Thymidylate synthase (TS), an essential enzyme for catalyzing the biosynthesis of thymidylate, is a critical therapeutic target in cancer therapy. Recent studies have shown that TS functions as an RNA-binding protein by interacting with two different sequences on its own mRNA, thus, repressing translational efficiency. In this study, peptides binding TS RNA with high affinity were isolated using mRNA display from a large peptide library (>1013 different sequences). The randomized library was subjected up to twelve rounds of in vitro selection and amplification. Comparing the amino acid composition of the selected peptides (12th round, R12) with those from the initial random library (round zero, R0), the basic and aromatic residues in the selected peptides were enriched significantly, suggesting that these peptide regions might be important in the peptide-TS mRNA interaction. Categorizing the amino acids at each random position based on their physicochemical properties and comparing the distributions with those of the initial random pool, an obvious basic charge characteristic was found at positions 1, 12, 17 and 18, suggesting that basic side chains participate in RNA binding. Secondary structure prediction showed that the selected peptides of R12 pool represented a helical propensity compared with R0 pool, and the regions were rich in basic residues. The electrophoretic gel mobility shift and in vitro translation assays showed that the peptides selected using mRNA display could bind TS RNA specifically and inhibit the translation of TS mRNA. Our results suggested that the identified peptides could be used as new TS inhibitors and developed to a novel class of anticancer agents.  相似文献

BmK M4 is a neutral neurotoxin in the BmK toxin series.It is medially toxic and belongs to group III α-toxins.The purified sample was crystallized in rhombic space group P61.Using an X-ray diffraction technique,the crystal structure of BmK M4 was revealed by molecular replacement at 0.20 nm resolution.The model was refined.The final crystallographic R factor was 0.142 and the free R factor was 0.173.The root mean square deviation is 0.001 5 nm for the bond length and 1.753°for the bond angles.64 water molecules were added to the asymmetric unit.The refined structure showed an unusual non-prolyl cis peptide bond at residue 10.The structure was compared with group II α-toxin BmK M8 (an acidic,weak toxin).The potential structural implications of the cis peptide bond were discussed.  相似文献

Abstract The time course of loading and transport of assimilate in sunflower leaves was examined by pulse labelling with 14CO2, followed by freeze drying or freeze substitution, and dry autoradiography at both low and high resolution. The five classes of veins, V1-V5 (V5 being smallest), show a division of function: V5 and V4 are engaged in loading and short distance transport; V3 to V1, in long distance translocation. The first high concentration of 14C is found in two or three phloem parenchyma cells (intermediary cells) of V5 and V4 veins. The sieve elements of V5 and V4 veins do not show comparable concentrations of 14C at any time. Recently assimilated 14C is transported by the intermediary cells for distances of about 0.5 mm to the V3 veins. In V3 to V1 veins translocation is in the sieve tubes. Transport in V5 and V4 veins is in two directions, that in V3 to V1, in one direction towards the petiole. The high concentration of 14C formed in the intermediary cells does not increase further as the assimilate moves to the sieve tubes of the V3 veins, and so is probably the origin of the gradient that drives translocation.  相似文献

A high degree of genetic diversity among 125 peanut bradyrhizobial strains and among 32 peanut cultivars collected from different regions of China was revealed by using the amplified fragment length polymorphism (AFLP) technique. Eighteen different peanut bradyrhizobial genotypes and six peanut cultivars were selected for symbiotic cross-inoculation experiments. The genomic diversity was reflected in the symbiotic diversity. The peanut cultivars varied in their ability to nodulate with the strains used. Some cultivars had a more restricted host range than the others. Also the strains displayed a range of nodulation patterns. In yield formation there were clear differences between the plant cultivar/bradyrhizobium combinations. There was good compatibility between some peanut bradyrhizobial strains and selected cultivars, with inoculation resulting in well-nodulated, high-yielding symbiotic combinations, but no plant cultivar was compatible with all strains used. The strains displayed a varying degree of effectiveness, with some strains being fairly effective with all cultivars and others with selected ones. The AFLP genotypes of the strains did not explain the symbiotic behavior, whereas the yield formation of the plant cultivars was more related to the genotype. It is concluded that to obtain optimal nitrogen fixation efficiency of peanut in the field, compatible plant cultivar-bradyrhizobium combinations should be selected either by finding inoculant strains compatible with the plant cultivars used, or plant cultivars compatible with the indigenous bradyrhizobia.  相似文献

We investigated the roles of flooding, salinity, and plant competition in creating a bimodal zonation pattern of the marsh dominant annual plant, Suaeda salsa, along coastal topographic gradients on the Pacific coast of northern China. In two consecutive years, we manipulated salinity and flooding, salinity, and competition for S. salsa seedlings that had been transplanted into the mudflat, the high marsh, and the upland, respectively. S. salsa plants that had been transplanted into the mudflat were completely eliminated in the non-elevated treatments whereas they performed much better in the 10 cm elevated treatments, regardless of salinity treatments. Although the performance of S. salsa transplanted into the high marsh did not differ between the fresh (watered) and the salt (control) treatments, S. salsa seedling emergence in the high marsh was nearly completely inhibited in the salt treatments. In contrast, a large number of S. salsa seedlings did emerge in the fresh treatments. S. salsa transplanted into the upland performed well when neighbors were removed, whereas it appeared to be strongly suppressed when neighbors were present. These data indicated that flooding, salinity, and competition all played a role in determining the zonation pattern of S. salsa. Furthermore, the importance of salinity was found to vary with life-history stage. Based on the results from these field manipulative experiments, we suggest that the marsh plant zonation paradigm may hold true for plant distributions along landscape-scale topographic gradients from mudflats to uplands in general. The muroid Cricetops Matthew and Granger, 1923 commonly occurred in the Oligocene terrestrial deposits in central and northern Asia. Here we report the first record of Cricetops in the southern part of Asia. Isolated rodent molars named as a new species, Cricetops auster sp. nov., were discovered from the early Oligocene sediments at the Lijiawa locality in Yunnan Province in southwestern China. Compared to previously known Cricetops, C. auster is smaller than Cricetops dormitor Matthew and Granger, 1923 and Cricetops aeneus Shevyreva, 1965, but larger than Cricetops minor Wang, 1987. The cusps of C. auster are less conical. The ridges and crests are longer, higher and thicker. Relatively long and high crests, ridges and arms extending from the main cusps in the new species make those cusps more crescent in appearance than in C. dormitor, C. aeneus and C. minor. C. auster is a rare species in the Lijiawa mammalian fauna. Well-developed shearing tooth crests and ridges of C. auster probably suggest a different diet from the Cricetops from the northern part of Asia.  相似文献

A simple chromogenic assay for human alpha thrombin is developed through aptamer affinity capture and a subsequent enzyme reaction. Thrombin is captured on the aptamer-modified magnetic beads, and catalyzes the conversion of chromogenic substrates to optically measured products. The measurement of the generated products by an absorbance spectrometer allows for the final quantification of thrombin. This assay shows high sensitivity by taking advantage of sample enrichment and enzyme amplification, and exhibits good specificity by involving the selective aptamer binding and the specific enzyme reaction. A concentration detection limit of 40 fM can be reached when the tripeptide substrate of tosyl-Gly-Pro-Arg-p-nitroanilide is used in a 24 h enzyme reaction, and the use of 2h enzyme reaction in the assay enables the detection of 400 fM thrombin for a rapid analysis. This assay can be applied to detect thrombin in dilute human serum.  相似文献

To test the hypothesis that leaf‐level photosynthetic‐related traits might confer late successionals a competitive advantage over early successionals in low‐light growth conditions, steady photosynthetic assimilation‐ and dynamic photosynthetic induction‐related traits were examined in low‐light‐grown seedlings with contrasting successional status. Compared with the early successionals, late successionals as a group significantly exhibited lower leaf gas exchange rates. While late successionals required a longer time to respond to simulated sunflecks, they had lower rates of induction losses after sunflecks. Such photosynthetic induction traits allowed late successionals to more effectively utilize subsequent sunflecks. It was observed that plants with lower gas exchange rates responded more slowly to simulated sunfelcks, but they had lower rates of induction losses after sunflecks. In addition, the rate of response to sunflecks was positively correlated with the rate of induction loss after sunflecks across the successional status of species. A principal components analysis (PCA) demonstrated that early and late successionals were separated along the first axis of the PCA, and that early successionals were grouped on the right and were associated with higher gas exchange rates, fast responses to sunflecks, and rapid rates of induction loss after sunflecks; late successionals held an opposite pattern. Overall, our results suggest that smaller respiratory carbon losses and lower metabolic costs give late successionals a competitive advantage in low‐light growth conditions, that late successionals have an advantage over early successionals in utilizing sunflecks, and thus that the successional status of species are mainly associated with the leaf‐level photosynthetic‐related traits.  相似文献
