Upon exposure to waterlogged growing conditions two-year-old alder trees reduced total root mass. Roots were concentrated in the uppermost soil horizon, and only few coarse roots penetrated into deeper soil layers. Root porosity was only slightly affected and did not exceed 8 % in fine roots. Porosity of coarse roots was higher (27 %) but unaffected by growing conditions. The stem base area covered by lenticels increased strongly and so did the cross section diameter of the stem base. The latter showed a highly significant correlation with O (2) transport into the roots, measured by a Clark type oxygen electrode. Exposure of the lower 5 cm of the stem base, where lenticels were concentrated, to pure N (2) led to a cessation of O (2) transport, confirming that lenticels were the major site of air entry into the stem. In alder plants grown under waterlogged conditions, temperature had a pronounced effect on O (2) gas exchange of the root system. The temperature compensation point, i.e., the temperature where O (2) transport equals O (2) consumption by respiration, was 10.5 degrees C for the entire root system, when measured in a range of 0.15 - 0.20 mmol dissolved O (2) L (-1), which is typical for an open water surface equilibrated with air. O (2) net flow was inversely related to O (2) concentration in the rooting media, indicating that higher root and microbial respiration induced higher net fluxes of O (2) into the root system. With 0.04 mmol dissolved O (2) L (-1) nutrient solution, the temperature compensation point increased to 20 degrees C. Measurement of O (2) gradients in the rhizosphere of agar-embedded roots using O (2) microelectrodes showed a preference for O (2) release in the tip region of coarse roots. Increasing stem temperature over air temperature by 5 degrees C stimulated O (2) flux into the roots as suggested by the model of thermo-osmotic gas transport. However determination of stem and air temperature in a natural alder swamp in northern Germany revealed that within the experimental period of almost one year, temperature gradients required for thermo-osmotic gas transport were very seldom. From this it is concluded that under natural conditions in northern Germany, oxygen diffusion along the stem into the root system is driven by O (2) concentration gradients rather than by thermo-osmosis.  相似文献

Hypoxia-induced changes in net H+, K+ and O2 fluxes across the plasma membrane (PM) of epidermal root cells were measured using the non-invasive microelectrode ion flux measurement (MIFE) system in elongation, meristem and mature root zones of two barley (Hordeum vulgare L.) varieties contrasting in their waterlogging (WL) tolerance. The ultimate goal of this study was to shed light on the mechanisms underlying effects of WL on plant nutrient acquisition and mechanisms of WL tolerance in barley. Our measurements revealed that functionally different barley root zones have rather different O2 requirements, with the highest O2 influx being in the elongation zone of the root at about 1 mm from the tip. Oxygen deprivation has qualitatively different effects on the activity of PM ion transporters in mature and elongation zones. In the mature zone, hypoxic treatment caused a very sharp decline in K+ uptake in the WL sensitive variety Naso Nijo, but did not reduce K+ influx in the WL tolerant TX9425 variety. In the elongation zone, onset of hypoxia enhanced K+ uptake from roots of both cultivars. Pharmacological experiments suggested that hypoxia-induced K+ flux responses are likely to be mediated by both K(+) -inward- (KIR) and non-selective cation channels (NSCC) in the elongation zone, while in the mature zone K(+) -outward- (KOR) channels are the key contributors. Overall, our results suggest that oxygen deprivation has an immediate and substantial effect on root ion flux patterns, and that this effect is different in WL-sensitive and WL-tolerant cultivars. To what extent this difference in ion flux response to hypoxia is a factor conferring WL tolerance in barley remains to be answered in future studies.  相似文献

In flood-tolerant species, a common response to inundation is growth of adventitious roots into the water column. The capacity for these roots to become photosynthetically active has received scant attention. The experiments presented here show the aquatic adventitious roots of the flood-tolerant, halophytic stem-succulent, Tecticornia pergranulata (subfamily Salicornioideae, Chenopodiaceae) are photosynthetic and quantify for the first time the photosynthetic capacity of aquatic roots for a terrestrial species. Fluorescence microscopy was used to determine the presence of chloroplasts within cells of aquatic roots. Net O2 production by excised aquatic roots, when underwater, was measured with varying light and CO2 regimes; the apparent maximum capacity ( P max) for underwater net photosynthesis in aquatic roots was 0.45  µ mol O2 m−2 s−1. The photosynthetic potential of these roots was supported by the immunolocalization of PsbA, the major protein of photosystem II, and ribulose-1-5-bisphosphate carboxylase/oxygenase (Rubisco) in root protein extracts. Chlorophyllous aquatic roots of T. pergranulata are photosynthetically active, and such activity is a previously unrecognized source of O2, and potentially carbohydrates, in flooded and submerged plants.  相似文献

The aim of this study was to determine the effect of waterlogging on annual regeneration in the dwarf birch (Betula nana L.), and to determine which demographic factors are responsible for the absence of B. nana in the waterlogged parts of a peat-bog. Growth parameters and demographic data were collected from 2001 to 2003 at four sites in the Linje peat bog in the Chełmno Lake District in northern Poland. Three of the sites represented waterlogged open bog communities. The fourth site served as the reference. The density of first-year ramets, annual shoot growth, and the proportion of flowering shoots was significantly lower at the waterlogged sites than at the reference site. No seedlings were found during the course of the study, even though flowering and fruit production took place at the reference site. The flowering shoots were mainly between nine and thirteen years old. Ramets in this age range were found only at the reference site due to high mortality in mature ramets at the waterlogged sites. Mean ramet height was significantly lower at the waterlogged sites than at the reference site, and was strongly correlated with mean ramet age. The distribution of B. nana in peat bogs is limited primarily by two demographic factors: the high mortality rate in older ramets, and the low rate of vegetative reproduction. Another contribution factor is that the proportion of older fertile ramets is low. The effects of fluctuations in the water table on distribution of B. nana are also discussed.  相似文献

分别构建了含cry3Acry3A+vhb基因的植物表达载体pBCry3A和pBC3Vhb,并通过根癌农杆菌介导转化了马铃薯. 对转化再生植株进行PCR和DNA印迹分析表明,外源基因已整合到马铃薯基因组中, 且连续三代无性繁殖后转基因仍存在. ELISA分析表明cry3A基因在转基因植株中得到了高效表达, 在单转cry3A植株中最高表达量达0.1%, 转cry3Avhb双基因株系中为0.065%. 水涝试验显示,转双基因且vhb mRNA的RT-PCR呈阳性的马铃薯植株,对低氧胁迫有较好的耐受性, 表明获得的上述转双基因马铃薯株系可能会具有很好的抗虫和耐涝性能.  相似文献

High-density olive orchards are increasing around the world, many of which may be potentially affected by salinity and waterlogging (hypoxia), two important stresses common in irrigated fields in arid and semi-arid climates. However, the response of olive to these stresses under field conditions is not well established. Therefore, our objective was to evaluate the vegetative growth response of young olive trees (Olea europaea L., cv. Arbequina) grown in a spatially variable waterlogged, saline-sodic field. We monitored the growth in trunk diameter of 341, 3-year-old olives between September 1999 and September 2000. Field contour maps were developed delineating soil salinity (ECa), relative ground elevation (RGE) and water table depth (WTD). Soil samples were also collected and analyzed for ECe and SARe in order to characterize the salinity and sodicity profiles and develop the ECa-ECe calibration equation. The infiltration rate (IR) of the crusted and uncrusted soil and the penetration resistance (PR) were also measured. The field was characterized by spatially variable ECe (2–15 dS m–1), SARe (3–40), RGE (–4 to +4 cm) and WTD (0.5–1.9 m, with corresponding ground water EC values between 12 and 6 dS m–1). Steady-state IR of crusted soil was only 7% of the uncrusted soil. Since the field was heavily irrigated by flooding, waterlogging conditions were related to low RGE values. Soil salinity was negatively correlated (R 2 = 0.83, P<0.001) with RGE (ponded water) and WTD (upward flux), due to the evapo-concentration of water and salts at the soil surface. Thus, inverted salinity profiles developed in high salinity areas. Fifty-five percent of the olives were dead 3.5 years after planted, and most of them were located in areas of high ECe (> 10 dS m–1), low RGE (< – 1.5 cm) and low WTD (< 1.2 m). The surviving trees had vegetative salinity tolerance values of ECe threshold = 4 dS m–1 and slope = –12% (i.e., percent decline per unit increase in ECe above the treshold), indicating that the Arbequina olive is moderately tolerant to salinity. The RGE and WTD thresholds for olive's survival were > 0.1 cm and > 1.6 m, respectively. Thus, very small changes in ground elevation had a significant effect on olive's survival or death. The coupled effects of salinity and waterlogging (hypoxia) stresses were most detrimental for olive's growth.  相似文献

Excessive soil wetness is a common feature where bananas (Musa spp.) evolved. Under O2 deficiency, a property of wet soils, root growth and functions will be influenced by the respiratory demand for O2 in root tissues, the transport of O2 from the shoot to root and the supply of O2 from the medium. In laboratory experiments with nodal roots of banana, we examined how these features influenced the longitudinal and radial distributions of O2 within roots, radial O2 loss, solute accumulation in the xylem, root hydraulic conductivity, root elongation and root tip survival. In aerated roots, the stele respired about 6 times faster than the cortex on a volume basis. Respiratory O2 consumption decreased substantially with distance from the root apex and at 300–500 mm it was 80% lower than at the apex. Respiration of lateral roots constituted a sink for O2 supplied via aerenchyma, and reduced O2 flow towards the tip of the supporting root. Stelar anoxia could be induced either by lowering the O2 partial pressure in the bathing medium from 21 to 4 kPa (excised roots) or, in the case of intact roots, by reducing the O2 concentration around the shoot. The root hair zone sometimes extended to 1.0 mm from the root surface and contributed up to a 60% drop in O2 concentration from a free-flowing aerated solution to the root surface. There was a steep decline in O2 concentration across the epidermal-hypodermal cylinder and some evidence of a decline in the O2 permeability of the epidermal-hypodermal cylinder with increasing distance from the root apex. The differences in O2 concentration between cortex and stele were smaller than reported for maize and possibly indicated a substantial transfer rate of dissolved O2 from cortex to stele in banana, mediated by a convective water flow component. An O2 partial pressure of 4 kPa in the medium reduced net nutrient transfer into the vascular tissue in the stele within 1 or 2 h. Hypoxia also caused a temporary decrease in radial root hydraulic conductivity by an order of magnitude. In O2 deficient environments, the stele would be among the first tissues to suffer anoxia and O2 consumption within the root hair zone might be a major contributor to root anoxia/hypoxia in banana growing in temporarily flooded soils.  相似文献

The present study evaluated waterlogging tolerance, root porosityand radial O2 loss (ROL) from the adventitious roots, of sevenupland, three paddy, and two deep-water genotypes of rice (Oryzasativa L.). Upland types, with the exception of one genotype,were as tolerant of 30 d soil waterlogging as the paddyand deep-water types. In all but one of the 12 genotypes, thenumber of adventitious roots per stem increased for plants grownin waterlogged, compared with drained, soil. When grown in stagnantdeoxygenated nutrient solution, genotypic variation was evidentfor root porosity and rates of ROL, but there was no overalldifference between plants from the three cultural types. Adventitiousroot porosity increased from 20–26 % for plants grownin aerated solution to 29–41 % for plants grown instagnant solution. Growth in stagnant solution also induceda ‘tight’ barrier to ROL in the basal regions ofadventitious roots of five of the seven upland types, all threepaddy types, and the two deep-water types. The enhanced porosityprovided a low resistance pathway for O2 movement to the roottip, and the barrier to ROL in basal zones would have furtherenhanced longitudinal O2 diffusion towards the apex, by diminishinglosses to the rhizosphere. The plasticity in root physiology,as described above, presumably contributes to the ability ofrice to grow in diverse environments that differ markedly insoil waterlogging, such as drained upland soils as well as waterloggedpaddy fields.  相似文献

Abstract

The Schöningen 13II-4 ‘Spear Horizon’ site is famous for the excellent preservation of 300,000-year-old Palaeolithic hunting weapons, including nine wooden spears and a lance, deposited on the shores of a former interglacial lake in association with a large assemblage of well-preserved and butchered animal bones, mainly from horse. Some bones show distinct areas of dark staining, thought to be derived from contact with decaying plant remains along the shores of the lake. It was decided to test this theory and try to determine experimentally where bone staining was most likely to occur on the littoral zone. Modern horse and cow bones were fastened along parallel transects at two locations and the installations were left for several months. Black stains appeared on some bones in the shallows, but not on bones deposited on permanently dry land or in deeper water. Within the 10 m wide band of bones in the main concentration at the Schöningen site, there is a high incidence of bone staining, indicating accumulation of finds along a shallow lake margin. By using GIS, additional clusters of stained bones in the eastern part of the site were revealed and may indicate shorelines when water levels in the lake were lower.  相似文献
