This study focused on effects from Monoporeia affinis reworking and ventilation activities on benthic fluxes and mineralization processes during a simulated bloom event. The importance of M. affinis density for benthic solute (O2, ΣNO2 + NO3, NH4+ and HPO42−) fluxes and sediment reactivity (mobilization of NH4+ and HPO42−) following additions of organic material to the sediment surface was experimentally investigated using sediment-water and closed sediment (jar) incubations. Three different densities of M. affinis were used to resemble a low, medium and high density situation (1300, 2500 and 6400 ind. m− 2, respectively) of a natural amphipod community. The degradation of phytodetritus (Tetraselmis sp., 5 g C m− 2) added to the sediment surface was followed over a period of 20 days. Benthic solute fluxes of O2, ΣNO2 + NO3 and NH4+ were generally progressively stimulated with increasing number of M. affinis, while no such correlation was found for HPO42−. Solute fluxes were initially enhanced 1 to 2 days after the addition of phytodetritius, caused by mineralization of the most labile organic material and a food-stimulated irrigation by the amphipods. There was no effect from the activity of M. affinis on total denitrification (Dtot = Dn + Dw) or denitrification utilizing nitrate from coupled nitrification/denitrification (Dn) for any of the densities examined. Denitrification utilizing overlying water nitrate (Dw) was only about 10% of Dtot. Dw was significantly enhanced for the highest M. affinis density investigated. The reactivity of the sediment decreased progressively with increasing density of M. affinis and with time of the experiment. However, enhanced ammonium production at least 6 days after the organic addition indicated excretion of N-containing organic compounds by M. affinis. In conclusion, large spatial and temporal variations in density of M. affinis may be of significant importance for benthic solute fluxes and overall mineralization of organic material in Baltic Sea sediments.  相似文献

Chloride (Cl?) has been recently described as a beneficial macronutrient, playing specific roles in promoting plant growth and water‐use efficiency (WUE). However, it is still unclear how Cl? could be beneficial, especially in comparison with nitrate (NO3?), an essential source of nitrogen that shares with Cl? similar physical and osmotic properties, as well as common transport mechanisms. In tobacco plants, macronutrient levels of Cl? specifically reduce stomatal conductance (gs) without a concomitant reduction in the net photosynthesis rate (AN). As stomata‐mediated water loss through transpiration is inherent in the need of C3 plants to capture CO2, simultaneous increase in photosynthesis and WUE is of great relevance to achieve a sustainable increase in C3 crop productivity. Our results showed that Cl?‐mediated stimulation of larger leaf cells leads to a reduction in stomatal density, which in turn reduces gs and water consumption. Conversely, Cl? improves mesophyll diffusion conductance to CO2 (gm) and photosynthetic performance due to a higher surface area of chloroplasts exposed to the intercellular airspace of mesophyll cells, possibly as a consequence of the stimulation of chloroplast biogenesis. A key finding of this study is the simultaneous improvement of AN and WUE due to macronutrient Cl? nutrition. This work identifies relevant and specific functions in which Cl? participates as a beneficial macronutrient for higher plants, uncovering a sustainable approach to improve crop yield.  相似文献

We studied the dispersion patterns of the exotic endoparasitoid, Diachasmimorpha longicaudata (Ahsmed) (Hymenoptera: Braconidae), in 1999 (summer) and in 2000 (winter) in a citrus orchard in southeast Brazil. Different population densities of D. longicaudata were released in the centre of the orchard, and their dispersion was determined by using yellow, sticky, rectangular traps, placed in various distances and heights around the release point. Our results suggest that during summer, climatic conditions did not affect dispersion. However, in winter, dispersion rates were positively affected by temperature, and negatively by rainfall. Both estimated dispersal distance and surface were higher in summer than in winter for all release densities. Dispersion peaked at 2000 parasitoids ha?1 in summer and 8000 parasitoids ha?1 in winter. The importance of our results for the biological control of fruit flies by augmented or innoculative releases of D. longicaudata in southeast Brazil is discussed.  相似文献

Expression of Hox genes located on different chromosomes is precisely regulated and synchronized during development. In order to test the hypothesis that the Hox loci might cluster in nuclear space in order to share regulatory components, we performed 3D FISH on cryosections of developing mouse embryos and differentiating embryoid bodies. We did not observe any instances of co-localization of 4 different Hox alleles. Instances of 2 different alleles touching each other were found in 20-47% of nuclei depending on the tissue. The frequency of such “kissing” events was not significantly different in cells expressing a high proportion of the Hox clusters when compared to cells expressing none or only a few Hox genes. We found that the HoxB and HoxC clusters, which are located in gene-rich regions, were involved more frequently in gene kissing in embryonic nuclei. In the case of HoxB, this observation correlated with the positioning of the corresponding chromosome towards the interior of the nucleus. Our results indicate that co-regulation of the different Hox clusters is not associated with co-localization of the loci at a single regulatory compartment and that the chromosomal context may influence the extent to which they contact each other in the nucleus.  相似文献

Xiangxi Bay, a typical bay of the Three Gorges Reservoir (TGR) in China, was formed in June 2003 after the impoundment of the TGR. We investigated the community structure of zoobenthos in Xiangxi Bay from August 2003 to August 2004. Benthic abundance and biomass were calculated per unit area of sediment. A total of 26 taxa were identified. They included two families of Lamellibranchia (7.7% of the total taxa number), two families of Oligochaeta (50%), Chironomidae (38.5%), and one other animal (3.8%). The average density and biomass were 276 ind./m2 and 0.301 g/m2, respectively. Oligochaetes and chironomids constituted the major groups, and Limnodrilus hoffmeisteri, Procladius sp., Branchiura sowerbyi, Tubificidae sp1. and Polypedilum scalaenum group sp. were the common taxa in this reservoir system. Among them, L. hoffmeisteri and Procladius sp. were the dominant species, and contributed 37.0% and 28.3% of the total density, respectively. The standing crop of zoobenthos in Xiangxi Bay was relative low, compared with other similar reservoirs. Our studies demonstrate that the standing crop increased gradually with time. The total density and biomass in August 2004 were 8.8 and 14.3 times those of the initial impoundment, separately. Carbon stocks in managed forests of Ontario, Canada, and in harvested wood products originated from these forests were estimated for 2010–2100. Simulations included four future forest harvesting scenarios based on historical harvesting levels (low, average, high, and maximum available) and a no‐harvest scenario. In four harvesting scenarios, forest carbon stocks in Ontario's managed forest were estimated to range from 6202 to 6227 Mt C (millions of tons of carbon) in 2010, and from 6121 to 6428 Mt C by 2100. Inclusion of carbon stored in harvested wood products in use and in landfills changed the projected range in 2100 to 6710–6742 Mt C. For the no‐harvest scenario, forest carbon stocks were projected to change from 6246 Mt C in 2010 to 6680 Mt C in 2100. Spatial variation in projected forest carbon stocks was strongly related to changes in forest age (r = 0.603), but had weak correlation with harvesting rates. For all managed forests in Ontario combined, projected carbon stocks in combined forest and harvested wood products converged to within 2% difference by 2100. The results suggest that harvesting in the boreal forest, if applied within limits of sustainable forest management, will eventually have a relatively small effect on long‐term combined forest and wood products carbon stocks. However, there was a large time lag to approach carbon equality, with more than 90 years with a net reduction in stored carbon in harvested forests plus wood products compared to nonharvested boreal forest which also has low rates of natural disturbance. The eventual near equivalency of carbon stocks in nonharvested forest and forest that is harvested and protected from natural disturbance reflects both the accumulation of carbon in harvested wood products and the relatively young age at which boreal forest stands undergo natural succession in the absence of disturbance.  相似文献

Summary

One hundred and sixty-two agarics are recorded for Hirta and two from Dùn, two islands situated off the West coast of Scotland in the St. Kilda complex. The agarics are described in relation to the ecological noda proposed by McVean for the higher plant communities of the islands. Omphalina ericetorum and Nolanea staurospora were by far the commonest species: eighttaxa which are not in the New British Check List are recorded from Hirta. An appendix dealing with the taxonomy and nomenclature of the more critical species in the list is given.  相似文献

Hypothesis/objective: Prolonged QT interval is an index of propensity for dangerous ventricular tachyarrhythmias. The aim of this article is to establish an automatic algorithm for QT interval measurement.

Method: The proposed method is based on the continuous wavelet transform. In this method, the concepts of the rescaled wavelet coefficients and dominant scales of the electrocardiogram (ECG) components are used to perform detection of ECG characteristic points. A new concept of rescaled maximum energy density is introduced so as to perform the estimation of the QT interval.

Results and conclusion: We have applied the algorithm to the PTB database of the Physiobank?Physionet in lead II. Then, the results were evaluated using pertinent reference QT. The criterion used for evaluation of the method's performance is the root mean square (RMS) error. The method approached the RMS error of 27.89 ms for 549 subjects. The proposed method is fast, simple and is applicable to a wide range of ECG cardio cycle morphologies.  相似文献

ABSTRACT

Although process intensification by continuous operation has been successfully applied in the chemical industry, the biopharmaceutical industry primarily uses fed-batch, rather than continuous or perfusion methods, to produce stable monoclonal antibodies (mAbs) from Chinese hamster ovary (CHO) cells. Conventional fed-batch bioreactors may start with an inoculation viable cell density (VCD) of ~0.5 × 106 cells/mL. Increasing the inoculation VCD in the fed-batch production bioreactor (referred to as N stage bioreactor) to 2–10 × 106 cells/mL by introducing perfusion operation or process intensification at the seed step (N-1 step) prior to the production bioreactor has recently been used because it increases manufacturing output by shortening cell culture production duration. In this study, we report that increasing the inoculation VCD significantly improved the final titer in fed-batch production within the same 14-day duration for 3 mAbs produced by 3 CHO GS cell lines. We also report that other non-perfusion methods at the N-1 step using either fed batch or batch mode with enriched culture medium can similarly achieve high N-1 final VCD of 22–34 × 106 cells/mL. These non-perfusion N-1 seeds supported inoculation of subsequent production fed-batch production bioreactors at increased inoculation VCD of 3–6 × 106 cells/mL, where these achieved titer and product quality attributes comparable to those inoculated using the perfusion N-1 seeds demonstrated in both 5-L bioreactors, as well as scaled up to 500-L and 1000-L N-stage bioreactors. To operate the N-1 step using batch mode, enrichment of the basal medium was critical at both the N-1 and subsequent intensified fed-batch production steps. The non-perfusion N-1 methodologies reported here are much simpler alternatives in operation for process development, process characterization, and large-scale commercial manufacturing compared to perfusion N-1 seeds that require perfusion equipment, as well as preparation and storage vessels to accommodate large volumes of perfusion media. Although only 3 stable mAbs produced by CHO cell cultures are used in this study, the basic principles of the non-perfusion N-1 seed strategies for shortening seed train and production culture duration or improving titer should be applicable to other protein production by different mammalian cells and other hosts at any scale biologics facilities.  相似文献

Summary This study examined whether nonconfluent endothelial cell cultures reacted differently than confluent ones toward thrombin-stimulated platelets or a heparinized salt solution. The adherence to the endothelial cell cultures of51Cr-labeled human platelets stimulated at different thrombin concentrations was studied. There was significantly higher adherence of stimulated platelets to nonconfluent cultures compared with confluent ones. This was confirmed by scanning electron microscopy, which also revealed a tendency for the platelets to adhere at the cell periphery. Electron microscopy also showed that thrombin-stimulated platelets induced endothelial cell contraction. Part of the peripheral endothelial cell surface toward the bottom of the culture dish was inverted, facing the lumen of the dish. This phenomenon was particularly seen in nonconfluent cultures. When51Cr-labeled endothelial cultures were incubated with a mildly injurious fluid as heparinized sodium acetate and 20% serum, at 20° C for 30 min, the nonconfluent cultures showed significantly more cell detachment and release of51Cr than the confluent ones. We conclude that under the conditions of the present experiments there are differences in the reactivity of confluent and nonconfluent endothelial cell cultures. These differences probably reflect biological dissimilarities. In experiments where properties of cultured endothelium are studied, care should be taken that the degree of confluency is standardized.  相似文献
