The use of bifunctional reagents to form cross-links between subunits in protein oligomers and subsequent disruption of noncovalent interactions with SDS allows comment upon the number of subunits and the symmetry in the original assembly. In existing treatments the number of equations needed to describe theoretically the proportions of all the cross-linked species that can be formed as a function of time in this way makes the analysis of the system unmanageable for proteins with more than four subunits. A method is presented that allows the required equations for any oligomer to be formulated as an algorithm suitable for solution by computer. Its application is illustrated with reference to experimental results obtained with two protein hexamers, Jasus hemocyanin and alpha-urease from jack bean.  相似文献

For microtubule assembly, the data reported here support an initial nucleation phase followed by a growth or elongation phase. The nucleation phase was not detected kinetically. Evidence for this step was given by the existence of the critical concentration and the dependence of the number of microtubules on oligomer concentration.Kinetic evidence indicated the existence of two consecutive steps in the growth phase of microtubules. The fast process increased and the slow one decreased with the concentration of microtubule protein. Similar kinetics were found upon recombination of tubulin oligomer and dimer which had been resolved by agarose chromatography. The fast process increased with oligomer and decreased with dimer concentration while the slow one depended positively on dimer concentration. Microtubules were formed when the oligomeric fraction only was employed. In contrast, under identical conditions, no microtubule formation was detected turbidimetrically or by electron microscopy from dimer alone. When dimer caused elongation of seed tubules, there was only one growth step with a rate constant of the same order of magnitude as the slow process for the other experiments.  相似文献

A helical, prehairpin trimer covering the majority of the HR1 region of human immunodeficiency virus gp41 was achieved by chemically coupling three identical 51 amino acid peptides. A 1,3,5‐tris(aminomethyl)‐2,4,6‐triethylbenzene pinwheel ‘cap’ was used to trimerize the peptides by taking advantage of the unique property of triacyl fluoride and orthogonal protection and deprotection. 脂肪酸不仅是细菌细胞膜组分,还是许多生物活性物质的合成原料。不饱和脂肪酸(unsaturated fatty acid, UFA)具有更低的相变温度,是细菌调节细胞膜流动性的重要分子,因此UFA合成途径是重要的抗菌药物筛选靶点。细菌可利用厌氧途径合成UFA,其中模式生物大肠杆菌利用经典的FabA-FabB途径合成UFA,但不同细菌中UFA合成的厌氧途径具有多样性,相关催化酶类也不尽相同;细菌还可以利用需氧途径合成UFA,利用脂肪酸脱饱和酶直接将饱和脂肪酸(saturated fatty acid, SFA)转化为不饱和脂肪酸,而不同脱饱和酶会生成不同结构的UFA,在逆境耐受、致病力等多方面发挥重要作用;细菌还可以利用单加氧酶,将脂肪酸合成途径中癸酰酰基载体蛋白(acyl carrier protein, ACP)转化为顺-3-癸烯酰ACP,并最终合成UFA。细菌脂肪酸合成相关的其他酶类在UFA合成或不同种类UFA调节中也发挥着重要作用。本文系统地总结了细菌UFA合成途径与相关酶类的多样性研究进展,旨在为进一步了解细菌UFA合成机制,并以此为靶点开发抗菌药物等方面提供理论支撑。  相似文献

Lipoxygenases (LOXs) are iron- or manganese-containing oxidative enzymes found in plants, animals, bacteria and fungi. LOXs catalyze the oxidation of polyunsaturated fatty acids to the corresponding highly reactive hydroperoxides. Production of hydroperoxides by LOX can be exploited in different applications such as in bleaching of colored components, modification of lipids originating from different raw materials, production of lipid derived chemicals and production of aroma compounds. Most application research has been carried out using soybean LOX, but currently the use of microbial LOXs has also been reported. Development of LOX composition with high activity by heterologous expression in suitable production hosts would enable full exploitation of the potential of LOX derived reactions in different applications. Here, we review the biological role of LOXs, their heterologous production, as well as potential use in different applications. LOXs may fulfill an important role in the design of processes that are far more environmental friendly than currently used chemical reactions. Difficulties in screening for the optimal enzymes and producing LOX enzymes in sufficient amounts prevent large-scale application so far. With this review, we summarize current knowledge of LOX enzymes and the way in which they can be produced and applied.  相似文献

Variation in traits across species distribution ranges is often indicative of diversifying evolution that can lead to speciation. Of particular interest is whether traits vary clinally or abruptly because the latter pattern can be indicative of incipient speciation. Understanding of intraspecific variation in chemical traits is still in its infancy because studies of population variation have tended to focus on morphology or neutral genetic markers. To address these issues, the composition of cone volatile odours was examined in ten populations of the South African cycad Encephalartos villosus across its range in the Eastern Cape and KwaZulu Natal using headspace sampling and analysis by gas chromatography‐mass spectrometry. Because volatiles play a key role in attracting pollinators to cones of Encephalartos cycads and may thus reflect local adaptation to pollinators, pollinator assemblages were also investigated in the ten populations of E. villosus. Volatile compounds from populations in the north of the distribution range were dominated by unsaturated hydrocarbons, whereas, in the southern populations, nitrogen‐containing compound and terpenoids were the major compounds. A shift between southern and northern populations appeared to occur at the Umtamvuna River, where populations had odour profiles with components of both the northern and southern populations. However, one population in the north (Vernon Crookes Nature Reserve) had a quantitatively similar odour profile to the populations in the extreme south of the range. These results reveal strong interpopulation variation in the cone scent of E. villosus, including variation in the relative emission of dominant compounds that may play key functional role in this pollination system. However, pollinator assemblages did not differ across the different populations, which suggest that these patterns were produced by co‐evolution or drift, rather than by pollinator shifts. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 106 , 514–527.  相似文献

Many proteins need to form oligomers to be functional, so oligomer structures provide important clues to biological roles of proteins. Prediction of oligomer structures therefore can be a useful tool in the absence of experimentally resolved structures. In this article, we describe the server and human methods that we used to predict oligomer structures in the CASP13 experiment. Performances of the methods on the 42 CASP13 oligomer targets consisting of 30 homo-oligomers and 12 hetero-oligomers are discussed. Our server method, Seok-assembly, generated models with interface contact similarity measure greater than 0.2 as model 1 for 11 homo-oligomer targets when proper templates existed in the database. Model refinement methods such as loop modeling and molecular dynamics (MD)-based overall refinement failed to improve model qualities when target proteins have domains not covered by templates or when chains have very small interfaces. In human predictions, additional experimental data such as low-resolution electron microscopy (EM) map were utilized. EM data could assist oligomer structure prediction by providing a global shape of the complex structure.  相似文献

Bovine whey from the cheese-making industry contains several bioactive factors that promote health and prevent disease. Although many efforts have been made over the years to show that immunoglobulins, lactoperoxidase, lactoferrin, lysosyme and small peptides present in whey have antimicrobial activities against several pathogenic microorganisms, such activities have not been investigated so far for the lipid fraction of whey. Here, we have used an in vitro assay-based fractionation procedure to show that free fatty acids derived from whey cream specifically inhibit the germination of Candida albicans, a morphologic change associated with pathogenicity. Further fractionation by HPLC demonstrated that this activity can be mainly attributed to lauric acid, myristoleic acid, linoleic acid and arachidonic acid.  相似文献
