The interaction of a plasma in the accelerating gap of an open discharge with a strong external electric field and with the cathode surface has been investigated theoretically and experimentally. In a pulsed nanosecond discharge, the ion inertia and plasma screening of the electric field cause a fast growth of the electric field E in the cathode region and a decrease in the length of the latter. Along with a reduction of the electron multiplication factor at high electric fields, this leads to a substantial decrease in the ion flux toward the cathode, which allows one to develop highly efficient open-discharge light sources with a long lifetime and low cathode sputtering. In this respect, continuous and quasi-continuous discharges are less advantageous because of the smaller increase in the electric field in the cathode region. The Townsend coefficients of charge multiplication and electron emission at high electric fields typical of open discharges have been measured for the first time. Fast ions and atoms extracted from the plasma of the accelerating gap significantly affect the cathode emission properties. In particular, photoemission is enhanced by more than one order of magnitude and becomes the main mechanism for electron generation. This also increases the efficiency and lifetime of open-discharge light sources.  相似文献

Results of a comparative study of the sensitivity of the system of respiratory control to increases in the CO2 concentration and the intensity of free-radical processes in young and elderly subjects are described. It is shown that normal (natural) aging is accompanied by a decrease in the sensitivity of the respiratory system to hypercapnic stimulation and a parallel significant decrease in the activity of catalase in the blood of examined subjects. Mechanisms responsible for the modifications of the sensitivity of the system of respiratory control to hypercapnia are discussed; these shifts can be at least partly related to changes in the intensity of production of free radicals observed in elderly subjects. In twoMacaca rhesus monkeys that received repeated N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) injections (single dose 0.2 mg/kg, i.m.; cumulative dose 11.2–13.3 mg), changes in characteristics of spontaneous saccadic eye movements and vestibulo-ocular reflex (VOR) were evaluated. With the development of severe behavioral disturbances, amplitude of spontaneous saccadic eye movements gradually decreased. Pronounced changes in duration of saccadic eye movements, frequency of spontaneous saccades, and their pattern were observed. Neuronal spike activity in the substantia innominata was investigated when establishing the instrumental conditioned reflex of lever-pressing with food reinforcement during chronic experiments on cats. Spike activity changed in 44.3% of neurons during this process; 24.3% responded directly to conditioned acoustic stimulation. Reactions associated with conditioned reflex movement were less pronounced. Response of peak intensity and duration was observed during food reinforcement, when the reaction occasionally occurred 500 msec in advance of the reinforcement provided. Involvement of cholinergic substantia innominata neurons in reinforcement is thought to be responsible for the persistence of residual effects in neocortical neurons.A. A. Structural proteins of the intermediate filament family are an early indicator of differentiation before organogenesis becomes apparent. Keratin intermediate filaments are characteristically expressed only by epithelial and not by mesenchymal cells. Here we show, using monoclonal antibodies, a transient expression of the keratin pair 8 and 18 in a population of mesenchymal cells in the regenerating newt limb, specifically in the undifferentiated progenitor cells (blastemal cells) which give rise to the new tissues. These keratins are also expressed in cultured limb cells that can differentiate into muscle. In contrast no reactivity with anti-keratin 8 and 18 antibodies was observed in the newt limb bud at an early stage of development, indicating a molecular difference between the developing and regenerating limb. The molecular weights of the newt proteins detected by these antibodies are very similar to those of human keratins 8 and 18, further supporting the immunocytochemical evidence that the newt homologs of these keratins are expressed in blastemal cells. This is the first demonstration of keratin expression in mesenchymal progenitor cells in an adult animal.  相似文献

Eye movements were investigated in cats while following a visual target. Wire coils implanted into the eyes served as transducers; the animal was placed in a revolving magnetic field (the magnetic search coil technique). The linear nature of amplitude-velocity relationships in saccadic eye movements was demonstrated. With combined head and eye movements, slope of plot was unrelated to maximum velocity of head movement over the entire test range (of up to 250 deg/sec); saccades decelerated when the head was immobile. Duration of gaze shift rose as it increased in amplitude. Amplitude of gaze was found to depend on head velocity. Experimentally obtained data on the interaction between head and eye movements when combined in following a target may be interpreted from the aspect of a mechanism operating to suppress saccadic signals by an efferent copy signal for head movement.M. Acetylcholine-induced membrane conductance was investigated in superior cervical ganglion neurons using a patch-clamp technique. It was found that hyperpolarization and depolarization produce an increase and a reduction in acetylcholine (ACh) conductance. This reduction was unconnected with either reversal of the current induced by iontophoretic ACh application or the presence of Ca ions in the external solution. The time constant of relaxation (r) of this current, produced by a jump in membrane potential, was found to increase e-fold when the membrane was hyperpolarized by 70 mV, matching the voltage dependence of ACh conductance. This led to the hypothesis that voltage-dependent ACh-induced conductance is entirely determined by the voltage dependence of nicotinic receptor channel gating kinetics.A. A. In anaesthetized dogs renal function was investigated in four successive 20-min periods in four experimental series. (1) In the first series following the first period (serving as control) 2.5 micrograms/kg/min of dopamine (DA) dissolved in 0.5 ml/min of Ringer's solution was infused into the left renal artery (period 2), than during periods 3 and 4. It was found that first (period 2) and second (period 3) doses of DA induced a significant decrease of about 20-30% in renal vascular resistance, and an increase of about 15-25% in renal blood flow. At the same time, systemic arterial blood pressure fell by 10%. The other investigated parameters of the left kidney (Cinulin, CPAH, sodium, potassium and water excretion) did not differ from the respective parameters of the intact right kidney. (2) In the second experimental series following the first period (prior to period 2) 1.0 mg/kg of the DA antagonist EGYT 2509 was administered intravenously. Prior to the period 3 again 1.0 mg/kg of EGYT 2509 and prior to period 4 2.0 mg/kg of EGYT 2509 was given intravenously. During periods 2 through 4 2.5 micrograms/kg/min of DA was infused into the left renal artery. It could be ascertained that EGYT 2509 abolished the renal effects of DA while not inducing any decrease in arterial blood pressure. (3) In the third experimental series, following the control period, prior to periods 2,3 and 4, 1.0 mg/kg, 1.0 mg/kg and 2.0 mg/kg chlorpromazine respectively, was administered i.v. followed by the infusion of DA into the left renal artery. After the administration of chlorpromazine arterial blood pressure and renal vascular resistance fell concomitantly and DA failed to induce any further changes in these parameters. According to our experiments chlorpromazine abolishes the effect of DA on kidney function. (4) In the fourth series, prior to DA infusion the dogs were given 0.5 mg/kg (period 2) then again 0.5 mg/kg and finally 1.0 mg/kg of haloperidol intravenously. Haloperidol decreased arterial blood pressure as well as renal vascular resistance, thus renal blood flow did not change. Renal blood flow could then be increased by DA infused into the left renal artery. It seems that haloperidol could not abolish the vascular effects of DA in the kidney. Our experiments indicate that substance EGYT 2509 possesses the most marked dopaminergic antagonistic effect, chlorpromazine had also been effective, while haloperidol had proved to be practically ineffective.  相似文献

Adult urodele amphibians can regenerate their limbs after amputation by a process that requires the presence of axons at the amputation plane. Paradoxically, if the limb develops in the near absence of nerves (the 'aneurogenic' limb) it can subsequently regenerate in a nerve-independent fashion. The growth zone (blastema) of regenerating limbs normally contains progenitor cells whose division is nerve-dependent. A monoclonal antibody that marks these nerve-dependent cells in the normal blastema does not stain the mesenchymal cells of developing limb buds and only stains the amputated limb bud when axons have reached the plane of amputation. This report shows that the blastemal cells of the regenerating aneurogenic limb also fail to react with the antibody in situ. These data suggest that the blastemal cells arising during normal regeneration have been altered by the nerve. This regulation may occur either at the time of amputation (when the antigen is expressed) or during development (when the limb is first innervated).  相似文献

Using the voltage clamp technique under conditions of intracellular perfusion, we investigated changes in high-threshold voltage-dependent calcium current (ICa) in the surface membrane of PC 12 cells caused by intracellular administration of the aromatic amino acids L-tyrosine and L-phenylalanine. Administration of L-tyrosine (20 mmole/1) prevented decrease in ICa caused by perfusion of the cell with an artificial saline solution and had a transient restorative effect upon the cell. Administration of L-phenylalanine (20 mM) quickened the decrease in amplitude of ICa observed in the control. These effect of aromatic amino acids are maintained when ATP (2 mM) in the intracellular solution is replaced by an equivalent quantity of ADP. The tyrosine hydroxylase blocker -methyl D,L-tyrosine (20 MM) had an effect upon ICa analogous to that of L-tyrosine.A. A.
