The very large acrosome of Pteropus species spermatozoa is prone to damage during cooling procedures. Cryogenic succuss has been linked to membrane composition, therefore the lipid composition of five Pteropus species sperm acrosomal and plasma membranes were investigated to provide insight into reasons for cold shock susceptibility. Rapid chilling and re-warming of spermatozoa from three Pteropus species resulted in a decrease (< 0.05) in acrosomal integrity. Biochemical analysis of lipids revealed that stearic acid (18:0) was the predominant saturated fatty acid and oleic acid (18:1, n-9) the predominant unsaturated fatty acid in both acrosomal and plasma membranes. Linolenic acid (18:3, n-3) was only detected in plasma membranes of Pteropus hypomelanus and was detected in acrosomal membranes of all Pteropus spp. studied (except Pteropus giganteus). Although detected in both plasma and acrosomal membranes of Pteropus vampyrus, docosahexaenoic acid (22:6) was not detected at all in Pteropus poliocephalus, only in trace levels in the acrosomal and plasma membranes of P. giganteus and P. hypomelanus and not in acrosomal membranes of Pteropus rodricensis. No difference was seen in the levels of polyunsaturated fatty acids (PUFAs) within plasma membranes, however PUFAs were lower (< 0.05) in acrosomal membranes of P. giganteus compared with P. vampyrus. Pteropus spp. spermatozoa have a very low ratio of unsaturated/saturated membrane fatty acids (<0.5). Membranes containing more PUFAs are more fluid, so the use of cryogenic media which improves membrane fluidity should improve Pteropus spp. spermatozoal viability post-thaw.  相似文献

Effective contraception would enhance genetic management of captive Pteropus species, which typically breed well in captivity. Male reproductive seasonality was monitored (15-mo interval) in captive P. alecto (6 controls and 5 treated with 4.7 mg deslorelin). In untreated males, there were seasonal changes in testicular volume, body weight and testosterone secretion; testicular volume and body weight peaked in February and March, respectively, whereas testosterone concentration remained >5 ng/ml before rising (P < 0.001) to 24.9 ± 3.6 ng/ml (mean ± SEM) in April. However, there was no corresponding change in sperm quality, and seminal vesicle gland (SVG) secretions remained present in ejaculates. In treated males, testosterone concentration had an initial ‘flare’ response (mean ± SEM peak: 19.95 ± 3.27 ng/ml) before declining (P < 0.001) by 32 d to basal levels, where it remained. In these males, there was reduced sperm motility after 1 mo (P < 0.001) and the absence of SVG secretions after 4 mo. However, aspermic ejaculates were first recorded 5 mo post-treatment. At 10 mo after treatment, spermatogenesis was still disrupted, when membrane-intact, but non-motile sperm were present in two individuals. Motile sperm were first recovered from one of these males 13 mo after deslorelin treatment. We concluded that captive P. alecto males: (a) had seasonal reproductive changes in testicular volume, body weight and testosterone secretion; (b) produced motile, membrane-intact sperm and SVG secretions throughout the year; and (c) had a rapid decline in testosterone concentration and consequent suppression of testicular function for at least 5 mo following deslorelin administration.  相似文献

The long‐tongued nectar bat Macroglossus minimus lagochilus (Megachiroptera, Pteropodidae) is a small fruit bat inhabiting South‐east Asia. Its reproduction was studied in Brunei, Borneo, where individuals were usually captured in rainforest (Ulu Temburong). Adult females (n = 23) had body masses of 13.0–19.0 g, with forelimbs 3.83–4.33 cm in length. All adult females were pregnant, with pregnancies observed during every month except May, September and November when no adult females were captured. Female M. minimus in Brunei have a reproductive cycle that is consistent with a pattern of seasonal bimodal polyoestry. Most births appear to occur from November to May, during which time females experience two births: the first centred around November and December, and the second centred around April and May. These two birth seasons appear correlated to the two seasons of greater rainfall in Brunei: October to December, and April to July. Oestrous and mating apparently occur soon after each birth; however, there appears to be a 2‐ to 3‐month period of delayed embryonic development following the births centred around April and May. The data are consistent with embryonic development commencing around July and August (after a delay) for the births centred around November and December, and around December and January for the births centred around April and May. The data suggest that all adult females participate in each breeding season. Adult males (n = 20) had body masses of 12.5–18.0 g, with forelimbs 3.91–4.26 cm in length.  相似文献

Cypridoidean ostracods are one of a number of animal taxa that reproduce with giant sperm, up to 10 000 µm in length, but they are the only group to have aflagellate, filamentous giant sperm. The evolution and function of this highly unusual feature of reproduction with giant sperm are currently unknown. The hypothesis of long-term evolutionary persistence of this kind of reproduction has never been tested. We here report giant sperm discovered by propagation phase contrast X-ray synchrotron micro- and nanotomography, preserved in five Miocene ostracod specimens from Queensland, Australia. The specimens belong to the species Heterocypris collaris Matzke-Karasz et al. 2013 (one male and three females) and Newnhamia mckenziana Matzke-Karasz et al. 2013 (one female). The sperm are not only the oldest petrified gametes on record, but include three-dimensional subcellular preservation. We provide direct evidence that giant sperm have been a feature of this taxon for at least 16 Myr and provide an additional criterion (i.e. longevity) to test hypotheses relating to origin and function of giant sperm in the animal kingdom. We further argue that the highly resistant, most probably chitinous coats of giant ostracod sperm may play a role in delaying decay processes, favouring early mineralization of soft tissue.  相似文献

We evaluated the sterilizing effect of 3-monochloropropane-1, 2-diol (3-MCPD) in male Egyptian fruit bats (Rousettus aegyptiacus). We used three groups. One was treated with 70 mg/kg 3-MCPD for 4 days. The second group was treated with 3-MCPD as a bait formulation (known concentration of 3-MCPD mixed with a known amount of food). The third group was untreated controls. We compared the weights of the reproductive organs, histology of the testes, occurrence of spermatogenesis, and the count, motility and abnormalities of epididymal sperm of treated males with those of the untreated control group. 3-MCPD caused significantly decreased weights of reproductive organs, several testicular histological alterations and spermatogenic arrest accompanied by significant decreases in sperm count and motility, and significantly increased number of abnormal sperm. 3-MCPD bait was readily accepted by the animals. 3-MCPD, even in low doses and after limited exposure, disrupted spermatogenesis in males of the Egyptian fruit bat. Our findings have potential value for public health and agricultural authorities, and for vertebrate pest managers. 3-MCPD may have application for control of this pest.  相似文献

ABSTRACT. The auditory characteristics of two populations (laboratory reared and wild) of North American gypsy moths (Lymantriidae: Lymantria dispar L.) were sampled and the neurally derived thresholds of wild males and females to frequencies from 5 to 150 kHz compared. The noctuoid auditory receptors, Al and A2-cell, and putative proprioceptor, B-cell, were identified. Both sexes possess neurally responsive ears but females exhibit median best frequencies significantly lower than those of males. Audiogram comparisons reveal significantly different thresholds at 5–15 kHz, 30–120 kHz and 130–140 kHz, with females less sensitive to all but the lowest frequencies. Wild male populations reveal less audiogram variability than laboratory-reared individuals, while females' tuning curves appear more similar. The high variability present in colony moths warrants caution in the use of laboratory-reared insects for studies that assume natural levels of selection pressure. We suggest that male L. dispar possess adaptively functional ears tuned to the frequencies in the echo-location signals of bats but that the flightless females of this species are not exposed to bat predation and therefore possess ears in a state of evolutionary degeneration.  相似文献

A preliminary analysis of the use by bats of National Trust properties revealed that all 14 resident species have been recorded. Comparison with a national database of records for Britain managed by English Nature (NCC Bat database) showed that the scarcer species are better represented roosting in buildings on Trust properties than in other buildings. In addition, survey of Trust properties has shown that a high proportion of buildings are used by bats. Bats provide unusual wildlife links between buildings, gardens and countryside through which the Trust can promote nature conservation, and resolve conflicts between building or landscape restoration and the bat habitat requirements. The role of the County Bat Groups in survey and promotion of bats on Trust properties is becoming increasingly important, especially if we are able to identify key properties for bats.  相似文献
