The stationary radial volume flows across maize (Zea mays L.) root segments without steles (sleeves) were measured under isobaric conditions. The driving force of the volume flow is an osmotic difference between the internal and external compartment of the root preparations. It is generated by differences in the concentrations of sucrose, raffinose or polyethylene glycol. The flows are linear functions of the corresponding osmotic differences ( ) up to osmotic values which cause plasmolysis. The straight lines obtained pass through the origin. No asymmetry of the osmotic barrier could be detected within the range of driving forces applied ( =±0.5 MPa), corresponding to volume-flow densities of jv, s=±7·10–8 m·s–1. Using the literature values for the reflection coefficients of sucrose and polyethylene glycol in intact roots (E. Steudle et al. (1987) Plant Physiol.84, 1220–1234), values for the sleeve hydraulic conductivity of about 1·10–7 m·s–1 MPa–1 were calculated. Erythrina lectins possess similar structural and carbohydrate binding properties. Recently, tri- and tetra-antennary complex type carbohydrates with non-reducing terminal galactose residues have been shown to be precipitated as tri- and tetravalent ligands, respectively, with certainErythrina lectins [Bhattacharyya L, Haraldsson M, Brewer CF (1988) Biochemistry 271034-41]. The present work describes a comparative study of the binding and precipitating activities of fourErythrina lectins,viz. E. corallodendron, E. cristagalli, E. flabelliformis, andE. indica, with multi-antennary complex type carbohydrates and synthetic cluster glycosides. The results show that though their binding affinities are very similar, theErythrina lectins show large differences in their precipitating activities with the carbohydrates. The results also indicate significant dependence of the precipitating activities of the lectins on the core structure of the carbohydrates. Abstract Water storage and nocturnal increases in osmotic pressure affect the water relations of the desert succulent Ferocactus acanthodes, which was studied using an electrical circuit analog based on the anatomy and morphology of a representative individual. Transpiration rates and osmotic pressures over a 24-h period were used as input variables. The model predicted water potential, turgor pressure and water flow for various tissues. Plant capacitances, storage resistances and nocturnal increases in osmotic pressure were varied to determine their role in the water relations of this dicotyledonous succulent. Water coming from storage tissues contributed about one-third of the water transpired at night: the majority of this water came from the nonphotosynthetic, water storage parenchyma of the stem. Time lags of 4 h were predicted between maximum transpiration and maximum water uptake from the soil. Varying the capacitance of the plant caused proportional changes in osmotically driven water movement but changes in storage resistance had only minor effects. Turgor pressure in the chlorenchyma depended on osmotic pressure, but was fairly insensitive to doubling or halving of the capacitance or storage resistance of the plant. Water uptake from the soil was only slightly affected by osmotic pressure changes in the chlorenchyma. For this stem succulent, the movement of water from the chlorenchyma to the xylem and the internal redistribution of water among stem tissues were dominated by nocturnal changes in chlorenchyma osmotic pressure, not by transpiration.  相似文献

Abstract. Two models of the relationship between diurnal variation in shoot water potential and transpiration in 14-year-old Picea sitchensis (Bong.) Carr. were compared. The first model was a physiologically based resistance-capacitance (R-C) analogue with its associated differential equations. The second was a non-physiological discrete-difference (D-D) or stochastic transfer function model. The RC model included only the effect of water storage in the phloem and bark while the D-D model implicity included all storage mechanisms. The R-C and D-D models explained similar fractions (62% and 68% respectively) of the variation in shoot water potential due to diurnal changes in transpiration rate. However, the D-D model had fewer parameters than the R-C model. The results from the D-D model showed that the resistance to flow from soil to shoots along the trunk, (RT), was 5 × 103 MPa kg-1s and the capacitance of the phloem and bark treated as a single store, (Cs), was 1.6 kg MPa-1. It is suggested that the resistance to flow into storage (Rs) is much greater than RT and can be disregarded. A non-linear version of the D-D model suggested [hat resistance to flow in the trunk increases with increasing transpiration rate.  相似文献

Spatial variations in soil processes regulating mineral N losses to streams were studied in a small watershed near Toronto, Ontario. Annual net N mineralization in the 0–8 cm soil was measured in adjacent upland and riparian forest stands using in situ soil incubations from April 1985 to 1987. Mean annual rates of soil N mineralization and nitrification were higher in a maple soil (93.8 and 87.0 kg.ha–1) than in a pine soil (23.3 and 8.2 kg.ha–1 ). Very low mean rates of mineralization (3.3 kg.ha–1) and nitrification (3.4 kg.ha–1) were found in a riparian hemlock stand. Average NO3-N concentrations in soil solutions were 0.3–1.0 mg.L–1 in the maple stand and >0.06mg.L–1 in the pine stand. Concentrations of NO3–N in shallow ground water and stream water were 3–4× greater in a maple subwatershed than in a pine subwatershed. Rapid N uptake by vegetation was an important mechanism reducing solution losses of NO3–N in the maple stand. Low rates of nitrification were mainly responsible for negligible NO3–N solution losses in the pine stand.  相似文献

Helicopters can be used to collect water samples from many lakes over a wide geographic area within a relatively short time period. Here we report the results from an experiment in which sequential water samples from a lake were collected first from a nonmotorized boat and then immediately afterward from a helicopter. No significant differences were found between the means of the measurement of 20 chemical parameters for the two methods of collection. When compared to obtaining samples from a boat, collection of samples from a helicopter platform had no effect on the content of the water samples.  相似文献

The increased tightening of controls on industrial and municipal wastewater discharges has resulted in steady improvements in the quality of many important rivers over recent years. However, episodic pollution, particularly from farm wastes and combined sewer overflows continues to pose a major problem, and is one of the main causes of poor quality rivers today. Despite our acknowledgement of this continuing problem, very little is known of the mechanistic basis of responses and recovery of aquatic organisms and communities exposed to intermittent pulses of common pollutants. The majority of ecotoxicological studies to date have been concerned with the effects of continuous exposure. Although such studies may provide a means of predicting the impact of episodic pollution events, a more appropriate test design would be to assess toxicity under pulsed and fluctuating exposure. Studies should also include a post-exposure observation period and should consider recovery of individuals and communities. This paper reviews the results of reported studies relevant to the investigation of episodic pollution and pays particular attention to the effects of magnitude, duration and frequency of exposure. Results of field investigations using an in situ bioassay technique are also presented to emphasize the importance of field validation of proposed water quality criteria for intermittent pollution events.  相似文献

The geographical distribution, morphometry and water quality of lakes within large calderas (> 2 km in diameter) were evaluated through a review of the literature and maps. Eighty-eight lakes in 75 calderas were located in 31 volcanic subregions. As a group, the lakes varied greatly in elevation, surface area, maximum depth, and shoreline development. The average surface area was 16.9 km2, surface elevation 873 m, depth 151.1 m, and shoreline development 1.35. Water quality ranged from ultraoligotrophic to highly eutrophic. None of the lakes had an inlet that originated outside the calderas. Most lakes did not have a surface outlet, were circular or subcircular in shape, and covered only parts of the caldera basins. Water clarity in some lakes was among the highest recorded for freshwater systems, but there are indications of possible declining clarity in some cases. Secondary volcanic activity, such as primary (hydrothermal) water and eruptions, has been associated with deteriorated water quality conditions in some lakes.  相似文献

Changes in epilithic algal communities colonizing introduced substrata were determined in a stream polluted with oil refinery effluent at Digboi (Assam, India). The number of algal taxa was reduced but the growth of blue-green algae, particularly two species ofOscillatoria, was encouraged. Epilithic biomass (as chlorophylla) also declined at polluted stations. The algal community of the upstream station was markedly different from the community occurring just after the confluence of effluent; however, the differences were gradually reduced downstream, indicating improvement in water quality. Of the various criteria tested for possible relationships with the level of pollutants, species richness, Shannon diversity and biomass showed significant relationships. The study demonstrates the usefulness of algal criteria for monitoring oil pollution in running waters.  相似文献

Summary Knowledge of the metabolic changes that occur in insulin-resistant type 2 diabetes is relatively lacking compared to insulin-deficient type 1 diabetes. This paper summarizes the importance of the C57BL/KsJ-db/db mouse as a model of type 2 diabetes, and illustrates the effects that insulin-deficient and insulin-resistant states have on hepatic glycogen metabolism. A longitudinal study of db/db mice of ages 2–15 weeks revealed that significant changes in certain parameters of hepatic glycogen metabolism occur during this period. The liver glycogen levels were similar between diabetic and control mice. However, glycogen particles from db/db mice were on average smaller in mass and had shorter exterior and interior chain lengths. Total phosphorylase and phosphorylase a activities were elevated in the genetically diabetic mice. This was primarily due to an increase in the amount of enzymic protein apparently the result of a decreased rate of degradation. It was not possible to find a consistent alteration in glycogen synthase activity in the db/db mice. Glycogen synthase and phosphorylase from diabetic liver revealed some changes in kinetic properties in the form of a decrease in Vmax, and altered sensitivity to inhibitors like ATP. The altered glycogen structure in db/db mice may have contributed to changes in the activities and properties of glycogen synthase and phosphorylase. The exact role played by hormones (insulin and glucagon) in these changes is not clear but further studies should reveal their contributions. The db/db mouse provides a good model for type 2 diabetes and for fluctuating insulin and glucagon ratios. Its use should clarify the regulation of hepatic glycogen metabolism and other metabolic processes known to be controlled by these hormones. The other animal models of type 2 diabetes, ob/ob mouse and fatty Zucker (fa/fa) rat, show similar impairment of hepatic glycogen metabolism. The concentrations of glycogen metabolizing enzymes are high and in vitro studies indicate enhanced rate of glycogen synthesis and breakdown. However, streptozotocin-induced diabetic animals and BB rats which resemble insulin-deficient type 1 diabetes are characterized by decreased glycogen turnover as a result of reduction in the levels of glycogen metabolizing enzymes.  相似文献
