The present study deals with the topography of retinal ganglion cells (GCs) and spatial resolution in the smelt Hypomesus japonicus. The eyes and retinae were examined by light microscopy and computerized tomography. DAPI labelling was used to visualize cell nuclei in the ganglion cell and inner plexiform layers. Two zones of increased GC density in the nasal and temporal retina were bridged by a horizontal streak with the GC density ranging from 5600 to 8000 cells/mm2. The maximum cell density (area retinae temporalis) ranged from 9492 to 14,112 cells/mm2, and the total number of GCs varied from 286 x 103 to 326 x 103 cells in three individuals. The theoretical anatomical spatial resolution (the anatomical estimate of the upper limit of visual acuity) was minimum in the ventral periphery (smaller fish, 1.43 cpd; larger fish, 1.37 cpd) and maximum in area retinae temporalis (smaller fish, 2.83 cpd; larger fish, 2.41 cpd). The relatively high density of GCs and presence of the horizontal streak and area retinae temporalis in the H. japonicus are consistent with its highly visual behaviour. The present findings contribute to better understanding of the factors affecting the topography of retinal ganglion cells and mechanisms of visual adaptation in fish.  相似文献

A new fluorescent zinc (II) complex-based probe 1 encompassing a Schiff's base (E)-2-methoxy-6-((2-[5-nitropyridin-2-ylamino]ethylimino)methyl)phenol ( HL ) was designed, synthesized, and used for the highly selective detection of Cu2+. Ligand HL and complex 1 were characterized using various spectroscopic techniques such as 1H, 13C-NMR, and FTIR spectroscopy, high-resolution mass spectronomy (HRMS), UV/visible light spectroscopy, and fluorescence studies. Ligand HL did not exhibit any considerable change in fluorescence in the presence of various cations. Notably, its Zn(II) complex 1 exhibited highly selective ‘TURN-OFF’ fluorescence signalling towards Cu2+ that remained uninterrupted with competing analytes. Probe 1 interacted with Cu2+ in 1:2 (1:Cu2+) stoichiometry as estimated through a Job's plot. Moreover, the selectivity of 1 was further confirmed through the interaction of the 1 + Cu2+ complex with some possible interfering metal ions inducing an insignificant response. Additionally, the association and quenching constant were determined to be 3.30 × 104 M−1 and 0.21 × 105 M−1 through the Benesi–Hildebrand method and Stern–Volmer plot, respectively.  相似文献

Salvia miltiorrhiza Bunge is a natural drug for treating myocardial infarction (MI). However, the targets and mechanisms of S. miltiorrhiza Bunge in the treatment of MI are yet to be elucidated. Traditional Chinese medicine systems pharmacology (TCMSP) data were used to screen out chemical constituents, and UniProt was used to predict relevant targets. Disease targets were obtained from the Online Mendelian Inheritance in Man and GeneCards databases. We used the STRING platform to build a protein–protein interaction network and used Cytoscape_v3.8.1 software to make a Drug–Ingredients–Gene Symbols–Disease network map. The Metascape database was used to perform gene ontology and Kyoto Encyclopaedia of Genes and Genomes (KEGG) enrichment analyses for drug–disease overlapping gene symbols. The targets identified by network pharmacology were further verified by in vitro and in vivo experiments. Seventy-five active components of S. miltiorrhiza Bunge were obtained from the TCMSP database, while 370 disease targets and 29 cross-targets were obtained from the Genecards database. The KEGG pathway enrichment results suggested that the mechanism of S. miltiorrhiza Bunge in the treatment of MI was significantly related to the VEGF signalling pathway. In vitro and in vivo experiments were used to evaluate the reliability of some important active ingredients and targets. S. miltiorrhiza Bunge alleviated the damage to cardiac function, attenuated myocardial fibrosis and protected endothelial cell function by increasing the expression of TGF-β and VEGFA. S. miltiorrhiza Bunge showed the therapeutic effect of MI by promoting the expression of VEGFA signalling pathway, providing a reliable basis for exploring herbal treatment of MI.  相似文献

The length of the time young gypsy moth, Lymantria dispar L. (Lepidoptera: Lymantriidae), larvae remain in litter after failing from trees would help in understanding the impact of predation by ants, as ants appear to be important predators of gypsy moths only in litter. To obtain information on caterpillar litter residence times, visual activity of larvae was investigated in the laboratory by presenting them with vertical dark silhouettes of various widths in a 1 m wide cylindrical arena. Their limit of angular resolution was about 1°. This value helped to determine distances to trees caterpillars could be expected to orient toward from randomly-located points in a forest. These data were used with results on litter crawling speed of instars 1, 2, and 3 in a simulation model that generated litter residence times averaging serveral hours. By combining results with a previously-developed model of ant predation in forest litter, it was estimated that about half of unfed first instars falling into the litter would survive, whereas only about 10% of 2nd and 3rd instars would. The implications these results have for control of the gypsy moth are discussed.
Résumé La prédation par les fourmis du genre Formica spp. a une influence importante sur les premiers stades de L. dispar (Lepido., Lymantriidae) en Amérique du Nord. La durée du temps passé dans la litière par les jeunes chenilles après être tombées des arbres doit permettre de comprendre l'impact de la prédation par les fourmis. Pour connaître le temps de séjour des chenilles dans la litière, l'acuité visuelle des chenilles a été examinée au laboratoire en leur présentant des silhouettes noires verticales de différentes largeurs dans une arène circulaire de 1 m de diamètre. La limite de leur angle de résolution était d'environ 1°. Cette valeur a été utilisée pour déterminer les distances (jusqu'à 5 m) quelles que soient les orientations dans la forêt, d'arbres supposés susceptibles d'orienter les chenilles. Les arbres aux silhouettes acceptables étaient distribués uniformément entre 1 et 5 m.Les vitesses de déplacement dans la litière des stades 1, 2, et 3 ont été mesurées par observation directe dans la forêt (elles variaient de 0.9 à 1,54 cm/min). La distance des arbres et la vitesse de déplacement on servi à construire un modèle de simulation qui a donné un temps moyen de séjour de plusieurs heures. En combinant ces résultats avec un modèle précédemment publié de la prédation par les fourmis dans la litière de la forêt, qui utilisait le temps de contact des chenilles avec les fourmis et les taux de captures victorieuses pour estimer la mortalité, nous avons déterminé que la moitié environ des chenilles non alimentées de premier stade tombées dans la litière pourraient survivre, tandis que la survie pour le second et le troisième stade ne serait que de 10%. La discussion porte sur l'importance de ces résultats dans la lutte contre L. dispar.

嗅觉器官是鱼类的一种重要感觉器官,它感受由化学而引起的刺激。各种鱼类的感觉器官是十分不同的,不少学者研究了它的构造及其生理机能。以往对鲀形目鱼的嗅觉器官构造的研究报道甚少,大多注意了它的外鼻孔的形态变化,仅在少数文章(Hora,1975)提到鲀形目鱼的嗅觉器官极不发达。为了弄清它的构造特点,解剖观察了中国鲀形目代表性种类的嗅觉器官,对它们的构造特征进行了比较研究,发现它们在各不同分类阶元有独特的构造。  相似文献

The aim of this work was to define the metabolic factors which regulate the respiratory pathways in trangenic potato tubers. We previously found that respiration is enhanced in transgenic tubers which express a yeast invertase and a glucokinase from Zymomonas mobilis . In this study we investigated glycolysis in three further transgenic potato lines with profound changes in the mobilization of sucrose. We studied antisense ADPglucose pyrophosphorylase lines which are characterized by a reduction in starch accumulation and a significant build up of sucrose and related metabolic intermediates. We also report the generation of two novel double transgenic lines where the yeast invertase is expressed specifically in tubers of the ADPglucose pyrophosphorylase antisense line, targeted to either the cytosol or apopolast. We evaluated whether the localization of sucrose cleavage had an impact on the glycolytic induction, and assessed if invertase expression in the high-sucrose background had any further effects on glycolysis. We found that induction of the glycolytic enzymes only occurs when the invertase is targeted to the cytosol, and that the extent of this induction was comparable in the wild type and antisenseADPglucose pyrophosphorylase backgrounds. We conclude that the signal regulating glycolysis is directly linked to cytosolic sucrose hydrolysis.  相似文献

Simple cells in mammalian visual cortex are quasi-linear mechanisms whose behavior departs from true linearity in a very consistent manner. Empirical research on direction selectivity (DS) clearly illustrates these characteristics. A linear DS cell will be DS for all stimuli, whereas a linear non-DS cell will not be DS for any stimuli. However, many simple cells have opposite preferred directions for stimuli of reversed polarity, and some cells are DS for some stimuli (e.g., moving bars) but not for others (e.g., drifting gratings). Also, linear non-DS cells must have separable spatiotemporal receptive fields (RFs), and linear DS cells must have inseparable RFs. Yet many actual DS cells have separable RFs. Here we present a nonlinear model of simple-cell behavior that reproduces all of these empirical behaviors. The model is a variant of the current linear model, amended to include an interleaved nonlinearity (half-wave rectification) that allows it to mimic the (im)balance of push-pull mechanisms. We present simulation results showing that balanced push-pull mechanisms result in linear behavior, while imbalanced push-pull arrangements produce all of the incongruent DS-related behaviors that have been reported for simple cells.  相似文献
