In the angelfish ( Pterophyllum scalare scalare ) numerous rodlet cells were found in the large post-orbital blood vessel caudal to the eye and in the surrounding extravascular space. Within the vessel the rodlet cells formed striking regular arrays, along the inner aspect of the wall. The rodlets within the cells were positive to PAS but negative to Sudan Black B, Masson's, and the Fuelgen stain. The capsule around the cells was negative for all these stains. These rodlet cells appeared to be traversing the vessel endothelium, and to be pushing the endothelium aside without damaging it. Some discharged their contents into the vessel, but we never observed the release of intact rodlets. The nuclei of rodlet cells in actual contact with the vessel were at the end of the cell more distant from the endothelial wall. Cell-to-cell adhesion structures or communications junctions between rodlet cells and the endothelium were not evident. A putative rodlet cell precursor in the extravascular space contained large electron-dense granules, and extended pseudopodia that contacted nearby rodlet cells. Based on their morphology, tissue distribution, and their behaviour, we conclude that the rodlet cell is an endogeneous teleost cell type, and possibly represents a form of matured granulocyte.  相似文献

Thirty-two partial phytochrome sequences from algae, mosses, ferns, gymnosperms, and angiosperms (11 of them newly released ones from our laboratory) were analyzed by distance and characterstate approaches (PHYLIP, TREECON, PAUP). In addition, 12 full-length sequences were analyzed. Despite low bootstrap values at individual internal nodes, the inferred trees (neighbor joining, Fitch, maximum parsimony) generally showed similar branching orders consistent with other molecular data. Lower plants formed two distinct groups. One basal group consisted of Selaginella, Equisetum, and mosses; the other consisted of a monophyletic cluster of frond-bearing pteridophytes. Psilotum was a member of the latter group and hence perhaps was not, as sometimes suggested, a close relative of the first vascular plants. The results further suggest that phytochrome gene duplication giving rise to a- and b- and later to c-types may have taken place within seedfern genomes. Distance matrices dated the separation of mono- and dicotyledons back to about 260 million years before the present (Myr b.p.) and the separation of Metasequoia and Picea to a fossil record-compatible value of 230 Myr B.P. The Ephedra sequence clustered with the c- or a-type and Metasequoia and Picea sequences clustered with the b-type lineage. The paleoherb Nymphaea branched off from the c-type lineage prior to the divergence of mono- and dicotyledons on the a- and b-type branches. Sequences of Piper (another paleoherb) created problems in that they branched off from different phytochrome lineages at nodes contradicting distance from the inferred trees' origin. The possible use of vanadium compounds in the treatment of diabetic patients is now being evaluated. However, previously to establish the optimal maximum dose for diabetes therapy, it should be taken into account that vanadium is a highly toxic element to man and animals. The toxic effects of vanadium are here reviewed. The tissue vanadium accumulation, which would mean an additional risk of toxicity following prolonged vanadium administration is also discussed. Recently, it has been shown that coadministration of vanadate and TIRON, an effective chelator in the treatment of vanadium intoxication, reduced the tissue accumulation of this element, decreasing the possibility of toxic side effects derived from chronic vanadium administration without diminishing the hypoglycemic effect of vanadium. However, previously to assess the effectiveness of this treatment in diabetic patients, a critical reevaluation of the antidiabetic action of vanadium and its potential toxicity is clearly needed.  相似文献

本文是利用扫描电子显微镜对陆栖恐龙tvrannosaurid与海栖渐增mosasaurid”牙齿结构进行的比较解剖学研究。化石采自加拿大RedDeerhiverValley上白里统Horse-shoeCanyon组。通过研究地层中出现的生物化石,特别是动物牙齿的组织结构,可以了解动物为了适应生活环境而发生的进化过程,也可以推测它们的系统发育关系。tyrannosaund与mosas。id都拥有锥状的同形齿,牙齿侧向扁平,且略向后弯曲。研究结果确认了tyrannosaurid的牙齿由于薄层的无柱釉质bPrismaticenamel)向齿质的侵人而造成许多的凹凸构造,此锯齿状构造沿着牙齿的前后缘,由牙齿的顶端分布至基部。因此tyrannosaurid的牙齿呈现着锐利的切缘;在这些凹凸状切缘的沟与小窝的深部可观察到有机物的沉积。但是类似的锯齿状构造只能在齿冠呈钝圆状的mosasaurid牙齿的基部附近观察到。我们以扫描电子显微镜(SEM)检索,确认两爬行类的齿质皆是属于中间型的真性齿质(intermediatetypeorthodentine);所谓orthodentine即是细管齿质(tubulardentine)。tyrannosaurid的真性齿质的齿质小管只在齿质一釉质相接处Uentino—enamaljunction)附近放散出规则性的分歧与末枝。但mosasaurid的真性齿质的齿质J。管,在齿质的中间层与表层中,呈现着由复杂  相似文献

Thrombin is a potent mitogen for human vascular smooth muscle cells (HVSMC) and its enzymatic activity is required for this function. The present study demonstrates that prothrombin is also mitogenic for HVSMC due to the generation of enzymatically active thrombin which occurs upon incubation of prothrombin with the cells. Analysis by SDS-PAGE, immunoblotting, and amino acid sequencing revealed that prothrombin incubated with HVSMC undergoes limited proteolysis. Prethrombin 1 was formed through cleavage at R155-S156. Cleavage at R271-T272 generated fragment 1.2 and prethrombin 2 whilst cleavage at R284-T285 yielded truncated prothrombin 2 (prethrombin 2′). However, cleavage at R320-I321 which, during prothrombin activation produces two-chain α-thrombin, was not detectable. Studies on HVSMC-conditioned medium revealed that a similar pattern of prothrombin cleavage occurred by a cell-secreted factor(s). Amidolytic activity analysis indicated that 1–3% catalytically active thrombin-like activity was generated upon incubation of prothrombin with HVSMC-conditioned medium. By treating conditioned medium with various classes of proteinase inhibitors or hirudin, it was determined that prothrombin is cleaved by a cell-derived serine proteinase-like factor(s) at R271-S272 and by α-thrombin at R155-S156 and R284-T285. Antibodies neutralising the activity of either urokinase, tissue plasminogen activator, or factor Xa failed to alter the prothrombin cleaving activity of conditioned medium. This activity which may catalyse an alternative pathway for the generation of thrombin, was eluted from a gel filtration column as a single peak with apparent molecular mass of 30–40 kDa. © 1995 Wiley-Liss, Inc.  相似文献

A procedure for the immunoassay of cohydrolase sphingolipid-I in mouse tissue is described. This cohydrolase (actually a mixture of at least four related proteins) stimulates or activates the beta-glucosidase which hydrolyzes ceramide glucoside, a widely occurring glycosphingolipid. The method involves extraction of cohydrolase from tissue homogenate with a salt-buffer solution, removal of proteins by adjustment to pH 6, further removal of proteins by heating, and removal of interfering materials with a small size exclusion column. Antibodies were raised to bovine cohydrolase in rabbits and purified with an affinity column made from cohydrolase. The immunoassay involves binding of antibody by the cohydrolase sample (20-200 pg) in competition with cohydrolase that has been chemically linked to horseradish peroxidase. The mixture is treated with particle-linked second antibody and centrifuged; the pellet is then assayed fluorometrically for peroxidase content. Initial application of the method showed that cohydrolase was present in all mouse tissues studied and that its concentration paralleled that of glucocerebrosidase relatively closely. Changes with age (14 and 92 days) occurred in a similar fashion for the two substances.  相似文献

在15只麻醉开胸犬中,用在体器官恒流灌注法研究了骨胳肌、皮肤、肾和心脏的血管在5-HT 诱发的主动脉区化学感受性升血压反射中的反应,同时还观察了5-HT 对这些器官血管的直接效应。左心房注入5-HT(200μg)后,在动脉血压急剧上升的同时,股薄肌、皮肤(后爪)和肾脏的灌流压明显增加,其增值分别为43±8.47±5和56±27mmHg(P<0.01),而冠脉灌流压无明显改变。用阿托品和/或心得安阻断骨胳肌、皮肤和心脏灌流区域的 m 和同β-受体后,上述效应无进一步变动。向灌流环路直接注入5-HT(10—50μg),可引起皮肤和肾脏的血管收缩,而股薄肌和心肌的血管则扩张;后一效应不被心得安阻断。以上结果提示:1.在5-HT 诱发的主动脉区化学感受性升血压反射中,骨胳肌、皮肤和肾脏呈现明显的交感性缩血管反应,而并不伴有交感性扩血管活动;2.在外周血管可能存在两种不同的5-HT 受体,一种主要分布在皮肤和肾脏,引起缩血管效应;另一种主要分布于骨胳肌和冠状血管,引起扩血管效应。  相似文献

在15只戊巴比妥钠麻醉的开胸狗身上观察高渗溶液对血流动力学的影响,主要结果如下:1.静脉内注射50%葡萄糖溶液(3m1/kg,15s 内注毕),规律地引起心动徐缓、动脉血压降低、左心室(-dp/dt)减小、左心室 dp/dt 和心输出量增加,以及肾和股薄肌的血流阻力降低。25%甘露醇溶液具有类似作用。2.切断两侧颈迷走神经后,注射高渗溶液不再能诱发动脉低血压以及肾和股薄肌血流阻力的反射性降低,提示此类效应的传入通路主要为迷走神经。3.在切断迷走神经后注射高渗溶液,还使左心室 dp/dt 进一步增加,表明高渗溶液增强心肌收缩性。根据以上结果似可认为,静脉注射高渗溶液所致动脉血压降低,实质上反映着心输出量的增加不足以抵销外周阻力的减小。  相似文献

本文是应用模糊聚类分析方法研究中国散生竹类分类的一次尝试。分类特征采用了竹秆上、中、下三段各类型维管束数,方法上使用了模糊(Fuzzy)直接聚类分析进行综合分析。经电子计算机运算后,不仅取得了与传统分类基本一致的分类结果,同时也表明这种方法较之其它一些植物数量分类方法简便易行,此外还讨论了一些中国散生竹类分类上的问题。  相似文献
