The initiation of meiotic recombination by the formation of DNA double-strand breaks (DSBs) catalysed by the Spo11 protein is strongly evolutionary conserved. In Saccharomyces cerevisiae, Spo11 requires nine other proteins for meiotic DSB formation, but, unlike Spo11, few of these proteins seem to be conserved across kingdoms. In order to investigate this recombination step in higher eukaryotes, we have isolated a new gene, AtPRD1, whose mutation affects meiosis in Arabidopsis thaliana. In Atprd1 mutants, meiotic recombination rates fall dramatically, early recombination markers (e.g., DMC1 foci) are absent, but meiosis progresses until achiasmatic univalents are formed. Besides, Atprd1 mutants suppress DSB repair defects of a large range of meiotic mutants, showing that AtPRD1 is involved in meiotic recombination and is required for meiotic DSB formation. Furthermore, we showed that AtPRD1 and AtSPO11-1 interact in a yeast two-hybrid assay, suggesting that AtPRD1 could be a partner of AtSPO11-1. Moreover, our study reveals similarity between AtPRD1 and the mammalian protein Mei1, suggesting that AtPRD1 could be a Mei1 functional homologue. 相似文献

Aims: To evaluate the positive influence of the probiotic strain Lactobacillus paracasei LMGP22043 carried by artichokes into the human gut with special reference to faecal bacterial balance, short‐chain fatty acid concentrations and enzyme activities in a randomized, double‐blind human trial in comparison with probiotic‐free artichokes (control). Methods: Twenty subjects were randomized into two groups, which consumed daily 180 g of the artichoke product (probiotic or control) during two 15‐day study periods (periods 1 and 2) separated by a 15‐day washout in a crossover manner. Faecal samples were subjected to microbiological and biochemical analyses, and a strain‐specific PCR was performed to monitor the probiotic strain. Results: The probiotic strain, transported by the vegetable matrix, transiently colonized the gut of 17/20 subjects (median 6·87 log CFU g?1 faeces), antagonized Escherichia coli and Clostridium spp. and increased the genetic diversity of lactic population based on REP‐PCR profiles, mainly after period 1. Conclusions: The probiotic L. paracasei LMGP22043 successfully colonized the human gut and positively influenced faecal bacteria and biochemical parameters. Significance and Impact of the Study: The association of the probiotic L. paracasei with a food carrier rich in fibre can represent a new strategy for favouring a daily supply of probiotics and attracting more consumers to vegetable food fortified with probiotic strains. 相似文献

Unequal crossing over between direct DNA repeats of sister chromosomes occurs during DNA replication in Escherichia coli. Such exchanges yield tandem duplications and thereby increase the expression of the genes involved. Nonhomologous cohesion of sister chromosomes and unequal crossing over were assumed to take place when the replication fork stops. The various iterations of the HapMap Project and many genome-wide association studies (GWAS) have identified hundreds of potential genes involved in monogenic and multifactorial traits. We constructed an arrayed 115,000-member human genomic library in the PAC shuttle vector pJCPAC-Mam2 that can be propagated in both bacterial and human cells. The library appears to represent a two-fold coverage of the human genome. Transient transfection of a p53-containing PAC clone into p53-null Saos-2 human osteosarcoma cells demonstrated that both p53 mRNA and protein were produced. Additionally, expression of the p53 protein triggered apoptosis in a subset of the Saos-2 cells. This library should serve as a valuable resource to validate potential disease genes identified by GWAS in human cell lines and in animal models. Also, individual library members could potentially be used for gene therapy trials for a variety of recessive disorders. 相似文献

The complex [Fe(III)(salten)(mepepy)]BPh4 (salten = 4-azaheptamethylene-1,7-bis(salicylideneiminate; mepepy = 1-(pyridin-4-yl)-2-(N-methylpyrrol-2-yl)-ethene; BPh4 = tetraphenyl borate) has been investigated to determine the volume and enthalpy changes associated with the room temperature photo-induced spin crossover. Here we report the photophysical properties of the trans to cis isomerization of the mepepy ligand as well as the spin crossover of the Fe(III)(salten)(mepepy) complex in acetonitrile:water mixtures using photoacoustic calorimetry (PAC). The PAC studies indicate that the trans to cis transition of the mepepy ligand occurs faster than the ∼20 ns response time of the acoustic detector and gives rise to a negligible volume change (0.7 ± 0.3 mL mol−1) and an enthalpy change of 33 ± 10 kcal mol−1. These results are consistent with the loss of a charge assisted hydrogen bond between a water molecule and the pyridyl ring of the mepepy upon photoisomerization. In the case of Fe(III)(salten)(mepepy) photoexcitation, PAC results indicate that the high-spin to low-spin transition, also occurring in ?20 ns, gives rise to small volume and enthalpy changes (0.9 mL mol−1 and 4 kcal mol−1). Analysis of the results indicate that the observed thermodynamics are related to a distortion of the Fe(II)(salten)(mepepy) complex associated with the cleavage of an Fe?N bond upon spin conversion. 相似文献
