High-resolution genetic and physical maps were constructed for the region of chromosome 2 containing the major fruit-shape locus ovate. A total of 3,000 NIL F2 and F3 NILs derived from Lycopersicon esculentum cv. Yellow Pear (TA503) x L. pennellii (a wild tomato) were used to position ovate adjacent to the marker TG645 and flanked by markers TX700 and BA10R (a 0.03-cM interval). BAC libraries and a BIBAC library were screened with the closest marker, TG645. Genetic mapping with the ends of isolated BAC clones revealed that two BAC clones (100 and 140 kb) both contained the ovate locus. Screening of sequences from these BAC clones revealed synteny between this segment of tomato chromosome 2 and the chromosome-4 region of Arabidopsis containing the BAC clone ATAP22. Microsynteny between the two genomes was exploited to find additional markers near the ovate locus. The placement of ovate on a BAC clone will now allow cloning of this locus and, hence, may open the door to understanding the molecular basis of fruit development and also facilitate the genetic engineering of fruit-shape characteristics. This also represents the first time that microsynteny with Arabidopsis has been exploited for positional cloning purposes in a different plant family. 相似文献

The present study was designed to examine therapeutic efficacy of the root extract of Stephania Tetrandra S. Moore (STMS) (traditional Chinese medicine; Han Fang Ji) for treatment of neovascularization of the retinal capillary (retinopathy) in streptozotocin (STZ)-induced diabetic rats (STZ diabetic rats) in culture. Recently we have established the culture system in which fetal bovine serum (FBS) in Dulbecco modified Eagle medium (DMEM) induced neovascularization of the retinal capillary and choroidal capillary in normal rats in culture. STZ diabetic rats showed more neovascularization of the retinal capillary and choroidal capillary than did normal rats in culture. In this study, the retinal tissue was removed for the posterior ocular region and cultured in DMEM containing FBS. The choroidal tissue of the posterior ocular region was also removed and cultured as an internal reference. Administration of STSM (0.91, 9.1 and 91 microg/ml) significantly suppressed neovascularization of the retinal capillary in both STZ diabetic rats and normal rats in a dose-dependent manner. Similar results were obtained with the choroidal capillary; administration of STSM suppressed neovascularization of the choroidal capillary in both STZ diabetic rats and normal rats. In order to determine the component of STSM inhibiting neovascularization of the retinal capillary, tetrandrine (a major chemical constituent of STSM) was administered and neovascularization of the retinal capillary was examined in culture. The effect of tetrandrine on the choroidal capillary was also examined as an internal reference. Administration of tetrandrine (0.1, 1.0 and 10 microM) suppressed neovascularization of the retinal capillary in both STZ diabetic rats and normal rats in a dose-dependent manner. Similar results were obtained with the choroidal capillary of both STZ diabetic rats and normal rats. We infer, therefore, that STSM has a direct effect on the retinal capillary of posterior ocular region and suppresses neovascularization of retinal capillary in STZ diabetic rats through the activation of tetrandrine. Studies on the regeneration-restore and karyotype of protoplast in Metarhizium anisopliae var. majus
Abstract: The characteristics and regeneration-restore of protoplasts and its karyotype of an insect pathological fungus, Metarhizium anisopliae var. majus were studied. Among the protoplasts, 25.3% were without a nucleus, and 74.7% contained a nucleus. Among the nucleus protoplasts, 53.6% contained a single nucleus. The regeneration-restore of protoplasts was of three distinct shapes. Considering the frequency of regeneration and the growing speed of the colony, 0.7 mol/l glucose was the optimum as osmotic stabilizer of culture medium in the regeneration-restore of the protoplasts. The chromosomal DNA molecules of M. anisopliae var. majus have been separated into seven bands by pulsed-field gel electrophoreses. Using the Schizosaccharomyces pombe chromosomes as size standard, the size of chromosomal DNA was estimated to be 1.1–6.5 Mb and its karyotype exhibited polytypism among strains. 相似文献

Abstract: The leaf extracts of Populus nigra were collected and identified by steam distillation, air entrainment and gas chromatographic–mass spectrometric analysis. Electroantennograms were recorded from Helicoverpa armigera adults in response to the chemicals identified. Both aromatic compounds and green-leaf volatiles elicited strong responses. Field experiments revealed that the active compounds responsible for attracting H. armigera moths are mainly short-side-chain aromatic alcohols and aldehydes. We, for the first time, used agaropectin as the controlled-release matrix of insect attractants. A five-component lure containing all the aromatics without phenolics, mixed in the proportions as found in the steam distillate of the leaves collected in August, produced the best trap catch. The results showed that the volatiles of wilted leaves of P. nigra can attract H. armigera adults by feeding attraction. 相似文献

本文用细胞免疫化学方法,在冰冻切片上,检测了胎儿不同组织和器官内带γδ和αβ受体的T细胞(TCR)的分布,结果发现TCR细胞的分布与,般T细胞不同,有相对固定的分布区,如胸腺内TCR细胞主要分布在皮筋质交界处和髓质部;脾脏内的γδT主要位于边缘区,而αβT主要位于动脉周围淋巴鞘,在红髓和血窦两种细胞共存;淋巴结内只有少数TCR细胞位于滤泡间或副皮质,滤泡内则未见。消化管内的TCR细胞主要分布在小肠的固有膜,而胃、大肠和阑尾的固有膜内很少见;肝内TCR细胞主要集中在血管和血窦周围;皮肤切片内的少数TCR细胞见于真皮内,表皮基底层细胞内未见。这些细胞在胎儿期的免疫皮应及其生理功能还不清楚。 相似文献

本文用ABC—GDN免疫组织化学方法,研究了γ-氨基丁酸(Gamma—Aminobutyric Acid,GABA)在大鼠胰腺的定位和分布,并用相邻切片法,观察它与胰岛素的共存关系。结果发现GABA免疫反应阳性细胞主要分布于胰腺内分泌部(胰岛)。在外分泌部亦有少许分布。大部分胰岛细胞呈GABA免疫反应阳性,集中位于胰岛的中央部。相邻连续切片免疫染色证实GABA与胰岛素共存于胰岛B细胞中。外分泌部胰腺GABA免疫反应阳性细胞,呈零散分布于腺泡和导管上皮间。本文为进一步探讨GABA在胰腺的生理作用提供了形态学依据。 相似文献
