Four field trials were done with narrow-leafed lupins (Lupinus angustifolius) in 1988 - 1989, to examine the effect of sowing seed with 5% and 0.5% cucumber mosaic virus (CMV) infection on subsequent virus spread, grain yield and percentage of infection in harvested seed. A proportion of the CM V-infected seed failed to produce established plants and thus, plots sown with 5% and 0.5% infected seed contained 1.5-2.9% and 0.2-0.3% of seed-infected plants respectively. The rate of virus spread by aphids was faster and resulted in more extensive infection at maturity in plots sown with 5% infected seed than with 0.5% infected seed. In three trials, sowing 5% infected seed resulted in yield losses of 34 - 53% and CMV infection in the seed harvested of 6 - 13%. The spread of CMV infection resulting from sowing 0.5% infected seed did not significantly decrease yield. However, late CMV spread in these plots caused > 1% seed infection. In the fourth trial, which was badly affected by drought, CMV spread only slowly, there was no significant effect of CMV on grain yield and the percentage of infected seed harvested was 3–5 times less than that in the seed sown. When CMV-infected seed was sown at different depths, target depths of 8 and 11 cm decreased the incidence of seed-infected plants by c. 15% and c. 50% respectively compared with sowing at 5 cm. However, in glasshouse tests, treatment with the pre-emergence herbicide simazine failed to selectively cull out seed-infected plants. The field trials were colonised by green peach (Myzus persicae), blue-green (Acyrthosiphon kondoi) and cowpea (Aphis craccivora) aphids. When the abilities of these aphid species and of the turnip aphid (Lipaphis erysimi) in transmitting CMV from lupins to lupins were examined in glasshouse tests, short acquisition access times favoured transmission. With 5–10 min acquisition access times, overall transmission efficiencies were 10.8%, 9.4%, 6.1% and 3.9% for the green peach, cowpea, blue-green and turnip aphids respectively.  相似文献

The evidence for the mechanisms involved in virus transmission by fungi is reviewed in relation to the non-persistent and persistent categories usually recognised. Non-persistent transmission by Olpidium spp. has been little studied in the last 20 years, but appears to depend on adsorption of virus to the outside of the fungal zoospores. This seems to be under the genetic control of both the virus (via its coat protein) and the vector. Such viruses are not transmitted in the fungal resting spores. The route by which the virus is transferred from the vector to the host may involve uptake into the zoospore but this deserves further study. Persistent transmission by Olpidium, Polymyxa and Spongospora spp. is less well characterised. Some of the evidence used for its support is inconclusive. The viruses are probably always carried inside zoospores, and they also persist in the fungal resting spores. Transmission depends on the genome of the vector and the virus, but not exclusively on the virus coat protein.  相似文献

The mechanisms involved in the impaired spermatogenesis of male goldfinch x canary hybrids were investigated by transmission electron microscopy and compared with spermatogenesis in the testes of the parent species. In the parent species the testes were of normal structure, with the only unusual observation being that the Sertoli cells were variable in cytoplasmic electron density. In hybrid birds the Sertoli cells were either electron dense or electron lucent with respect to both nucleus and cytoplasm. In the hybrids examined in this study, no spermatozoa were produced. Spermatogenic stages were arrested without formation of synaptonemal complexes. Centrioles were abnormally arranged in both somatic and germ cells. When they moved away from the basement lamina the germ cells degenerated and were phagocytosed. No focal tight junctions were present between Sertoli cells overlying what would normally have been the basal compartment of the tubule. The basement lamina was unusually thickened, peritubular cells were abnormal in structure, and numerous plasma cells were present in the interstitial tissue. The observations reported here suggest that there was an immunological basis for degeneration of germ cells in the hybrid testis.  相似文献

Urinary bladder activity involves central and autonomic nervous systems and bladder wall. Studies on the pathogenesis of voiding disorders such as the neurogenic detrusor overactivity (NDO) due to suprasacral spinal cord lesions have emphasized the importance of an abnormal handling of the afferent signals from urothelium and lamina propria (LP). In the LP (and detrusor), three types of telocytes (TC) are present and form a 3D‐network. TC are stromal cells able to form the scaffold that contains and organizes the connective components, to serve as guide for tissue (re)‐modelling, to produce trophic and/or regulatory molecules, to share privileged contacts with the immune cells. Specimens of full thickness bladder wall from NDO patients were collected with the aim to investigate possible changes of the three TC types using histology, immunohistochemistry and transmission electron microscopy. The results show that NDO causes several morphological TC changes without cell loss or network interruption. With the exception of those underlying the urothelium, all the TC display signs of activation (increase in Caveolin1 and caveolae, αSMA and thin filaments, Calreticulin and amount of cisternae of the rough endoplasmic reticulum, CD34, euchromatic nuclei and large nucleoli). In all the specimens, a cell infiltrate, mainly consisting in plasma cells located in the vicinity or taking contacts with the TC, is present. In conclusion, our findings show that NDO causes significant changes of all the TC. Notably, these changes can be interpreted as TC adaptability to the pathological condition likely preserving each of their peculiar functions.  相似文献

Acquired immune deficiency syndrome (AIDS), caused by infection with human immunodeficiency virus (HIV), is associated with gastrointestinal disease, systemic immune activation and changes in the gut microbiota. Here, we aim to investigate the gut microbiota patterns of HIV‐infected individuals and HIV‐uninfected individuals in populations from South China. We enrolled 33 patients with HIV (14 participants treated with highly active antiretroviral therapy [HAART] for more than 3 months; the remaining 19 individuals had not received treatment) and 35 healthy controls (HC) for a cross‐sectional comparison of gut microbiota using stool samples. Gut microbial communities were profiled by sequencing the bacterial 16S rRNA genes. Dysbiosis was more common among patients with AIDS compared with healthy individuals. Dysbiosis was characterized by decreased α‐diversity, low mean counts of Bacteroidetes, Faecalibacterium, Prevotella, Bacteroides vulgatus, Dialister and Roseburia inulnivorans, and high mean counts of Proteobacteria, Enterococcus, Streptococcus, Lactobacillus, Lachnociostridium, Ruminococcus gnavus and Streptococcus vestibularis. Increased abundance of Bacilli was observed in homosexual patients. Proteobacteria were higher among heterosexual patients with HIV infections. Tenericutes were higher among patients with history of intravenous drug abuse. Restoration of gut microbiota diversity and a significant increase in abundance of Faecalibacterium, Blautia and Bacteroides were found in patients receiving HAART compared to those who did not receive. HIV infection‐associated dysbiosis is characterized by decreased levels of α‐diversity and Bacteroidetes, increased levels of Proteobacteria and the alterations of gut microbiota correlate with the route of HIV transmission. The imbalanced faecal microbiota of HIV infection is partially restored after therapy.  相似文献

Identifying patterns and drivers of infectious disease dynamics across multiple scales is a fundamental challenge for modern science. There is growing awareness that it is necessary to incorporate multi‐host and/or multi‐parasite interactions to understand and predict current and future disease threats better, and new tools are needed to help address this task. Eco‐phylogenetics (phylogenetic community ecology) provides one avenue for exploring multi‐host multi‐parasite systems, yet the incorporation of eco‐phylogenetic concepts and methods into studies of host pathogen dynamics has lagged behind. Eco‐phylogenetics is a transformative approach that uses evolutionary history to infer present‐day dynamics. Here, we present an eco‐phylogenetic framework to reveal insights into parasite communities and infectious disease dynamics across spatial and temporal scales. We illustrate how eco‐phylogenetic methods can help untangle the mechanisms of host–parasite dynamics from individual (e.g. co‐infection) to landscape scales (e.g. parasite/host community structure). An improved ecological understanding of multi‐host and multi‐pathogen dynamics across scales will increase our ability to predict disease threats.  相似文献

Wireless electric energy transmission is an important energy supply technology. However, most wireless energy supply based on electromagnetic induction cannot be used for energy transmission through a metal chamber. Herein, a novel idea for wireless electric energy transmission through various isolated solid media based on triboelectric nanogenerator (TENG) is presented. The electric energy is first transformed into mechanical vibration energy in mechanical wave that can propagate well in solid medium, and then the vibration energy is harvested by a TENG. By employing the spring steel sheets and freestanding triboelectric‐layer structure, the vibration TENG as an energy conversion unit has the advantages of high efficiency and facilitation, boosting this wireless energy transmission technology to be an alternative way of delivering electric energy through metal medium. The working principle and output performance have been systematically studied. A commercial capacitor can be charged from 0 to 10 V in 33 and 86 s isolated by an acrylic plate and a copper plate in thickness of 3 mm, respectively. The wireless electric transmission technology is also applied to deliver electric energy into a vacuum glove box and across glass wall successfully. This novel technology has great potential applications in implantable microelectronic devices, encrypted wireless communication, and even nondestructive testing.  相似文献
