In cells, mitochondria, endoplasmic reticulum, and peroxisomes are the major sources of reactive oxygen species (ROS) under physiological and pathophysiological conditions. Cytochrome c (cyt c) is known to participate in mitochondrial electron transport and has antioxidant and peroxidase activities. Under oxidative or nitrative stress, the peroxidase activity of Fe3+cyt c is increased. The level of NADH is also increased under pathophysiological conditions such as ischemia and diabetes and a concurrent increase in hydrogen peroxide (H2O2) production occurs. Studies were performed to understand the related mechanisms of radical generation and NADH oxidation by Fe3+cyt c in the presence of H2O2. Electron paramagnetic resonance (EPR) spin trapping studies using 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) were performed with NADH, Fe3+cyt c, and H2O2 in the presence of methyl-β-cyclodextrin. An EPR spectrum corresponding to the superoxide radical adduct of DMPO encapsulated in methyl-β-cyclodextrin was obtained. This EPR signal was quenched by the addition of the superoxide scavenging enzyme Cu,Zn-superoxide dismutase (SOD1). The amount of superoxide radical adduct formed from the oxidation of NADH by the peroxidase activity of Fe3+cyt c increased with NADH and H2O2 concentration. From these results, we propose a mechanism in which the peroxidase activity of Fe3+cyt c oxidizes NADH to NAD•, which in turn donates an electron to O2, resulting in superoxide radical formation. A UV-visible spectroscopic study shows that Fe3+cyt c is reduced in the presence of both NADH and H2O2. Our results suggest that Fe3+cyt c could have a novel role in the deleterious effects of ischemia/reperfusion and diabetes due to increased production of superoxide radical. In addition, Fe3+cyt c may play a key role in the mitochondrial “ROS-induced ROS-release” signaling and in mitochondrial and cellular injury/death. The increased oxidation of NADH and generation of superoxide radical by this mechanism may have implications for the regulation of apoptotic cell death, endothelial dysfunction, and neurological diseases. Despite the efforts of many natural resource professionals, wild pig (Sus scrofa) populations are expanding in many areas of the world. Although many creative techniques for controlling pig populations are being explored, trapping has been and still is the most commonly used method of population control for many public and private land managers. We conducted an observational study to examine the efficiency of 2 frequently used trap styles: a small, portable box-style trap and a larger, semi-permanent, corral-style trap. We used game cameras to examine patterns of trap entry by wild pigs around each style of trap, and we conducted a trapping session to compare trapping success between trap styles. Adult female and juvenile wild pigs entered both styles of trap more readily than did adult males, and adult males seemed particularly averse to entering box traps. Less than 10% of adult male visits to box traps resulted in entries, easily the least percentage of any class at any style of trap. Adult females entered corral traps approximately 2.2 times more often per visit than box traps and re-entered corral traps >2 times more frequently. Juveniles entered and re-entered both box and corral traps at similar rates. Overall (all-class) entry-per-visit rates at corral traps (0.71) were nearly double that of box traps (0.37). Subsequent trapping data supported these preliminary entry data; the capture rate for corral traps was >4 times that of box traps. Our data suggest that corral traps are temporally and economically superior to box traps with respect to efficiency; that is, corral traps effectively trap more pigs per trap night at a lower cost per pig than do box traps. © 2011 The Wildlife Society. 相似文献

Rice is an important crop and major model plant for monocot functional genomics studies. With the establishment of various genetic resources for rice genomics, the next challenge is to systematically assign functions to predicted genes in the rice genome. Compared with the robustness of genome sequencing and bioinformatics techniques, progress in understanding the function of rice genes has lagged, hampering the utilization of rice genes for cereal crop improvement. The use of transfer DNA (T‐DNA) insertional mutagenesis offers the advantage of uniform distribution throughout the rice genome, but preferentially in gene‐rich regions, resulting in direct gene knockout or activation of genes within 20–30 kb up‐ and downstream of the T‐DNA insertion site and high gene tagging efficiency. Here, we summarize the recent progress in functional genomics using the T‐DNA‐tagged rice mutant population. We also discuss important features of T‐DNA activation‐ and knockout‐tagging and promoter‐trapping of the rice genome in relation to mutant and candidate gene characterizations and how to more efficiently utilize rice mutant populations and datasets for high‐throughput functional genomics and phenomics studies by forward and reverse genetics approaches. 1. A number of planktonic cyanobacteria species form resting stages that survive in the sediments of lakes. The significance of this life history strategy to the ecology of new planktonic populations was investigated in Esthwaite Water, a mesotrophic lake in the English Lake District.
2. A simple trapping technique was used to quantify vertical movements of five species of buoyant gas-vacuolate cyanobacteria from close to the sediments, along a depth transect.
3. 'Recruitment' from the sediments was found to be widespread amongst the cyanobacteria species associated with the summer phytoplankton community.
4. Estimates of the vertical upward fluxes of cyanobacteria based upon trap catches could not account for observed increases in the planktonic populations suggesting that 'recruitment' was not a significant source of biomass.

Bleomycin, in the presence of ferric salts, oxygen and a suitable reductant, degrades DNA with the release of base propenals, detected as thiobarbituric acid (TBA) reactivity, and the formation of 8-hydroxydeo-xyguanosine (80HdG) detected by HPLC. When xanthine oxidase is added to the incubated mixture of DNA degradation products, TBA-reactivity is destroyed but 80HdG formation is increased. EPR Spin trapping experiments show that hydroxyl radicals (OH) are formed in the reaction mixture and can be inhibited by the inclusion of either superoxide dismutase or catalase. These findings suggest that the base propenals and possibly malondialdehyde, formed from them, are aldehydic substrates for xanthine oxidase and, the product of this reaction is superoxide (O2-) and hydrogen peroxide (H2O2). Thus, TBA reactivity is destroyed in the formation of O2- and H2O2 which stimulate further oxidative damage to DNA resulting in increased 8OHdG formation. 相似文献

Chiracanthium mildei, a common sac spider (Clubionidae) in Israel, is an important predator of pest insects in citrus and apple orchards. Field observations indicate that the females of this hunting spider are attracted to traps containing the parapheromone trimedlure and males of the Mediterranean fruit fly (Ceratitis capitata),which were caught in these traps. Laboratory testsshow that C. mildei females are attractedto olfactory cues from male C. capitata, but notto trimedlure. 【目的】研究旨在深入探讨中国黄杨绢野Diaphania perspectalis 的雌蛾性信息素组成及诱蛾活性。【方法】利用气质联用仪(GC-MS)对黄杨绢野螟正常型性成熟雌蛾的性腺体提取物和合成标样比较分析,并用反-11-十六碳烯醛(E11-16:Ald) 、顺-11-十六碳烯醛(Z-11-16:Ald)、顺-9-十六碳烯醛(Z-9-16:Ald)、顺-11-十六碳烯醇(Z-11-16:OH)等物质进行触角的电生理测定,最后开展田间诱集比较试验以筛选出最佳性信息素混合物。【结果】Z-11-16:Ald为中国黄杨绢野螟正常型性信息素主要组分,E-11-16:Ald的含量极低,Z-11-16:OH未检测到。正常型雄性黄杨绢野螟触角对Z-11-16:Ald, E-11-16:Ald, Z-9-16:Ald和Z-11-16:OH产生强烈的EAG反应,并随着浓度的提高而显著增加;而对Z-11-16:Ac和E-11-16:Ac的嗅觉反应较弱,低于对植物绿叶气味顺3-己烯乙酸酯(Z-3-6:Ac)的反应。单一Z-1-16:Ald对正常型雄性黄杨绢野螟具有强烈的诱集效果,加入E-11-16:Ald有一定的增效作用,但在统计上则不显著。单一Z-11-16:Ald组分对黑化型雄性黄杨绢野螟无引诱活性,必需加入一定比例的E-11-16:Ald才显示诱蛾活性。Z-11-16:Ald:E-11-16:Ald的比例为250 μg:250 μg时诱集到的黑化型雄性黄杨绢野螟数量最多,而Z-11-16:Ald:E-11-16:Ald的比例为429 μg:71 μg时则诱集到的正常型雄性黄杨绢野螟数量最多。同时,单一Z-11-16:Ald也可引诱大量雄性粘虫Mythimna separata,但E-11-16:Ald抑制其活性。【结论】中国黄杨绢野螟的性信息素主成分是Z-11-16:Ald,单一组分即可在田间强烈引诱雄蛾,E-11-16:Ald的功能只起到微弱的增效作用,但也可能起种的专一性的作用。正常型与黑化型黄杨绢野螟对性信息素的嗅觉反应存在差异,黑化型黄杨绢野螟的性信息素接近日本种,即性信息素组成为Z-11-16:Ald和E-11-16:Ald 的混合物,其比例为1:1,且E-11-16:Ald为必需。 相似文献
