was sprayed with insecticide. No significant effects of pesticides were recorded on Skylark chick condition or growth rate, but sample sizes were small. Invertebrate food abundance affected chick condition (Skylark) and the number of chicks fledging (Yellowhammer and Corn Bunting Miliaria calandra ; relationship for the latter derived from re-analysis of data from an earlier study). Other recent work is briefly reviewed and the current evidence for the indirect effects of pesticides is summarized. Significant knowledge gaps are identified and some of the issues involved in resolving these are discussed.  相似文献

Geographic variation in body size and sexual dimorphism of the short‐nosed fruit bat (Cynopterus sphinx) was investigated in peninsular India. Bats were sampled at 12 localities along a 1200 km latitudinal transect that paralleled the eastern flanks of the Western Ghats. The geographic pattern of variation in external morphology of C. sphinx conforms to the predictions of Bergmann's Rule, as indicated by a steep, monotonic cline of increasing body size from south to north. This study represents one of the first conclusively documented examples of Bergmann's Rule in a tropical mammal and confirms that latitudinal clines in body size are not exclusively restricted to temperate zone homeotherms. Body size was indexed by a multivariate axis derived from principal components analysis of linear measurements that summarize body and wing dimensions. Additionally, length of forearm was used as a univariate index of structural size to examine geographic variation in a more inclusive sample of bats across the latitudinal transect. Multivariate and univariate size metrics were strongly and positively correlated with body mass, and exhibited highly concordant patterns of clinal variation. Stepwise multiple regression on climatological variables revealed that increasing size of male and female C. sphinx was associated with decreasing minimum temperature, increasing relative humidity, and increasing seasonality. Although patterns of geographic size variation were highly concordant between the sexes, C. sphinx also exhibited a latitudinal cline in the magnitude and direction of sexual size dimorphism. The size differential reversed direction across the latitudinal gradient, as males averaged larger in the north, and females averaged larger in the south. The degree of female‐biased size dimorphism across the transect was negatively correlated with body size of both sexes. Canonical discriminant analysis revealed that male‐ and female‐biased size dimorphism were based on contrasting sets of external characters. Available data on geographic variation in the degree of polygyny in C. sphinx suggests that sexual selection on male size may play a role in determining the geographic pattern of sexual size dimorphism.  相似文献

ANTIBODY production to many antigens including heterologous erythrocytes1–3, serum proteins4,5 and hapten protein conjugates6,7, occurs as a result of an interaction between antigen and thymus-derived (T) and non-thymus-derived (B) lymphocytes. Although the specificity of the antibody response is determined by T as well as B cells8–10, T cells do not actively secrete any of the known classes of immunoglobulin molecules11. ABSTRACT. Leishmania tropica promastigotes transport α-aminoisobutyric acid (AIB), the nonmetabolizable analog of neutral amino acids, against a substantial concentration gradient. AIB is not incorporated into cellular material but accumulates within the cells in an unaltered form. Intracellular AIB exchanges with external AIB. Various energy inhibitors (amytal, HOQNO, KCN, DNP, CCCP, and arsenate) and sulfhydryl reagents (NEM, pCMB, and iodoacetate) severely inhibit uptake. The uptake system is saturable with reference to AIB-and the Lineweaver-Burk plots show biphasic kinetics suggesting the involvement of two transport systems. AIB shares a common transport system with alanine, cysteine, glycine, methionine, serine, and proline. Uptake is regulated by feedback inhibition and transinhibition.  相似文献

SYNOPSIS. The glucose transport system in Leishmania tropica promastigotes was characterized by the use of labeled 2-deoxy-D-glucose (2-DOG), a nonmetabolizable glucose analog. The uptake system has a Q10 of 2 and a heat of activation of 10.2 kcal/mole. The glucose transport system is subject to competitive inhibition by 2-DOG, glucosamine, N-acetyl glucosamine, mannose, galactose, and fructose which suggests that substitutions in the hexose chain at carbons 2 and 4 do not affect carrier specificity. In contrast, changes at carbon 1 (α-methyl-D-glucoside, 1,5-anhydroglucitol) and carbon 3 (3–0-methyl glucose) lead to loss of carrier affinity since these sugars do not compete for the glucose carrier. Sugars that compete with the glucose carrier have one common feature—they all exist in the pyranose form in solution. The carrier for D-glucose does not interact with L-glucose or any of the pentose sugars tested. Uptake of 2-DOG is inhibited by glycerol. This inhibition, however, is noncompetitive; it is evident, therefore, that glucose and glycerol do not compete for the same carrier. Glycerol does not repress the glucose carrier since cells grown in presence of glycerol transport the sugar normally.  相似文献

Close to the edge: predation risks for two declining farmland passerines   总被引:1,自引:1,他引:0  
Data on the breeding success of two crop-nesting passerines, Skylark Alauda arvensis and Yellow Wagtail Motacilla flava , were collected in relation to linear features within and surrounding arable crops. Both species were found to experience high rates of nest predation with increased proximity to field boundaries, although the exact nature of the relationship differed with species and, in the case of Skylark, with boundary type. Most nest losses were attributable to predation. During 2006 video cameras deployed on Skylark nests showed that all recorded predation was by mammals of various species, and that these were most active in or around grass margins. The results suggest that further research is needed into ways of minimizing negative impacts of predation on Skylarks. Possible solutions discussed include concentrating Skylark Plots in the field centres away from grass margins and promoting Skylark Plots in fields without grass margins in future agri-environmental schemes.  相似文献
