Because toothpaste and other menthol/surfactant‐containing products have residual effects, researchers should ensure that enough time has elapsed between use of toothpaste and evaluation of food products. In this study, orange juice, coffee, sausage and processed cheese were profiled under controlled conditions and at 4, 12, 30 and 60 min after panelists brushed with a strongly mentholated toothpaste. Residual effects of the toothpaste at 4 min reduced several aromatic attributes, but did not affect most fundamental taste or texture attributes. A mint aromatic was observed in some food products after toothpaste use, even after 12 min. Chemesthetic attributes were diminished when a product was presented within 12 min of brushing. Results suggested that the amount of time needed between brushing and tasting depended on the food product. Orange juice, an acidic beverage, required at least an hour's delay; fatty, solid foods could be tasted almost immediately after toothbrushing if provided a warm‐up sample first.  相似文献

Drying of immature seeds of Ricinus communis L. cv. Hale (castorbean) during the desiccation-tolerant phase of development causesthem to germinate upon subsequent rehydration. This desiccation-inducedswitch from development to germination is also mirrored by achange in the pattern of soluble and insoluble protein synthesiswithin the cotyledons of the castor bean. Following rehydrationof seeds prematurely dried at 40 d after pollination (DAP),cotyledonary proteins characteristic of development (e.g. storageproteins) are no longer synthesized; hydrolytic processes resultingin their degradation commence (after 12 h) in a manner similarto that observed following imbibition of the mature seed. Apattern of protein synthesis recognizable as germination/growth-associatedoccurs; premature drying has elicited a redirection in metabolismfrom a developmental to a germinative mode. Desiccation is alsorequired for the induction (within cotyledons of 35 DAP seeds)of enzymes involved in protein reserve breakdown (leucyl ß-naphthylamidase;LeuNAase) and lipid utilization (isocitrate lyase; ICL), anevent intimately associated with the post-germinative (growth)phase of seedling development. Thus, at a desiccation-tolerantstage of development, premature drying results in the suppressionof the developmental metabolic programme and a permanent switching-onof the germination/growth metabolic programme. DURING replication of RNA tumour viruses, the genetic information contained in the viral RNA seems to be transferred to DNA1,2. Studies on the enzymatic activities present in the virus particles suggest that this transfer is mediated by an RNA dependent DNA polymerase3,4. RNA-DNA hybrids have been demonstrated to occur as intermediates in this reaction5 and single stranded DNA is generated as an early reaction product6, which is then replicated to give a double stranded DNA product6–8. A diagenetic case study of phosphatized soft-bodied squids from the lower part of the Oxford Clay of Christian Malford, Wiltshire, has dated mineralization in relation to burial history. Complete squids with mantle tissues were examined under the SEM as uncoated specimens at low KV. A simple stage attachment designed for this purpose is described. Soft-parts are preserved in three dimensions and must therefore have been mineralized prior to decay-induced tissue collapse. Such mineralization occurred near to the anoxic/oxic boundary in the 'soupy' upper layer of organic-rich sediment. In acrobic conditions, phosphates were liberated to pore-water solution from decomposing organic matter and became adsorbed to ferric hydroxides within sediment. The reduction of these ferric hydroxides at the anoxic-oxic interface liberated phosphates to solutions which were conccntrated in the decomposing carcasses of the squids. This mineral concentration was favoured by the development of a high pH reducing environment around the carcass and possibly by complexing with protcolipids liberated by decomposition of animal matter. ▭ Soft-part preservation, phosphatization, SEM techniques, cephatopod. Background

Although a preponderance of pre-clinical data demonstrates the immunosuppressive potential of mesenchymal stromal cells (MSCs), significant heterogeneity and lack of critical quality attributes (CQAs) based on immunosuppressive capacity likely have contributed to inconsistent clinical outcomes. This heterogeneity exists not only between MSC lots derived from different donors, tissues and manufacturing conditions, but also within a given MSC lot in the form of functional subpopulations. We therefore explored the potential of functionally relevant morphological profiling (FRMP) to identify morphological subpopulations predictive of the immunosuppressive capacity of MSCs derived from multiple donors, manufacturers and passages.

Methods

We profiled the single-cell morphological response of MSCs from different donors and passages to the functionally relevant inflammatory cytokine interferon (IFN)-γ. We used the machine learning approach visual stochastic neighbor embedding (viSNE) to identify distinct morphological subpopulations that could predict suppression of activated CD4+ and CD8+ T cells in a multiplexed quantitative assay.

Results

Multiple IFN-γ–stimulated subpopulations significantly correlated with the ability of MSCs to inhibit CD4+ and CD8+ T-cell activation and served as effective CQAs to predict the immunosuppressive capacity of additional manufactured MSC lots. We further characterized the emergence of morphological heterogeneity following IFN-γ stimulation, which provides a strategy for identifying functional subpopulations for future single-cell characterization and enrichment techniques.

Discussion

This work provides a generalizable analytical platform for assessing functional heterogeneity based on single-cell morphological responses that could be used to identify novel CQAs and inform cell manufacturing decisions.  相似文献

Freshwater mussels (Unionidae) are among North America's most imperilled organisms. Mussels produce small larvae (glochidia) that parasitize aquatic vertebrates. We modified the Epicentre QuickExtract protocol to extract DNA from a single glochidium, collected directly from the marsupium of a female mussel, to use as template in polymerase chain reactions (PCRs). Yield per glochidium in a 40 µL extraction volume provided enough DNA for ≥ 15 PCRs per individual. We were successful in using this DNA for microsatellite analysis of up to three loci per individual. Offspring from one female showed evidence for multiple paternity within her brood. Our results are the first documentation of this phenomenon in freshwater mussels.  相似文献
