In the present study, the reaction mixtures (lead compounds with essential unsaturated fatty acids) were preincubated at 37°C for 24 h prior to the measurement of malondialdehyde (MDA) by HPLC. The metal-catalyzed reactions were also compared in the presence of butylated hydroxytoluene (BHT), a free radical scavenger. Our results showed that according to the difference in the number of double bonds of essential unsaturated fatty acids, the kinds of lead compounds, and the concentrations of lead compounds, the extent of lipid peroxidation was different. The addition of BHT to the reaction mixtures significantly reduced the production of MDA (P<0.01). These in vitro studies support prior in vivo reports that the important mechanism of the acute toxic effects of the lead compounds is owing at least in part, to metal-catalyzed peroxidation of polyun-saturated fatty acids.  相似文献

Summary Previously, we reported the isolation of a new microbial strain,Flavobacterium sp. DS5 (NRRL B-14859) which converted oleic and linoleic acids to their corresponding 10-keto- and 10--ydroxy-fatty acids. The hydration enzyme seemed to be specific to the C-10 position. Now we have identified, by GC/MS, NMR, and FTIR, the bioconversion products from -linolenic acid as 10-hydroxy-12(Z), 15(Z)-octadecadienoic acid and from -linolenic acid as 10-hydroxy-6(Z), 12(Z)-octadecadienoic acid. Products from 9(E)-unsaturated fatty acids were also identified as their corresponding 10-hydroxy or 10-keto fatty acids. From these results, it is concluded that strain DS5 hydratase is indeed a C-10 positional-specific enzyme and prefers an 18-carbon mono-unsaturated fatty acid. Among the C18 unsaturated fatty acids, an additional double bond on either side of the C-9 position lowers the enzyme hydration activity.  相似文献

Gasoline spills were simulated in the laboratory with three air‐dried soils, using a synthetic gasoline and unsaturated soil to quantify passive volatilization over a period of up to 16 d. The total and individual gasoline components were monitored as a function of time and depth in the soil.

The time required to deplete the overall gasoline concentration in the soil to 40% of the initial concentration ranged from 0.25 to 10 d for the three soils. Sand was the fastest, followed by loamy sand and silt loam. The volatilization rate of gasoline from soil was found to be dependent on soil, chemical type, and depth. Observation of individual components indicated that a wicking mechanism contributed to the gasoline flux toward the surface.  相似文献

Exposure to ethanol at 0 days of development induced changes in total membrane fatty acid composition at 18 days of development. When exposed to ethanol concentrations ranging from 0–743.27μm/kg egg wt, decreased levels of long-chain, unsaturated membrane fatty acids and increased levels of short-chain, saturated membrane fatty acids were observed in embryonic chick brains at 18 days of development. The ratios of unsaturated membrane/saturated membrane fatty acids correlated with an ethanol-induced reduction in neuron densities within the cerebral hemispheres and three different regions of the optic lobes with correlation coefficients (r) ranging from 0.44 [F = (1, 32) 7.84; P ≤ 0.009] to 0.59 [F = (1, 32) 17.38; P ≤ 0.0002]. The ratios of long-chain/short-chain membrane fatty acids also correlated with an ethanol-induced reduction in neuron densities within the cerebral hemispheres and three different regions of the optic lobes with correlation coefficients (r) ranging from 0.51 [F = (1, 32) 11.27; P≤ 0.002] to 0.66 [F = (1, 32) 24.40; P ≤ 0.0001]. Cell fractionation studies indicated that the ethanol-induced changes in brain membrane fatty acid composition were restricted to microsomal membranes.  相似文献

Electrophiles are electron-deficient species that form covalent bonds with electron-rich nucleophiles. In biological systems, reversible electrophile–nucleophile interactions mediate basal cytophysiological functions (e.g. enzyme regulation through S-nitrosylation), whereas irreversible electrophilic adduction of cellular macromolecules is involved in pathogenic processes that underlie many disease and injury states. The nucleophiles most often targeted by electrophiles are side chains on protein amino acids (e.g. Cys, His, and Lys) and aromatic nitrogen sites on DNA bases (e.g. guanine N7). The sulfhydryl thiol (RSH) side chain of cysteine residues is a weak nucleophile that can be ionized in specific conditions to a more reactive nucleophilic thiolate (RS?). This review will focus on electrophile interactions with cysteine thiolates and the pathophysiological consequences that result from irreversible electrophile modification of this anionic sulfur. According to the Hard and Soft, Acids and Bases (HSAB) theory of Pearson, electrophiles and nucleophiles can be classified as either soft or hard depending on their relative polarizability. HSAB theory suggests that electrophiles will preferentially and more rapidly form covalent adducts with nucleophiles of comparable softness or hardness. Application of HSAB principles, in conjunction with in vitro and proteomic studies, have indicated that soft electrophiles of broad chemical classes selectively form covalent Michael-type adducts with soft, highly reactive cysteine thiolate nucleophiles. Therefore, these electrophiles exhibit a common mechanism of cytotoxicity. As we will discuss, this level of detailed mechanistic understanding is a necessary prerequisite for the rational development of effective prevention and treatment strategies for electrophile-based pathogenic states.  相似文献

Normal human milk (NHM) has antiprotozoal activity unrelated to immunological components; this activity extends to sporozoites of Eimeria tenella . This activity may be due to free fatty acids (FFA) enzymatically hydrolyzed from tnacyl glycerols by a bile salt-stimulated lipase (BSSL) found in NHM. Sporozoites were therefore incubated in the presence of several saturated and unsaturated FFA. Anticoccidial activity was observed for many unsaturated fatty acids and for some saturated fatty acids. In addition, sporozoites were added to solutions of triglycerides (trilinolein, triolein and trilinolenin) preincubated with BSSL and sodium cholate. which resulted in killing of the parasites. Triglycerides alone showed no anticoccidial activity. These results were duplicated with first generation merozoites. Intracellular stages of E. tenella were affected by FFA only at concentrations that inhibited host cells.  相似文献

Variable temperature 1H-nuclear magnetic resonance (NMR) has been used to study the interaction of the RNA trimer, GpCpA, with the intercalators ethidium bromide and the acridine derivatives; proflavin, 9-amino-acridine, acridine orange, acridine yellow and acriflavin. The complexes formed were studied at nucleic acid to drug ratios of 1:1 and 5:1, the latter being useful in defining the effects of structural variation in the acridine series and in determining the site of intercalation. All the intercalators greatly stabilized the oligonucleotide duplex, the average melting temperature (Tm) increasing by up to 30 degrees C. Significant changes in individual Tms and chemical shifts were observed for all the GpCpA protons. 9-Amino-acridine and acriflavin did not stabilize the GpCpA duplex as substantially as the other acridine derivatives. It is suggested that this intercalator:GpCpA system, and its associated NMR-derived Tm, is a useful physical probe for potential mutagens.  相似文献
