天然林林下光质对乔木幼苗以及灌草的组成与更新具有重要的生态学意义。但目前对于林下光质的研究仍然有限。以吉林东部地区天然林为例,通过调查乔木数据和林下光质数据,基于移动窗口法分析不同空间尺度森林冠层结构与林下光质的关系。结果表明:不同林型下红光光子通量密度(R)与蓝光光子通量密度(B)存在差异。其中沙松-千金榆-花楷槭混交林林下蓝光光子通量密度最小,而沙松-紫椴-臭冷杉混交林和长白落叶松纯林林下最大。随着尺度的增大,天然林乔木胸高断面积与R/PFD(红光/光子通量密度比值)和B/PFD(蓝光/光子通量密度比值)的比值呈显著正相关(P<0.05)。并且随着尺度的增加,相关系数总体逐渐增大,在35m处达到峰值。在此基础上在南向、东向和西向各延伸10m时呈现显著正相关(P<0.05)。在该尺度下分析优势树种对林下R/PFD和B/PFD比值的影响时发现:R/PFD与B/PFD比值随着针叶林胸高断面积的增加而增加。相对于阔叶林来说,多数林型针叶林下的冠层结构与林下R/PFD和B/PFD比值之间显著正相关(P<0.05)。在不同树种下,乔木冠层结构对R/PFD和B/PFD比值的影响...  相似文献

本文介绍了本室建立的围堤式颅窗软脑膜微循环观察方法。大鼠暴露颅骨,顶骨钻孔后,固定一根4号头皮针于骨孔边缘,将牙托粉、牙托水调匀后于骨孔周围围一个环形堤,根据需要硬脑膜可以保持完整或剥去。经头皮针持续向颅窗内输恒温人工脑脊液(aCSF),并维持颅窗内aCSF的pH值和Pco2于正常水平。颅窗内数毫米厚的aCSF将外界隔开,使脑组织和软脑膜处于较好的生理环境。围堤式颅窗简便易行,并可达到与密闭颅窗相似的效果。  相似文献

寡聚蛋白质广泛地参与多种生命活动,对其预测研究有重要的意义。文章从蛋白质序列出发,提出多策略滑动伸缩窗特征提取方法,采用“ 一对一”的多类分类策略,对蛋白质同源寡聚体进行预测研究。结果表明,在Jackknife检验下,基于支持向量机的多策略滑动伸缩窗特征和氨基酸组成成分构成的特征集在加权情况下,其总分类精度最高达到了75.37%,比单纯的氨基酸组成成分法提高10.05%,比参考文献最好特征BG_Zhang提高了3.82%。 说明多策略滑动伸缩窗特征提取方法对于蛋白质同源寡聚体分类,是一种非常有效的特征提取方法。  相似文献

Biological treatment of many cancers currently targets membrane bound receptors located on a cell surface. To identify novel membrane proteins associated with migration and metastasis of breast cancer cells, a more migrating subpopulation of MDA‐MB‐231 breast cancer cell line is selected and characterized. A high‐resolution quantitative mass spectrometry with SILAC labeling is applied to analyze their surfaceome and it is compared with that of parental MDA‐MB‐231 cells. Among 824 identified proteins (FDR < 0.01), 128 differentially abundant cell surface proteins with at least one transmembrane domain are found. Of these, i) desmocollin‐1 (DSC1) is validated as a protein connected with lymph node status of luminal A breast cancer, tumor grade, and Her‐2 status by immunohistochemistry in the set of 96 primary breast tumors, and ii) catechol‐O‐methyltransferase is successfully verified as a protein associated with lymph node metastasis of triple negative breast cancer as well as with tumor grade by targeted data extraction from the SWATH‐MS data of the same set of tissues. The findings indicate importance of both proteins for breast cancer development and metastasis and highlight the potential of biomarker validation strategy via targeted data extraction from SWATH‐MS datasets.  相似文献

The effect of acid irrigation and liming on fine root growth of Norway spruce (Picea abies [L.] Karst.) was studied in an approximately 80-year-old forest stand in southern Germany (Höglwald). Root growth was measured mainly on root windows and in addition by soil core sampling. Root growth rate showed a typical pattern in the course of a year with a maximum in August. Acid irrigation depressed root growth rate, whereas liming, particularly in combination with acid irrigation, markedly increased root growth in the humic layer and the upper 0–5 cm of the mineral soil. The treatment effects on root growth in the mineral soil below 5 cm were small and not significant. Root growth rate was not correlated with the concentration of aluminium (Al) or the molar ratio of calcium (Ca) to Al in the soil solution. The results suggest that inhibition of root growth by acid irrigation is a direct effect of high proton concentrations in the irrigation water, and that enhancement of root growth by liming is caused by an improved supply of mineral nutrients and higher biological activity.  相似文献

Background

Genotype imputation can help reduce genotyping costs particularly for implementation of genomic selection. In applications entailing large populations, recovering the genotypes of untyped loci using information from reference individuals that were genotyped with a higher density panel is computationally challenging. Popular imputation methods are based upon the Hidden Markov model and have computational constraints due to an intensive sampling process. A fast, deterministic approach, which makes use of both family and population information, is presented here. All individuals are related and, therefore, share haplotypes which may differ in length and frequency based on their relationships. The method starts with family imputation if pedigree information is available, and then exploits close relationships by searching for long haplotype matches in the reference group using overlapping sliding windows. The search continues as the window size is shrunk in each chromosome sweep in order to capture more distant relationships.

Results

The proposed method gave higher or similar imputation accuracy than Beagle and Impute2 in cattle data sets when all available information was used. When close relatives of target individuals were present in the reference group, the method resulted in higher accuracy compared to the other two methods even when the pedigree was not used. Rare variants were also imputed with higher accuracy. Finally, computing requirements were considerably lower than those of Beagle and Impute2. The presented method took 28 minutes to impute from 6 k to 50 k genotypes for 2,000 individuals with a reference size of 64,429 individuals.

Conclusions

The proposed method efficiently makes use of information from close and distant relatives for accurate genotype imputation. In addition to its high imputation accuracy, the method is fast, owing to its deterministic nature and, therefore, it can easily be used in large data sets where the use of other methods is impractical.  相似文献

城市热环境是城市生态环境效应研究的热点之一,其演变规律的研究为缓解城市热岛带来的负效应、促进城市可持续发展提供依据。以2004年和2010年Landsat7 ETM+遥感影像数据和土地利用现状数据为数据源,在地表亮度温度反演的基础上,运用景观指数并结合GIS空间分析技术,采用移动窗口和梯度分析结合的方法,分析2004—2010年长沙市中心城区热环境的空间格局动态变化,通过分区统计法分析了不同热力景观等级下不同城市景观的空间格局变化,从景观尺度上阐明城市景观类型组成和空间格局与地表温度空间分异之间的关系。研究结果表明:2010年热岛区域扩大且更加分散,面积比2004年增加15.01km2,新增区域主要分布在金霞、岳麓和星沙的新兴工业园区;中心城区热力景观格局在景观水平上具有明显的空间分异特征,在从中心位置到偏北、偏东和偏南方向上,热力景观从市区向周边郊区呈现破碎化、多样性递增、形状复杂化,而从中心位置到偏西方向上与之相反;景观类型组成和空间布局对地表热环境产生不同影响,耕地、林地在热力景观内的优势度越大、分布越集中,地表降温效果越显著;反之,建设用地斑块越大、凝聚程度越高、形状越规整,地表温度越高,热岛效应显著。  相似文献

AIMS: To investigate whether two different wastewater treatment plants (WWTPs) -- treating the same pharmaceutical influent -- select for a different bacterial and/or ammonia oxidizing bacterial (AOB) community. METHODS AND RESULTS: Molecular fingerprinting demonstrated that each WWTP had its own total bacterial and AOB community structure, but Nitrosomonas eutropha and N. europea were dominant in both WWTP A and B. The DNA and RNA analysis of the AOB communities revealed different patterns; so the most abundant species may not necessarily be the most active ones. Nitritation failures, monitored by chemical parameter analysis, were reflected as AOB community shifts and visualized by denaturing gradient gel electrophoresis (DGGE)-based moving window analysis. CONCLUSIONS: This research demonstrated the link between functional performance (nitritation parameters) and the presence and activity of a specific microbial ecology (AOB). Clustering and moving window analysis based on DGGE showed to be valuable to monitor community shifts in both WWTPs. SIGNIFICANCE AND IMPACT OF THE STUDY: This study of specific community shifts together with functional parameter analysis has potential as a tool for relating functional instability (such as operational failures) to specific-bacterial community shifts.  相似文献

目的:探讨与对比心房颤动对不同时间窗内急性缺血性脑卒中患者静脉溶栓疗效的影响。方法:选择2014年8月到2016年5月在我院进行诊治的急性缺血性脑卒中患者98例,其中卒中前已诊断心房颤动定义为慢性心房颤动组(n=50),入院后诊断心房颤动者为新发心房颤动组(n=48);两组都给予重组组织型纤溶酶原激活剂(rt-PA)静脉溶栓治疗,记录两组预后情况。结果:两组患者的性别、年龄、时间窗、合并疾病、血糖与甘油三酯含量对比无明显差异(P0.05)。慢性心房颤动组与新发心房颤动组的有效率分别为94.0%和95.8%,组间比较差异无统计学意义(P0.05)。慢性心房颤动组与新发心房颤动组治疗后的m RS评分分别为6.22±1.83分和6.29±1.45分,都明显低于治疗前的9.24±1.31分和9.19±1.52分(P0.05),组间对比无明显差异(P0.05)。慢性心房颤动组的症状性脑出血与非症状性脑出血发生率分别为4.0%和2.0%,都明显低于新发心房颤动组的14.6%和12.5%(P0.05)。结论:发病4.5h之内静脉溶栓急性缺血性脑卒中是安全有效的,新发心房颤动不影响患者静脉溶栓后的神经功能结局,但是会增加症状性脑出血与非症状性脑出血,需要加强预防性管理。  相似文献
