The transfer of substances across the interface between water and a membrane or between water and a solvent occurs in series with transport up to and away from the interface. These processes have been difficult to resolve. Recently D. M. Miller (Biochim Biophys Acta 856: 27–35, 1986) has used a moving drop technique to measure the rates of transfer of short-chain alcohols and tritiated water between water andn-octanol. This technique produces equivalent unstirred layers which are less than about 10 m thick. Based on the trends in the observed rates of phase transfer, he proposes that the transfer is limited by the actual interfacial step. If so, water-oil interfacial transfer would be sufficiently slow to limit the rate of permeation of lipid membranes by these substances. It is shown here that the observed rates of phase transfer can be explained quantitatively if they are limited by convection or by diffusion across the combination of 5–10 m unstirred layers both inside and outside the moving drops. For water, comparison of the observed rates with the rate of evaporation from a clean surface, suggests that the interfacial step at the water-octanol interface is not rate-limiting. 相似文献

Five plant species were cultivated on a soil from the Neckar alluvial fan near Heidelberg (FRG) polluted by the emissions
of a cement plant. Thallium, cadmium and lead concentrations in seedlings and mature plants were determined by atomic absorption
analysis. AdditionallyBrassica napus L.napus was grown on soils containing 5 different concentrations of heavy metals, achieved by mixing two similar soils, from the
same area but with different metal concentrations.
Thallium and cadmium were shown to be taken up by roots whilst lead which was also absorbed, was deposited mainly on the plant
surface. However during cultivation in the winter months, a remarkable deposit of lead via the roots was found. Thallium in
the soil from a anthrorogen source was more available to plants than thallium of geological origin.
During the lifetime of a plant concentrations of thallium and cadmium were always highest in the seedling. The decrease in
metal concentration with maturity depended on the plant species and the element, but was not a function of the metal concentration

Summary The nuclei of plant cells harbor genes for two types of glyceraldehyde-3-phosphate dehydrogenases (GAPDH) displaying a sequence divergence corresponding to the prokaryote/eukaryote separation. This strongly supports the endosymbiotic theory of chloroplast evolution and in particular the gene transfer hypothesis suggesting that the gene for the chloroplast enzyme, initially located in the genome of the endosymbiotic chloroplast progenitor, was transferred during the course of evolution into the nuclear genome of the endosymbiotic host. Codon usage in the gene for chloroplast GAPDH of maize is radically different from that employed by present-day chloroplasts and from that of the cytosolic (glycolytic) enzyme from the same cell. This reveals the presence of subcellular selective pressures which appear to be involved in the optimization of gene expression in the economically important graminaceous monocots. 相似文献

Methanogenesis by a Syntrophomonas wolfei/ Methanospirillum hungatei coculture was inhibited in presence of ethylene and the hydrogenation catalyst Pd-BaSO4. However, butyrate oxidation by S. wolfei continued and ethylene was reduced to ethane. Per mol of butyrate oxidized, 2.4 mol acetate was produced and 0.8 mol ethylene was reduced. Acetylene, propylene and butene were less effective as H2 acceptors than ethylene, and addition of bromoethanesulfonic acid was necessary to inhibit methanogenesis in the presence of the two longer-chain olefins. Other hydrogenation catalysts were less effective in the order Pd-charcoal < PE-asbestos < Pd-PEI beads < Pt-Al2O3, Pd-CaCO3. Optimal ethylene hydrogenation was achieved with still incubation in presence of 7.2 mg Pd-BaSO4 and 0.7 g sand per ml medium. The higher catabolic rate of S. wolfei in presence of the methanogen indicated that the biological H2 removal mechanism was more efficient than the catalytic olefin reduction.Abbreviations BES

ABSTRACT. 1. Deme formation is the transformation of a generalist population into one which is adapted to its local conditions. This adaptation has been inferred from many things but should be inferred from higher survival or fecundity of scale insects on their natal tree compared to that of immigrant scales on the same tree.
2. Transfers of the scale insect Matsucoccus acalyptus Herbert within and between infested host trees ( Pinus monophylla (Torr. & Frem.) resulted in significant differences in scale survivorship among recipient trees. Survival on individual trees was correlated across years, indicating stable differences in tree susceptibility to scale.
3. A significant natal tree colonized tree interaction was observed for late stage scale survival in one experiment but the interaction was not caused by superior survivorship of scales transferred back to the natal tree. Hence, we found no evidence of deme formation in M.acalyptus.
4. Previous studies have concluded that deme formation occurs in the black pineleaf scale based on a significant natal tree by colonized tree interaction in scale survival. We question this conclusion because the experimental design employed did not include transfers back onto the natal tree. Expression vectors for cDNA of the κ and λ1 chains of a monoclonal antibody directed against creatine kinase were introduced into murine myeloma cells. κ and γ1 cDNA were either under the control of the SV40 early promoter or of the cognate promoters and enhancers of the light- and heavy-chain genes. Secretion of immuno-reactive κ and γ1 chains into the culture medium was demonstrated with the SV40 promoter as well as with the cognate promoters. Expression of y 1 cDNA with the SV40 early promoter was about twice as high as with the heavy-chain promoter and enhancer. Expression of κ cDNA under the control of the S V40 early promoter was about 17 times higher than with the light-chain promoter and enhancer. These expression levels were compared to those of a genomic immunoglobulin (Ig) κ determinant, including introns. Such an entire κ gene led to expression of the light chain at levels double those with the κ cDNA construction using the SV40 promoter and about 35 times as high when using κ cDNA and the cognate promoter and enhancer. This result might indicate that, besides the cognate promoter and enhancer elements, other intragenic elements are involved in the regulation of Ig expression. Diurnal variation in ion content of the solution bathing roots of two plants growing together in sand culture was analysed
for three pairs of grass-legume species (Lolium multiflorum andTrifolium pratense; Zea mays andGlycine hispida; Avena sativa andVicia sativa) and their monospecific controls. Biomass and nitrogen content of plants were determined.

, and K+) and pH of root solutions were measured for Lolium-Trifolium plant pairs and controls at 6 hours intervals over 36 h, starting
at 8 am within a circadian cycle. Root solutions were regularly depleted in NO
3
−
by the grasses (Lolium-Lolium control) throughout the cycle. For associations involving the legume (Lolium-Trifolium and
Trifolium-Trifolium), NO
3
−
depletion was followed by NO
3
−
enrichment at night, from late afternoon to early morning; the enrichment was more marked for the Lolium-Trifolium association.
Solutions which did not contain NO
2
−
ions, were enriched by trace amounts of NH
4
+
ions, largely depleted in K+ and alkalanized for all associations throughout the cycle.
Repeating the experiment with the three pairs of species at the vegetative phase of development confirmed the previous results:
NO
3
−
enrichment during the night for associations with legumes. When the experiment was repeated with older plants which had almost
completed their flowering stage, depletion only was observed and no NO
3
−
enrichment.
These data suggest that NO
3
−
enrichment results from N excretion from active nodulated roots of the legume, accounting for the increase in both biomass
and nitrogen content of the companion grass in grass-legume association. The quantitative importance and periodicity of nitrogen

Manganese in the oxygen-evolving complex is a physiological electron donor to Photosystem II. PS II depleted of manganese may oxidize exogenous reductants including benzidine and Mn2+. Using flash photolysis with electron spin resonance detection, we examined the room-temperature reaction kinetics of these reductants with Yz
+, the tyrosine radical formed in PS II membranes under illumination. Kinetics were measured with membranes that did or did not contain the 33 kDa extrinsic polypeptide of PS II, whose presence had no effect on the reaction kinetics with either reductant. The rate of Yz
+ reduction by benzidine was a linear function of benzidine concentration. The rate of Yz
+ reduction by Mn2+ at pH 6 increased linearly at low Mn2+ concentrations and reached a maximum at the Mn2+ concentrations equal to several times the reaction center concentration. The rate was inhibited by K+, Ca2+ and Mg2+. These data are described by a model in which negative charge on the membrane causes a local increase in the cation concentration. The rate of Yz
+ reduction at pH 7.5 was biphasic with a fast 400 s phase that suggests binding of Mn2+ near Yz

We have measured the extent of flash-induced electron transfer from the bacteriochlorophyll dimer, P, to the bacteriopheophytin in the M-subunit, HM, in reaction centers of Rhodopseudomonas viridis. This has been done by measuring the transient states produced by excitation of reaction centers trapped in the PHL
–HM state at 90 K. Under these conditions the normal forward electron transfer to the bacteriopheophytin in the L-subunit, HL, is blocked and the yield of transient P+HM
– can be estimated with respect to the lifetime of P*. Under these conditions flash induced absorbance decreases of the bacteriochlorophyll dimer 990 nm band suggest that a transient P+ state is formed with a quantum yield of 0.09±0.06 compared to that formed during normal photochemistry. These transient measurements provide an upper limited on the yield of a transient P+ HM
– state. An estimate of 0.09 as the yield of the P+ HM
– state is consistent with all current observations. This estimate and the lifetime of P* suggest that the electron transfer rate from P* to HM, kM, is about 5 × 109 sec–1 (M = 200ps). These measurements suggest that the a branching ratio kL/kM is on the order of 200. The large value of the branching ratio is remarkable in view of the structural symmetry of the reaction center. This measurement should be useful for electron transfer calculations based upon the reaction center structure. 相似文献
