The potato has tremendous potential as a transgenic crop and is a good model system by which to analyse metabolic regulation and gene expression. The potato’s difficult genetics, but ease of genetic transformation and its clonal means of propagation make it ideal for applied agricultural molecular genetics. Thus, the next 4 years promise to put the potato (with a diversity of transgenic constructs expressed) in the limelight as many of the first transgenic agricultural products enter the marketplace.  相似文献

The ethnic and geographic distributions of several common chronic diseases show distinct patterns that are consistent with the distribution of genes and genetic admixture. For example, diabetes and gallbladder disease occur most frequently among Amerindians, while those genetically admixed with them (such as Mexican-Americans) have intermediate rates, and lowest rates are found among Whites and Blacks. Because there will be heterogeneity from individual to individual in ancestral affinity within an admixed population, a method is developed for estimating each person's admixture probability. Results confirm that there is substantial heterogeneity of individual admixture among Mexican-Americans in Starr County, Texas, with a mean value indicating that 65% of genes in this population are Caucasian derived and 35% Amerindian derived. The individual estimates are shown to be unrelated to the probability of being diabetic and only marginally related to gallbladder disease, with those having the most Amerindian affinity being at increased risk. These results are a consequence of the independent assortment of loci and indicate that unless the markers employed are related (including linkage) to the disease of interest, the method will have limited utility. Individual admixture estimates will be useful, however, for examining aspects of population structure and will find increased utility for predicting disease and examining disease associations as more and more of the genome is represented by markers, a very probable prospect with the abundance of DNA polymorphism being identified by restriction enzymes.  相似文献

The knapweed nematode, Subanguina picridis, forms galls on the leaves, stems, and root collar of Russian knapweed, Acroptilon repens. After being revived from a dormant, cryptobiotic state, second-stage juveniles required at least 1 month in a free-living state before becoming infective. Galls were induced on relatively slow-growing host plants that retained their apical meristems at or near the soil surface for 2-5 weeks. Galls developed extensive areas of nutritive tissue. The nematode was introduced from the Soviet Union and released in Canada for the biological control of Russian knapweed.  相似文献

Grain sorghum can substitute for corn as a full season crop and replace soybeans in double cropping systems with wheat in the southeastern United States. Relatively few studies have been conducted to measure the response of grain sorghum to tillage, weed control method, and row spacing. These experiments were designed to determine the effects of weed control method and row spacing on no-till planted grain sorghum (Sorghum bicolor L. Moench G1516-BR) after wheat (Triticum aestivum L. Coker 68–15) and crimson clover (Trifolium incarnatum L. Bigbee) grown for winter forage in comparison to sorghum planted on a conventionally prepared seedbed. The experiment included 45, 60, and 90 cm row spacings and three weed control regimes: none, mechanical, and chemical. Grain sorghum planted no-till in crimson clover or wheat sod yielded considerably more grain than conventionally planted sorghum. Grain sorghum produced significantly higher yields in 45-cm rows than in 60-and 90-cm row spacings with all three planting methods. Effects of chemical weed control on weed population with all tillage methods and on grain yield with conventional tillage were significant. There were no significant differences in grain protein content due to row spacing or weed control method.  相似文献

Glyphosate is the active ingredient of the broad-spectrum, translocated herbicide Roundup. Glyphosate is cleared for safe use in or near watercourses, being rated virtually non-toxic by the World Health Organisation. Trials in and alongside Irish fishery watercourses first commenced in 1989 and are continuing to date. The aim of this work is to evaluate the product's efficacy in clearing nuisance reed species in recreational fisheries. The longevity of control and impact on the habitat and its fauna is also investigated. Trials in canal fisheries have demonstrated the capacity of glyphosate to remove obstructive stands of reeds (mainly Schoenoplectus lacustris, Glyceria maxima, Phragmites australis, Sparganium erectum and Typha latifolia), so creating reed-free areas and swims for anglers. These swims remained open for three years following a single application. In 1992 a trial over a 3 km length of the River Boyne, a renowned salmonid fishery, was undertaken. The results clearly demonstrated the ability of glyphosate to provide long-term control of dense (354 shoots m–2) Schoenoplectus infestations in a large watercourse. In the year following, less than one shoot per m2 was present in the channel. In 1994 a small increase in density (7.6 shoots m–2 was recorded, so enabling unobstructed angling in a stretch of river that had been virtually unfishable for years. Trout (Salmo trutta L.) and salmon (Salmo salar L.) also used the newly exposed gravels for spawning in the winter of 1993, thereby improving fish recruitment and production in the fishery.  相似文献

Uncultivated plants growing on disturbed sites may be useful for assessing the bioavailability of some metals in soils, and thus the potential for metal mobilization up the terrestrial food chain, an important element in ecological risk assessment. A planted chicory cultivar (Cichorium intybus L. var. foliosum Hegi.) and the uncultivated plants horseweed (Canada fleabane) (Erigeron canadensis L.) and dogfennel (Eupatorium capillifolium (Lam.) Small) were evaluated for their ability to act as index plant species for soil Cd, Cr, Ni, and V at two field sites where these metals had been applied five yr previously to two highly weathered sandy Ultisols. Soil Cd was available to all analyzed plant tissues of all three plant species at both sites, particularly on the sandier Blanton soil. Chicory was an effective index plant for Cd on the finer textured Orangeburg soil but functioned as an indicator plant (toxicity symptoms were observed) on the sandier Blanton soil. Horseweed and dogfennel were effective index plants for Cd in both contaminated soils. Soil Cr, Ni, and V were less bioavailable than soil Cd and plant metal uptake was more sensitive to residual soil Cr, Ni, and V than was soil extraction with double acid. Horseweed and chicory may have potential as index plants for soil Cr. Chicory may have potential as a Ni index plant. Chicory and dogfennel may have potential as V index plants.  相似文献

Effects of soil application of Ascochyta caulina spores on seedlings of Chenopodium album and five cultivated plant species were investigated under greenhouse conditions as a part of a study on biological control of C. album. Application of A. caulina spores to soils resulted in disease development on C. album and to a lesser degree on Spinacia oleracea seedlings, but not on Beta vulgaris subspecies vulgaris, Zea mays, Triticum aestivum and Pisum sativum seedlings. Affected C. album seedlings had an abnormal olive-green colour or necrotic spots on cotyledons and hypocotyls, and were stunted or died. Affected S. oleracea seedlings were pale in colour or had necrotic spots on the cotyledons, but did not die. Time courses of disease incidence and of mortality of C. album could be described by a monomolecular model. Effects of spore density, sowing depth, soil water content, soil type and time of sowing on disease development were examined. Disease incidence and mortality were influenced by spore density, soil water content and soil type, but not by sowing depth. Spores in a moist soil maintained infectivity at least 2 wk. Spore densities of 109 to 1010 spores m-2 were required for 50% mortality of emerged C. album plants. Aspects of the development of A. caulina into a soil-applied mycoherbicide are discussed.  相似文献

The quality of sediment was assessed in 46 sites in the delta of the rivers Rhine and Meuse (The Netherlands) by means of physical-chemical analysis, field observations on the macrobenthic community structure, accumulation studies and bioassays using Chironomus riparius, Daphnia magna and Photobacterium phosphoreum. The results of chemical analyses were classified using national criteria for sediment quality. Results of field studies and bioassays were classified using criteria derived from research in reference areas or based on data from literature. Risk assessment was carried out according to the sediment quality triad and by means of multi criteria analysis (MCA). The Triad approach was used to demonstrate causal relations between effects on the macrozoobenthos community structure, effects demonstrated in bioassays and sediment pollution. This was done by making a comparison of sediment contamination levels with toxicity data from literature for the test organisms of the bioassays. Using the MCA method, for each site a numerical value was derived for total environmental risk in the present situation, based on observed effects. In this way, a relative risk ranking of all sites was realized. The MCA values for the present situation were also compared with MCA scores based on estimated risks after remediation in 1995, in order to set priorities for sites where remediation is expected to cause a significant reduction in environmental risk. In most of the 46 sites studied so far, the macrofauna community was poorly developed, judged by a low number of benthic species, low abundances and a high dominance of species regarded as relatively tolerant to chemical pollution. In bioassays high sediment toxicity was demonstrated for 24 sites. Using the sediment quality triad approach, 25 sites were identified as areas where pollution can be held responsible for the effects observed in the field. From a comparison of contaminant concentrations in different types of food with maximum tolerable risk levels, and the application of a bioaccumulation model it was concluded that the sediment pollution also implies high risks for plant-, benthos- or fish-eating birds (secondary poisoning of top predators). In the Nieuwe Merwede highest MCA risk scores were found for shallow parts where highly polluted sediments are found. It is concluded that the sediment quality triad and the MCA provide additional information which can be used to establish priorities for remedial action. Based on an ecotoxicological evaluation of the improved quality of new sediments that will be deposited after removal of the polluted sediments in the Nieuwe Merwede, it is concluded that in this upstream part of the Rhine delta remedial action will be effective.  相似文献

Laboratory incubation experiments were used to study the effect of reduced concentrations of organic micropollutants in water from the rivers Rhine and Meuse on the specific growth rate of the river phytoplankton community. Before incubation, part of the water sampled was treated with XAD-4 and XAD-8 resins to absorb dissolved organic compounds. Four dilutions were made by mixing untreated water with XAD-treated water in the ratios 100:0 (control), 70:30, 40:60 and 0:100. The phytoplankton specific growth rate increased significantly with the increased fraction treated with XAD in all but one incubation experiment. In these experiments, the specific growth rate was on average 9% higher in the fraction in which 100% was treated with XAD than in the controls. In the Rhine and Meuse river water, phytoplankton growth seemed to be inhibited by organic compounds. This inhibition was ascribed to the presence of dissolved organic micropollutants. Removing organic micropollutants using XAD resins to study the toxic effects of these compounds on field phytoplankton communities can be concluded to be a promising tool for risk assessment of micropollutants but needs to be supported by additional methodological research.  相似文献
