An improved three-step Percoll density gradient centrifugation technique is described for simultaneous isolation of glial plasmalemmal vesicles (GPV) and synaptosomal vesicles (SYN) from a rat brain homogenate. While electron microscopy revealed that fractions contained intact vesicles with markedly distinct morphological features, measures of high-affinity [3H]choline uptake, glutamine synthetase and carbonic anhydrase activities, as well as Western blot analyses for glial fibrillary acidic protein and neuron specific enolase, served to confirm the low level of neuronal contamination in GPV fractions as well as the low level of glial contamination in SYN fractions. In addition, GPV and SYN fractions were used to characterize the kinetic and pharmacological properties of sodium-dependent [3H]L-glutamate transport. In conclusion, these results demonstrate the usefulness of this method for obtaining highly-enriched, functionally viable populations of glial and neuronal elements which are suitable for studies of their respective cell functions in vitro. 相似文献

人类对次生林生态系统的长期扰动必然会对地下生态过程产生深刻影响,而土壤动物群落结构和功能多样性对地下生态过程的演变有重要的指示意义.本文以小陇山林区恢复近30年的次生林和栽植近30年的油松、日本落叶松、欧洲云杉和粗枝云杉林为对象,通过对5种林型土壤动物群落密度调查,采用PCA排序和方差分析等方法,探讨了不同林型土壤动物群落结构特征和营养结构.结果表明:油松林和日本落叶松林土壤动物群落密度是次生林的3.0和2.1倍;油松和日本落叶松人工林土壤动物群落中消费者/分解者比值明显高于次生林,油松林和日本落叶松林消费者/分解者的比值显著高于欧洲云杉林和粗枝云杉林;不同人工林土壤动物群落结构存在明显差异,油松林和日本落叶松林土壤动物群落密度是粗枝云杉林的4.5和3.1倍,而油松林土壤动物群落类群丰富度是欧洲云杉林和粗枝云杉林的1.5倍. 相似文献

细菌在海洋生物地球化学循环中发挥着重要作用。为更好地了解海洋细菌的特征及其在海洋环境中的潜在作用, 本文利用纯培养与16S rRNA基因高通量测序技术对中印度洋与南海西部海域表层海水细菌多样性进行研究。纯培养结果表明, 自中印度洋与南海西部表层海水中共分离275株可培养海洋细菌, 隶属于4门49属75种。变形菌门是绝对优势类群(占总株数的68.7%), 其次是放线菌门(21.5%)、拟杆菌门(9.1%)和厚壁菌门(0.7%)。在属水平, 微杆菌属(Microbacterium)与弧菌属(Vibrio)是主要的优势属, 共占总株数的30.0%。在3种分离培养基中, 自1/10 × 2216E培养基中分离细菌的数目与种类最多(89株, 30属); 分离菌株中的细菌菌株有7、9与3个属分别仅在2216E、1/10 × 2216E及葡萄糖甘露糖(glucose-mannose, GM)培养基中生长。此外, 共分离培养出50株细菌(26种)可能代表潜在新分类单元。高通量测序结果显示, 中印度洋和南海西部表层海水中共有23个门531个属。优势门类为变形菌门(72.2%)和拟杆菌门(15.3%), 优势属为嗜冷杆菌属(Psychrobacter, 24.4%)、盐单胞菌属(Halomonas, 16.3%)和亚硫酸杆菌属(Sulfitobacter, 13.9%)。此外, 中印度洋表层海水细菌Shannon-Wiener指数与Pielou均匀度指数显著高于南海西部(P < 0.05), 且细菌群落结构显著不同(P < 0.05)。综合纯培养与原位细菌数据得出, 中印度洋与南海西部海洋细菌具有丰富的多样性, 具有进一步开发研究的价值。 相似文献

以课题组保存的连接在pMD-18T载体上的香蕉条斑病毒河口分离物全基因组为模板,设计一对特异引物,经过PCR扩增,切胶回收,并通过与pET32(b)载体共同双酶切及T4连接酶连接,得到重组载体pET32-ORF2。将获得的重组质粒pET32-ORF2转化表达菌株E.coli BL21(DE3)感受态细胞,得到表达BSV的ORFⅡ的工程菌。经0.1mmol/L的IPTG诱导表达,超声波破碎后,获得的融合蛋白大多为包含体,但仍有少部分为可溶性蛋白可以进行后续的纯化步骤。将上述可溶性融合蛋白通过Ni2+-NTA亲和层析柱纯化,获得纯度较高的融合蛋白。以纯化蛋白作为抗原免疫家兔,得到特异性抗血清。间接ELISA以及western blotting表明,所得抗血清效价和特异性都较高,可以用于检测。为BSV的快速检测以及一些相关后续理论研究工作奠定了一定基础。 相似文献

Life-history traits and ecological conditions have an important influence on primate social systems. Most of what we know about the life-history patterns and social structure of gorillas comes from studies of eastern gorillas (Gorilla beringei sp.), which live under dramatically different ecological conditions compared to western gorillas (Gorilla gorilla sp.). In this paper we present new data on western gorilla social structure and life histories from four study sites, and make comparisons with eastern gorilla populations. Data were obtained from two study sites with gorilla groups undergoing the habituation process (Lossi, Democratic Republic of Congo and Bai Hokou, Central African Republic) and two "bai" studies (Maya Nord and Mbeli Bai, Republic of Congo). The size and structure of these groups were similar to those seen in eastern gorillas. However, differences in the occurrence of various group transitions (group formations, changes between one-male and multimale composition, and group disintegrations) exist, and western gorillas notably exhibit much higher rates of male emigration and correspondingly fewer multimale groups compared to mountain gorillas. Certain phenomena have been observed only rarely, including predation by leopards. The preliminary data show no significant differences in birth rates between western gorillas and mountain gorillas. The ecological variability across gorilla habitats likely explains the flexibility in the social system of gorillas, but we need more information on the social relationships and ecology of western gorillas to elucidate the causes for the similarities and differences between western and eastern gorillas on the levels of individuals, social groups, and population dynamics. 相似文献

Surfactant protein-A (SP-A) belongs to a family of collagen-containing C-type lectins called collectins. SP-A is expressed by renal tubule epithelial cells. We investigated the distribution of SP-A in renal cell carcinomas (RCC) using immunohistochemical techniques and western blotting. We used 35 formalin fixed, paraffin embedded (FFPE) RCC tissue samples. We compared results with clinico-pathological parameters of RCC including age, sex, Fuhrman grade, tumor volume, tumor node metastasis (TNM) and clinical stage. SP-A was localized in the glomerulus and renal tubule epithelium in nontumor tissue and strong SP-A immunoreactivity was observed in tumor tissue. SP-A was expressed in the RCC tumor cells (64%) and nontumor cells (34%) in males and RCC tumor cells (90%) and nontumor cells (30%) in females. There was a significant correlation between SP-A immunoreactivity in tumor cells and gender, age, tumor diameter, Fuhrman grade and tumor diameter. Western blot analysis supported the immunohistochemical findings. We present evidence for involvement of SP-A in RCC and suggest that increased SP-A expression in RCC is associated with favorable prognosis. 相似文献

The endemic fauna of the Comoro Archipelago is composed of a mixture of taxa originating from Africa and Madagascar. Bats are the only native land dwelling mammals on this archipelago, but the biogeographical origins for the vast majority of species within this group are ambiguous. We report here genetic analyses based on two mitochondrial DNA markers to infer the origin of Comorian bats belonging to a reputed species complex of Miniopterus that is further distributed across Africa and Madagascar. Phylogenetic reconstructions show that east African M. minor are not closely related to the insular Miniopterus of Madagascar and the Comoros (Grande Comore and Anjouan). The latter cluster into two distinct, monophyletic clades (Clade 1 and Clade 2). Representatives of these clades occur sympatrically both on the Comoros and on Madagascar, and are distinguished by a large genetic distance (K2P: 9.9% for cytochrome b). No haplotypes are shared between any islands, suggesting the absence of contemporary gene flow. Populations of the widespread Clade 1 are furthermore characterized by a significant inter‐island structure (ΦCT = 0.249), and by high haplotype and nucleotide diversities (h = 0.90–0.98, π = 0.04–0.06). Demographic analyses of Clade 1 suggest secondary contact between two distinct phylogroups (Subclade 1 A and 1B) that reached Grande Comore and Anjouan, and a large, stable population with a long evolutionary history on Madagascar. These results and the current distribution of related lineages suggest that the Comoros were colonized independently at least two or three times by ancestors from Madagascar. 相似文献
