Abstract.  1. The sibling species Papilio glaucus and Papilio canadensis meet in a narrow hybrid zone believed to be maintained by temperature thresholds acting independently on both species. The present study tests if this assertion is true for the cold-adapted species, P. canadensis , which is presumed to be limited by the effect of high temperatures in late summer and/or autumn on pupal survival.
2. Three experiments were conducted examining the effects of: (i) short periods of high temperature stress in autumn, (ii) prolonged warm temperatures in autumn, and (iii) temperatures simulating warmer winters/longer springs upon the survival of P. canadensis and P. glaucus.
3. Results indicated that short periods of high temperatures did not induce the high mortality rates required to be the key factor limiting the range of P. canadensis . However, P. canadensis did exhibit a considerably lower tolerance to high temperature extremes, prolonged warm temperatures in autumn, and conditions simulating shorter/warmer winters than P. glaucus .
4. Differences in temperature tolerance throughout the pupal stage are likely to be a significant factor in maintaining the southern range limit of P. canadensis . Further warming as may occur during climate change, particularly in winter and spring, will likely affect the dynamics of southerly populations of P. canadensis.  相似文献

Translocations are a common management practice to restore or augment populations. Understanding the genetic consequences of translocation efforts is important for the long-term health of restored populations. The restoration of elk (Cervus canadensis) to Kentucky, USA, included source stocks from 6 western states, which were released at 8 sites in southeastern Kentucky during 1997–2002. We assessed genetic diversity in restored herds and compared genetic similarity to source stocks based on 15 microsatellite DNA loci. Genetic variation in the restored populations was comparable to source stocks ( allelic richness = 3.52 and 3.50; expected heterozygosity = 0.665 and 0.661 for restored and source, respectively). Genetic differentiation among all source and restored populations ranged from 0.000 to 0.065 for pairwise FST and 0.034 to 0.161 for pairwise Nei's DA. Pairwise genetic differentiation and Bayesian clustering revealed that stocks from Utah and North Dakota, USA, contributed most to restored populations. Other western stocks appeared less successful and were not detected with our data, though our sampling was not exhaustive. We also inferred natural movements of elk among release sites by the presence of multiple genetic stocks. The success of the elk restoration effort in Kentucky may be due, in part, to the large number of elk (n = 1,548), repeated releases, and use of diverse source stocks. Future restoration efforts for elk in the eastern United States should consider the use of multiple stock sources and a large number of individuals. In addition, preservation of genetic samples of founder stock will enable detailed monitoring in the future. © 2020 The Authors. Abstract.— Localized dispersal and mating may genetically structure plant populations, resulting in matings among related individuals. This biparental inbreeding has significant consequences for the evolution of mating systems, yet is difficult to estimate in natural populations. We estimated biparental inbreeding in two populations of the largely self-fertilizing plant Aquilegia canadensis using standard inference as well as a novel experiment comparing apparent selfing between plants that were randomly relocated within populations to experimental control plants. Using two allozyme markers, biparental inbreeding ( b ) inferred from the difference between single-locus and multilocus estimates of selfing ( b = ss – sm ) was low. Less than 3% of matings involved close relatives (mean b = 0.029). In contrast, randomly relocating plants greatly reduced apparent selfing (mean ss = 0.674) compared to control plants that had been dug up and replanted in their original locations ( ss = 0.953, P = 0.002). Based on this difference in ss , we estimated that approximately 30% of all matings involved close relatives (mean b = 0.279, 95% CL = 0.072–0.428). Inference from ss – sm underestimated b in these populations by more than an order of magnitude. Biparental inbreeding is thought to influence the evolution of self-fertilization primarily through reducing the genetic cost of outcrossing. This is unlikely to be of much significance in A. canadensis because inbreeding depression (a major cost of selfing) is much stronger than the cost of outcrossing. However, biparental inbreeding combined with strong inbreeding depression may influence selection on dispersal.  相似文献

Cloned DNA fragments coding for parts of strain WSN (H1N1) influenza virus hemagglutinin (HA) were fused to a bacterial leader DNA derived from the Escherichia coli trp operon. Fusion proteins produced consisted of 190 amino acids of trpLE' protein at the amino terminus, and HA amino acids, either 1-308, 1-396, or 1-548 (complete HA), at the carboxyl terminus. These proteins were expressed at high levels (10-20% of total protein) in E. coli starved for tryptophan. A CNBr fragment (HA1-211) was derived from HA-308. Each of the proteins was purified and used for immunizing mice and rabbits. The antibody produced was shown to bind to (i) the HA fusion proteins, (ii) detergent-treated viral HA, (iii) HA, on intact virions, and (iv) the HA on the surface of cells infected with influenza virus. This shows that the HA fusion proteins expressed in bacteria can elicit antibodies that recognize at least some determinants of the native viral HA, and probably could lead to development of an anti-influenza vaccine.  相似文献

BackgroundApoptosis played vital roles in the formation and progression of osteosarcoma. However, no studies elucidated the prognostic relationships between apoptosis-associated genes (AAGs) and osteosarcoma.MethodsThe differentially expressed genes associated with osteosarcoma metastasis and apoptosis were identified from GEO and MSigDB databases. The apoptosis-associated prognostic signature was established through univariate and multivariate cox regression analyses. The Kaplan–Meier (KM) survival curve, ROC curve and nomogram were constructed to investigate the predictive value of this signature. CIBERSORT algorithm and ssGSEA were used to explore the relationships between immune infiltration and AAG signature. The above results were validated in another GEO dataset and the expression of AAGs was also validated in osteosarcoma patient samples by immunohistochemistry.ResultsHSPB1 and IER3 were involved in AAG signature. In training and validation datasets, apoptosis-associated risk scores were negatively related to patient survival rates and the AAG signature was regarded as the independent prognostic factor. ROC and calibration curves demonstrated the signature and nomogram were reliable. GSEA revealed the signature related to immune-associated pathways. ssGSEA indicated that one immune cell and three immune functions were significantly dysregulated. The immunohistochemistry analyses of patients’ samples revealed that AAGs were significantly differently expressed between metastasis and non-metastasis osteosarcomas.ConclusionsThe present study identified and validated a novel apoptosis-associated prognostic signature related to osteosarcoma metastasis. It could serve as the potential biomarker and therapeutic targets for osteosarcoma in the future.  相似文献

Abstract Effectively managing habitat for threatened populations of Canada lynx (Lynx canadensis) requires knowledge of habitat conditions that provide for the ecological needs of lynx. We snow-tracked lynx to identify habitat conditions associated with hunting behavior and predation during winters of 2002–2003 and 2003–2004 in the northern Cascade Range in Washington state, USA. We recorded number and success of predation attempts, prey species killed, and trail sinuosity on 149 km of lynx trails. Lynx killed snowshoe hares (Lepus americanus), red squirrels (Tamiasciurus hudsonicus), and cricetids more than expected in Englemann spruce (Picea engelmannii) and subalpine fir (Abies lasiocarpa) forests, where snowshoe hare densities were highest. Lynx killed prey less than expected in Douglas-fir (Pseudotsuga menziesii) and ponderosa pine (Pinus ponderosa) forests and forest openings. We used the sinuosity of lynx trails as an index of quality of habitat hunted. Lynx trails that included predation attempts were more sinuous than trail segments without predation attempts. Lynx trails had greater sinuosity in forest stands with high hare densities dominated by Engelmann spruce and subalpine fir than in stands with low hare densities dominated by Douglas-fir and ponderosa pine or in forest openings. We encourage forest managers to maintain or create sufficient understory cover to support high densities of snowshoe hares as foraging habitat for lynx.  相似文献

Abstract. Photosynthetic utilization of HCO, in leaves of Poiamogeton and Elodea occurs at the lower leaf side, with subsequent OH∼ release at the upper side. It is accompanied by transport of cations, in the present experiment K +, across the leaf. The resulting pH and K+ concentration changes near the leaf surface were recorded with miniature electrodes. From the pH and K+ concentration the concentrations of the different inorganic carbon species were calculated and compared with photosynthetic O, production. HCO3 utilization is accompanied by a drastic increase in the free CO2 concentration near the lower epidermis. Experiments with CO2− and HCO3free solutions showed an oscillating acidification near the lower epidermis and alkalinization near the upper epidermis. It is concluded that the acidification results from the activity of light-dependent H+ pumps. The finding that an increase in pH at the upper side always coincided with a decrease at the lower in these experiments shows that the H+ pumps and the OH− extruding mechanism are coupled although occurring in different cell layers. Previously we have suggested that the first step in the process of photosynthetic HCO3 utilization is external conversion of HCO3" by acidification caused by light-dependent H+ pumps. The present results strongly support this hypothesis. Two possible pathways for the accompanying K + transport are discussed. The model presented here explains the known inhibiting effects of buffers and high pH on photosynlhetic HCO3 utilization.  相似文献

Three new fossils of saprotrophic fungi are presented and described from Baltic amber, dated to Eocene epoch (Paleogene, upper to mid-Eocene). All belong to Ascomycota and are represented by hyphae as well as asexual reproduction structures allowing to assign them to present genera, respectively Periconia, Penicillium and Scopulariopsis. These material provide both the first and the oldest known fossil record of the mentioned taxa, making these data valuable for the knowledge about the evolutionary history of the Ascomycota.  相似文献
