Plant genetic resource conservation strategies, informed by an understanding of the geographical distribution of genetic variation within species, are likely to result in a wider representation of conserved diversity in ex situ gene banks and in situ genetic reserves. The main objective of this study was to map the geographical distribution of genetic variation, as revealed by randomly amplified polymorphic DNA (RAPDs), in four wild relatives of the cultivated lentil, namely Lens culinaris ssp. orientalis, L. odemensis, L. ervoides and L. nigricans . Areas of high diversity and unique diversity were located for each taxon, and regions where further germplasm collection was most likely to yield novel genetic variation were identified. There were centres of diversity for L. culinaris ssp. orientalis in southeast Turkey and northwest Syria, and in south Syria and Jordan. A centre of diversity was found to exist in Sweida province, south Syria, for L. odemensis , and for L. ervoides along the coastal border region between Syria and Turkey stretching down along the Syrian coast. There was a centre of diversity for L. nigricans in west Turkey. Analytical techniques previously used at the species level were found to be useful at the genotypic level to objectively target areas for future collection missions, to increase diversity in ex situ collections and to target areas for in situ conservation. 相似文献

Chloroplast thylakoid protein changes induced by low growth temperature in maize revealed by immunocytology 总被引:4,自引:3,他引:4
Tissue-specific effects of low growth temperature on maize chloroplast thylakoid protein accumulation were analysed using immunocytology. Sections of leaves from plants grown at 25 and 14°C were probed with antibodies to specific chloroplast thylakoid proteins from the four major protein multisubunit complexes of the thylakoid membrane followed by fluorescein-conjugated goat anti-rabbit antibodies. At a normal growth temperature of 25°C, the 32 kDa D1 protein of the photosystem II reaction centre and the 33 kDa protein of the extrinsic oxygen-evolving complex of photosystem II are both accumulated to a greater degree in the mesophyll than in the bundle sheath chloroplasts. In contrast, subunit II of photosystem I, cytochrome f and the α- and β-subunits of ATP synthetase are predominant in the bundle sheath thylakoids at 25°C. A striking difference between the 25°C-grown and the 14°C-grown leaf tissue was the presence in the latter of (20–30%) cells whose chloroplasts apparently completely lack several of the thylakoid proteins. In plants grown at 14°C, the accumulation of the 33 kDa protein of the extrinsic oxygen-evolving complex of photosystem II was apparently unchanged, but other thylakoid proteins showed a significant reduction. The uneven distribution of proteins between the bundle sheath and mesophyll chloroplasts observed at 25°C was also maintained at 14°C. Reduction in the fluorescence at 14°C was manifested either as an overall reduction in the diffuse fluorescence across the chloroplast profiles or less frequently as a reduction to small discrete bodies of intense fluorescence. Indirect effects of pesticides, operating through the food chain, have been proposed as a possible causal factor in the decline of farmland bird species. To demonstrate such a link, evidence is needed of (1) an effect of food abundance on breeding performance or survival; (2) an effect of breeding performance or survival on population change; and (3) pesticide effects on food resources, sufficient to reduce breeding performance or survival, and hence to affect the rate of population change. Evidence under all three categories is only available for one species, the Grey Partridge Perdix perdix , although data showing effects of pesticides on food resources and relationships between food resources and breeding performance are also available for some other species. This paper reports on recent work investigating the effects of pesticides on Yellowhammer Emberiza citrinella and Skylark Alauda arvensis during the breeding season. The probability of brood reduction in Yellowhammer was affected by the proportion of the foraging area around the nest which was sprayed with insecticide. No significant effects of pesticides were recorded on Skylark chick condition or growth rate, but sample sizes were small. Invertebrate food abundance affected chick condition (Skylark) and the number of chicks fledging (Yellowhammer and Corn Bunting Miliaria calandra ; relationship for the latter derived from re-analysis of data from an earlier study). Other recent work is briefly reviewed and the current evidence for the indirect effects of pesticides is summarized. SUMMARY. 1. Regeneration of ammonium and phosphate by macro-zooplankton (Cladocera. adult copepods. and copepodites) was measured in Lake Calado. an Amazon floodplain lake, Macrozooplanktonabundances ranged between 1×104 and 3×105 individuals m−2 .

Phosphate regeneration ranged from 0.2 to 1.3 μ mol PO4 m−2 b−1 at station 1. located 2 km from the Solimoes River, and from 1.6 to8.3 μ mol PO4 m−2 h −1 at station 3, located 7 km from the SolimoesRiver. Ammonium regeneration at stations 1 and 3 ranged from 1.7 to11.9 and from 13.4 to 77.2 μ mol NH4 m−2 h−1 . respectively.
