In plants, knowledge about linkage disequilibrium (LD) is relevant for the design of efficient single-nucleotide polymorphism arrays in relation to their use in population and association genomics studies. Previous studies of conifer genes have shown LD to decay rapidly within gene limits, but exceptions have been reported. To evaluate the extent of heterogeneity of LD among conifer genes and its potential causes, we examined LD in 105 genes of white spruce (Picea glauca) by sequencing a panel of 48 haploid megagametophytes from natural populations and further compared it with LD in other conifer species. The average pairwise r(2) value was 0.19 (s.d.=0.19), and LD dropped quickly with a half-decay being reached at a distance of 65 nucleotides between sites. However, LD was significantly heterogeneous among genes. A first group of 29 genes had stronger LD (mean r(2)=0.28), and a second group of 38 genes had weaker LD (mean r(2)=0.12). While a strong relationship was found with the recombination rate, there was no obvious relationship between LD and functional classification. The level of nucleotide diversity, which was highly heterogeneous across genes, was also not significantly correlated with LD. A search for selection signatures highlighted significant deviations from the standard neutral model, which could be mostly attributed to recent demographic changes. Little evidence was seen for hitchhiking and clear relationships with LD. When compared among conifer species, on average, levels of LD were similar in genes from white spruce, Norway spruce and Scots pine, whereas loblolly pine and Douglas fir genes exhibited a significantly higher LD.  相似文献

Soil streptomycetes are commonly antagonistic against plant pathogens. However, interactions involving increased defense responses in the host plant, leading to suppression of plant disease development, have not yet been detailed. Here, the mechanisms were studied of disease suppression by Streptomyces sp. GB 4-2 against Heterobasidion root and butt rot in Norway spruce (Picea abies) seedlings. GB 4-2 promoted mycelial growth of the phytopathogenic fungus, germination rate of fungal spores, extension of germ tubes and early colonization of outer cortical layers of the plant root. Reduced colonization of the inner cortical cell layers was accompanied by the induction of cell wall appositions, and increased xylem formation in the vascular cylinder emerged after bacterium-fungus coinoculation. Bacterial treatment led to decreased water content in roots and needles and increased photosynthetic yield (F(v)/F(m)) and peroxidase activities in needles. The infection of needles by Botrytis cinerea was reduced by bacterial pretreatment. Complex interactions of GB 4-2 with Norway spruce and Heterobasidion abietinum were discovered. The bacterium promoted the growth of the phytopathogenic fungus but induced plant defense responses. Host responses indicate that GB 4-2 induces both local and systemic defense responses in Norway spruce.  相似文献

The green algal communities in podzolic soils under coniferous forest in N.E. European Russia were studied in areas that were subjected to different technogenic pollution intensities. Sixty-five green algal species belonging to 4 classes, 12 orders, 20 families and 27 genera were recorded. The algal communities in the investigated soils of the different type spruce phytocoenoses included from 15 to 38 taxa on one site. Indicator species were identified for background spruce forests (Chalmydomonas gelatinosa, Tetracystis aggregata, T. dissociata, Pseudopleurococcus botryoides, Myrmecia bisecta) and for aerotechnogenically polluted spruce forests (Actinochloris sphaerica). The results may be used for the monitoring of the ecological state of biota in soils under boreal forests of protected areas and spruce forests exposed to intense aerial technogenic pollution. Because nitrogen is considered to be the major growth-limiting element in boreal forests, the increasing nitrogen availability from deposition should lead to increasing growth. We have tested this assumption by simulating, with a simple model, carbon and nitrogen development in seven long-term fertilization experiments in three Nordic countries. The only differences between sites in the model are climate, the ambient nitrogen deposition, nitrogen fertilization regimes, and the initial conditions at the start of the experiment. The model simulates the observed stand development well as long as nitrogen remains the limiting factor. The simulated retention of deposited nitrogen is in general low (less than 50%), whereas retention of fertilizer nitrogen is higher. This seems to imply that the higher production in the fertilized stands will not be maintained once the fertilization is stopped. The model results also indicate that the major effect of climate on site productivity is through soil processes, not tree physiology.  相似文献

Oil shale mining and processing in northeast Estonia have brought about several ecological problems. The mined oil shale is used as fuel in power stations and in processing plants producing crude oil and about 40 manufactured articles. Pollutants emitted from oil shale processing and chemical plants include SO2, CO, NO x , oil shale fly ash, and organic compounds in which aromatic and aliphatic hydrocarbons, phenols, formaldehyde, etc., are represented. Pollution has caused changes in the condition of the forest ecosystem and the chemical character of soil and ground water. The condition of coniferous forest sites was investigated in 1995–1998. Because of the high concentration of alkaline fly ash in the air, the pH of rain water is somewhat elevated (pH = 7.0–7.1) and exceeds the level regarded as normal for rain water. The analysis of the soil samples showed that the concentrations of Ca, Mg and K, which dominate in the solid fraction of the pollutant mixture, are high, being respectively 18, 14, and 4 times as high as the control. The increases in the concentrations of K, Mg, Cu, Pb, and Ni in stemwood reflect increases in the regional oil shale fly ash deposition. Conifers influenced by high levels of air pollution emitted from the oil shale industry are characterized by retarded growth of needles and shoots and radial growth as a result of disturbances in their mineral nutrition and imbalance in their mineral composition.  相似文献

Abstract. Reproducibility of vegetation measurements is critical for large‐scale or long‐term studies, where numerous observers collect data, but past studies have questioned repro‐ducibility of some techniques. Five methods of evaluating understory composition were appraised for reproducibility among six observers in two forest types in south‐central Alaska: ocular estimates in quadrats, overall community species rank and cover estimates, nested rooted frequency, horizontal‐vertical profiles, and pin drop (systematic points). One forest type was selected to represent structure of coastal communities, another to represent structure of interior Alaska communities. Three general methods of evaluating reproducibility were considered: standard deviations (precision among observers), components of variance (percentage of total variance attributable to observers), and analysis of variance (significance of observer variance). Observer variances were generally similar among techniques and significant in most cases. No technique stood out as being more reproducible than others. Features of techniques other than reproducibility may be more important when selecting a technique. Management decisions based on vegetation cover data should consider the observer errors involved as well as biological significance.  相似文献

This is a study of the colonization pattern of herbs and dwarf shrubs on rotten logs in subalpine spruce forests (Plagiothecio Piceetum) in the Tatra Mountains. On four study plots (total area 1.43 ha.) all dead logs were measured and the decomposition stage was estimated using the 8-degree scale. For each log the cover of all vascular species, bryophytes and lichens was determined according to the methods of classical phytosociology. Constancy and an index of coverage were calculated for all vascular species growing on logs. The total volume of logs was relatively high (93 m3 ha–1) and constituted 22% of the volume of living trees. Logs and stumps covered 411 m2 ha–1. These values are similar to those known from natural spruce forest from Carpathians and Scandinavia. The 8 stages of decomposition were equally represented, which indicates a constant supply of dead wood to the forest floor over time. The colonization of dead wood starts with lichens, followed by bryophytes and finally herbs and tree saplings. The first vascular plant colonists of dead logs appear at decay stage nr. 3 at least 20 years after tree death. The most suitable condition for most of the herb species corresponds to decay stage nr. 6 ca. 50 years after tree death. The herb cover is distinctively dominated by Vaccinium myrtillus. Simultaneously with herb species, tree seedlings colonize the logs. Constancy and abundance of Norway spruce saplings increases with advanced decomposition. It seems that the herb cover of logs does not hinder the regeneration of spruce.  相似文献

We describe a protocol for total RNA isolation from black spruce dry seeds, which is an adaptation of a method used for mouse myeloma tissues. We documented 13 behaviors associated with oviposition in unfertilized and fertilized spruce budworm moths, Choristoneura fumiferana, by videotaping active moths with a macro lens. Apart from resting, the most pronounced behaviors were probing, drumming + probing, and egg laying. Probing was a bending of the abdomen and extension of the ovipositor to touch the substrate in a rocking, back and forth motion. Drumming + probing involved tapping of the substrate with the pro- and mesothocacic legs with concurrent bending of the abdomen as above. Egg laying was the actual process of egg deposition onto the needle surface of a balsam fir twig. Both the frequency and the duration of these behavioral elements varied depending on the mating status of the female. Unfertilized females exhibited a higher frequency of probing and egg laying. The duration of probing was longer in unfertilized females, while drumming + probing and egg laying were longer for fertilized females. This study is the basis for future work on the chemosensilla associated with the perception of host-plant surface chemicals by ovipositing females.  相似文献

The effect of wood ash applied as a fertiliser (0.25, 0.5 and 1 kg m–2) to nutrient poor sandy soil on 4-year-old Norway spruce (Picea abies) growing in small-scale sample plots was studied. Analyses carried out with roots, stems, needles and shoots of different age showed that an increase in the pH level and K and Ca concentrations and a decrease in N and P concentrations in the soil was accompanied by a disbalance of nutrients in tree compartments. Stimulation of pigment synthesis in Norway spruce needles was observed, and no disbalances occurred in the pigment system (Chl a/Chl b, TChl/Car) guaranteeing normal process of photosynthesis. Biomass responses to wood ash application depended on the age of needles and shoots, being only slightly notable in current year organs. Inhibition of height growth of seedlings, but stimulation of root biomass was established.  相似文献
