Progress has been made in the development of fixed, expanded and fluidized bed anaerobic processes by addressing fundamental reactor design issues. A convenient gas-liquid chromatography procedure to quantify poly-beta-hydroxybutyrate and poly-beta-hydroxyvalerate in activated sludge was developed by combining lyophilization of the samples, purification of the chloroform phase by water reextraction, and the use of capillary columns. The effects of dietary energy and suckling on adrenal function and luteinizing hormone (LH) concentrations were investigated in primiparous postpartum cows. Ten heifers were assigned at calving to either high (22.8 Mcal/day) or low (15.2 Mcal/day) energy diets. Blood samples were collected every 15 minutes for 8 hours on 28, 42, and 56 days post partum. Calves were allowed to suckle ad libitum during sampling periods. Serum samples were analyzed by radioimmunoassay for LH and cortisol. Concentrations of catecholamines were quantified by reverse-phase HPLC. Body weights were decreased (P<0.01) by low energy intake. In addition, low energy diet cows had lower mean LH concentrations (0.97 +/- 0.09 vs 1.57 +/- 0.07 ng/ml), P<0.05) than high energy diet cows. Luteinizing hormone concentrations in high energy diet cows increased with days post partum, resulting in a treatment-by-time interaction (P<0.005). Treatment did not affect mean cortisol concentrations. However, within 15 minutes of suckling cortisol release was significantly above baseline in 77% of the observed suckling events. Dihydroxyphenylalanine (DOPA) increased in high energy diet cows compared with that of low energy diet cows (2,833 +/- 243 vs 1,294 +/- 243 pg/ml, P<0.01). Norepinephrine (NE) and 3,4-dihydroxyphenylacetic acid (DOPAC) were not influenced by treatment. Plasma NE decreased during the postpartum interval (P<0.005). These data suggest that reduced energy intake may prevent the increase in LH associated with increasing days post partum and alter adrenal function. In addition, spontaneous suckling events elicit a release of cortisol.  相似文献

Many integral membrane proteins, particularly receptors on the cell surface, are made up of several polypeptide chains. After translation and insertion into the ER membrane, these subunits must assemble into the mature protein. However, the mechanisms controlling their faithful assembly are largely unknown. Recent evidence has shed some light on two cell surface receptors that use different strategies to assemble their subunits. Particulate cell wall polysaccharide synthetase from oat coleoptiles could use either guanosine diphosphate glucose or uridine diphosphate glucose; the latter was a much more effective glucose donor. The neutral polymer derived from uridine diphosphate glucose utilization yielded, after cellulase digestion, mostly cellobiose and to a lesser extent a substance tentatively identified as a mixed-linkage β1,4 = β1,3-trisaccharide; only cellobiose was found after guanosine diphosphate glucose utilization. The uridine diphosphate glucose utilizing system was inactivated by peroxyacetyl nitrate treatment of intact tissue and to a lesser extent by ozone treatment suggesting that this system is a possible site of interference with cellulose and non-cellulosic glucan biosynthesis in vivo. Direct treatment of the enzyme in vitro by peroxyacetyl nitrate, iodoacetamide or p-chloromercuribenzoate also inactivated the enzyme, indicating that the mechanism of inactivation possibly involves reaction with sulfhydryl groups.  相似文献

A biotype of Stellaria media (L.) Vill. has been identified that is highly resistant to the herbicide chlorsulfuron. Resistance is due to an altered acetolactate synthase (ALS) that is much less sensitive to chlorsulfuron than the ALS from the susceptible (S) biotype. The S biotype was extremely sensitive to D489 (N-[2,6-dichlorophenyl]-5,7-dimethyl-1,2,4-triazolo[1,5a] pyrimidine-2-sulfonamide), a member of a new class of triazolopyrimidine herbicides, while the chlorsulfuron-resistant biotype exhibited complete cross-resistance at both the whole plant and enzyme levels. ALS activity of the S biotype was reduced by approximately 90% in the presence of 0.1 micromolar D489, while that of the R biotype was reduced by less than 10%. This result suggests that the two herbicides share a common binding site on ALS. Only very slight cross-resistance at the ALS level was found to imazamethabenz, an imidazolinone herbicide.  相似文献
